TANK-binding kinase 1 (TBK1) missense mutations predispose to neurologic disease including frontotemporal dementia and amyotrophic lateral sclerosis (FTD-ALS). FTD-ALS TBK1 variants have also been observed in patients with a primary schizophrenia diagnosis. TBK1 upregulates the transcription of type I interferon genes in response to bacteria including B. burgdorferi, which causes Lyme disease.
