The electroneutral Na < sup > + < /sup > -HCO < sub > 3 < /sub > < sup > - < /sup > cotransporter NBCn1 (SLC4A7) modulates colonic enterocyte pH < sub > i < /sub > , proliferation and migration
Am J Physiol Cell Physiol. 2024 Apr 22. doi: 10.1152/ajpcell.00079.2024. Online ahead of print.ABSTRACTNBCn1 (SLC4A7) is one of the two major Na+-HCO3- cotransporters in the human colonic epithelium, expressed predominantly in the highly proliferating colonocytes at the cryptal base. Increased NBCn1 expression levels are reported in tumours, including colorectal cancer. The study explores its importance for maintenance of the intracellular pH (pHi),as well as the proliferative, adhesive, and migratory behavior of the self-differentiating Caco2bbe colonic tumour cell line. In the self-differentiating Caco2BBe cells, NBCn1 m...

Kinesin Facilitates Phenotypic Targeting of Therapeutic Resistance in Advanced Prostate Cancer
In this study, we use in vitro and in vivo preclinical MDA PCa PDX models of resistant human prostate cancer to determine molecular mechanisms of cross-resistance between anti-androgen therapy and taxane chemotherapy, underlying the therapeutically resistant phenotype. Transcriptomic profiling revealed that resistant and sensitive prostate cancer C4-2B cells have a unique differential gene signature response to cabazitaxel. Gene pathway analysis showed that sensitive cells exhibit increase in DNA damage, while resistant cells express genes associated with protein regulation in response to cabazitaxel. These PDX specimens a...

Promising anticancer activity of cromolyn in colon cancer: in vitro and in vivo analysis
CONCLUSION: Our study showed that cromolyn is a selective and strong drug in inhibiting the proliferation of colon cancer cells. Based on our results, the efficacy of C in vitro analysis (MTT assays and apoptosis), as well as animal studies is competitive with the FDA-approved drug doxorubicin. C is very promising as a low-complication and good-efficacy drug for cancer drug repositioning. This requires clinical research study designs to comprehensively evaluate its anti-cancer effects.PMID:38647571 | DOI:10.1007/s00432-024-05741-2 (Source: Cell Research)

Cell death as an architect of adult skin stem cell niches
Cell Death Differ. 2024 Apr 22. doi: 10.1038/s41418-024-01297-3. Online ahead of print.ABSTRACTOur skin provides a physical and immunological barrier against dehydration and environmental insults ranging from microbial attacks, toxins and UV irradiation to wounding. Proper functioning of the skin barrier largely depends on the interplay between keratinocytes- the epithelial cells of the skin- and immune cells. Two spatially distinct populations of keratinocyte stem cells (SCs) maintain the epidermal barrier function and the hair follicle. These SCs are inherently long-lived, but cell death can occur within their niches and...

Antibody-drug conjugates in lung and breast cancer: Current evidence and future directions - a position statement from the ETOP IBCSG Partners Foundation
Ann Oncol. 2024 Apr 20:S0923-7534(24)00108-X. doi: 10.1016/j.annonc.2024.04.002. Online ahead of print.ABSTRACTFollowing the approval of the first antibody-drug conjugates (ADCs) in the early 2000s, development has increased dramatically, with 14 ADCs now approved and >100 in clinical development. In lung cancer, trastuzumab deruxtecan (T-DXd) is approved in human epidermal growth factor receptor 2 (HER2)-mutated, unresectable or metastatic non-small cell lung cancer, with ADCs targeting HER3 (patritumab deruxtecan), trophoblast cell-surface antigen 2 (datopotamab deruxtecan and sacituzumab govitecan [SG]) and mesenchym...

Advances in molecular mechanisms of inflammatory bowel disease ‑associated colorectal cancer (Review)
Oncol Lett. 2024 Apr 9;27(6):257. doi: 10.3892/ol.2024.14390. eCollection 2024 Jun.ABSTRACTThe link between inflammation and cancer is well documented and colonic inflammation caused by inflammatory bowel disease (IBD) is thought to be a high-risk factor for the development of colorectal cancer (CRC). The complex crosstalk between epithelial and inflammatory cells is thought to underlie the progression from inflammation to cancer. The present review collates and summarises recent advances in the understanding of the pathogenesis of IBD-associated CRC (IBD-CRC), including the oncogenic mechanisms of the main inflammatory si...

Single-cell analysis revealed a potential role of T-cell exhaustion in colorectal cancer with liver metastasis
J Cell Mol Med. 2024 Apr;28(8):e18341. doi: 10.1111/jcmm.18341.ABSTRACTLiver metastasis (LM) is an important factor leading to colorectal cancer (CRC) mortality. However, the effect of T-cell exhaustion on LM in CRC is unclear. Single-cell sequencing data derived from the Gene Expression Omnibus database. Data were normalized using the Seurat package and subsequently clustered and annotated into different cell clusters. The differentiation trajectories of epithelial cells and T cells were characterized based on pseudo-time analysis. Single-sample gene set enrichment analysis (ssGSEA) was used to calculate enrichment scores...
