Tetrahydroxystilbene glycoside improves endothelial dysfunction and hypertension in obese rats: the role of omentin-1
CONCLUSIONS: Down-regulation of omentin-1 induces endothelial dysfunction and hypertension in obesity. TSG treatment (at least partially) increases omentin-1 via promoting binding of PPAR-γ and Itln-1 promoter in adipose tissues, subsequently exerts protective effects on endothelial function via activating Akt/eNOS/NO signaling and attenuating oxidative/nitrative stress. The effect of emodin on melanogenesis through the modulation of ERK and MITF signaling pathway.
In this study, emodin did not show antioxidant activity in DPPH radical and reducing power assays. However, it was found that emodin scavenged intracellular H2O2. Emodin increased not only tyrosinase activity but also melanin synthesis in vitro. Tetrahydroxystilbene glucoside alleviates Ang Ⅱ-induced senescence of HUVECs via SIRT1.
In this study, the possible protective mechanism of TSG on human umbilical vein endothelial cells (HUVECs) senescence induced by angiotensin Ⅱ (Ang Ⅱ) was investigated. The results revealed that TSG pretreatment could reduce the percentage of senescence-associated-β-galactosidase (SA-β-gal) positive cells, and decrease the expression levels of the cellular senescence biomarkers, p53 and PAI-1 proteins.
