Short-Term Evidence in Adults of Anorexigenic Drugs Acting in the Central Nervous System: A Meta-Analysis.
Abstract PURPOSE: Obesity is a chronic clinical condition that is considered one of the most serious health problems in the world because it can cause other chronic metabolic disorders. A meta-analysis was conducted to evaluate the safety and efficacy of 4 central-acting drugs, all approved in Brazil's market for weight loss. METHODS: PubMed, EMBASE, and Cochrane library databases were searched from inception until January 2018 to retrieve randomized controlled trials comparing sibutramine, diethylpropion, mazindol, and fenproporex versus placebo in overweight or obese patients. Language was not a ...

Liquid chromatography-quadrupole time of flight mass spectrometric method for targeted analysis of 111 nitrogen-based compounds in weight loss and ergogenic supplements
Publication date: Available online 31 May 2019Source: Journal of Pharmaceutical and Biomedical AnalysisAuthor(s): Bharathi Avula, Ji-Yeong Bae, Amar G. Chittiboyina, Yan-Hong Wang, Mei Wang, Ikhlas A. KhanAbstractThe use of supplements for weight loss and in sports as pre-workout (ergogenic) products is widespread. Many of these supplements were found to contain active components, which were not claimed on the labels. A validated liquid chromatography high-resolution mass spectrometry quadrupole time-of-flight (LC-QToF-MS) method was developed for the simultaneous analysis of 111 amine-based compounds belonging to anorecti...

WITHDRAWN: Interventions for treating obesity in children.
CONCLUSIONS: While there is limited quality data to recommend one treatment program to be favoured over another, this review shows that combined behavioural lifestyle interventions compared to standard care or self-help can produce a significant and clinically meaningful reduction in overweight in children and adolescents. In obese adolescents, consideration should be given to the use of either orlistat or sibutramine, as an adjunct to lifestyle interventions, although this approach needs to be carefully weighed up against the potential for adverse effects. Furthermore, high quality research that considers psychosocial det...

A simple and fast-portable method for the screening of the appetite-suppressant drug sibutramine in natural products and multivitamins supplements
Publication date: Available online 20 November 2018Source: Sensors and Actuators B: ChemicalAuthor(s): Jhonys M. Freitas, Thiago C. Oliveira, Mario H.P. Santana, Craig E. Banks, Rodrigo A.A. Munoz, Eduardo M. RichterAbstractSibutramine is one of the most widespread adulterants encountered in herbal formulations or dietary supplements used for weight-loss purposes. In this work, a simple and portable square-wave voltammetric (SWV) method was proposed for fast screening and quantification of sibutramine in herbal formulations and dietary supplements samples. The selective determination of sibutramine was possible using 0.1â€...

Sibutramine facilitates apoptosis and contraction of aortic smooth muscle cells through elevating production of reactive oxygen species.
Abstract Sibutramine had been prescribed as an oral anorexiant that reduces dietary intake, but was withdrawn from the market due to frequent occurrence of severe cardiovascular events including hypertension. To elucidate the pathogenic mechanism of hypertension, we here investigated whether sibutramine facilitates damage and contraction of human aortic smooth muscle (HASM) cells or not. Treatment with sibutramine provoked HASM cell apoptosis, which was attributed to production of reactive oxygen species and mitochondria dysfunction. In addition, the drug treatment of the cell promoted calcium influx, phos...

Combination Phentermine –Topiramate Extended Release for the Treatment of Binge Eating Disorder: An Open-Label, Prospective Study
Conclusion: Phentermine–topiramate could be helpful for weight loss and reduction of binge-eating symptoms in patients with obesity or overweight in addition to BED. Controlled studies are warranted. Binge-eating disorder (BED), an eating disorder recognized in the Diagnostic and Statistical Manual of Mental Disorders, fifth edition (DSM-5), is characterized by recurrent and distressing episodes of binge eating (eating an unusually large amount of food with a sense of loss of control over eating) without inappropriate compensatory weight-loss behaviors.1 Known as the most common eating disorder, BED is strongly associate...

S225. a systematic review and meta-analysis of pharmacological interventions for reduction or prevention of weight gain in schizophrenia
DiscussionAccumulating evidence supports the safe use of pharmacological interventions to achieve modest weight loss. Reboxetine may be slightly effective in preventing weight gain while metformin has the most evidence for use as treatment of weight gain in schizophrenia. The small number of studies, small sample size, and short study duration limits interpretation for other agents. Future studies that are adequately powered, with longer treatment duration, will be needed in evaluating the efficacy and safety of interventions for managing weight gain further. Drugs Involved in Dyslipidemia and Obesity Treatment: Focus on Adipose Tissue.
Authors: Dias S, Paredes S, Ribeiro L Abstract Metabolic syndrome can be defined as a state of disturbed metabolic homeostasis characterized by visceral obesity, atherogenic dyslipidemia, arterial hypertension, and insulin resistance. The growing prevalence of metabolic syndrome will certainly contribute to the burden of cardiovascular disease. Obesity and dyslipidemia are main features of metabolic syndrome, and both can present with adipose tissue dysfunction, involved in the pathogenic mechanisms underlying this syndrome. We revised the effects, and underlying mechanisms, of the current approved drugs f...

Molecules, Vol. 23, Pages 555: Enzyme Kinetics and Molecular Docking Studies on Cytochrome 2B6, 2C19, 2E1, and 3A4 Activities by Sauchinone
In conclusion, when sauchinone was co-treated with drugs metabolized via CYP2B6, 2C19, 2E1, or 3A4, sauchinone–drug interactions occurred because sauchinone inhibited the CYP-mediated metabolic activities. (Source: Molecules)
