Integrating network pharmacology and experimental validation reveals therapeutic effects of D-mannose on NAFLD through mTOR suppression
In this study, network pharmacology followed by molecular docking analysis was utilized to identify potential targets of mannose against NAFLD, and the leptin receptor-deficient, genetically obese db/db mice was employed as an animal model of NAFLD to validate the regulation of D-mannose on core targets. As a result, 67 targets of mannose are predicted associated with NAFLD, which are surprisingly centered on the mechanistic target of rapamycin (mTOR). Further analyses suggest that mTOR signaling is functionally enriched in potential targets of mannose treating NAFLD, and that mannose putatively binds to mTOR as a core mec...

Identification of mRNA expression profiles and their characterization in age-related hearing loss
In this study, we analyzed an mRNA expression dataset (GSE49543) from the GEO database. Differentially expressed genes (DEGs) between young control mice and presbycusis mice were analyzed using limma in R and weighted gene co-expression network analysis (WGCNA) methods. Functional enrichment analyses of the DEGs were conducted with the clusterProfiler R package and the results were visualized using ggplot2 R package. The STRING database was used for the construction of the protein-protein interaction (PPI) network of the screened DEGs. Two machine learning algorithms LASSO and SVM-RFE were used to screen the hub genes. We ...

Relationship between changes in blood glucose, glycosylated hemoglobin [HbA1c], cholesterol, and triglyceride and changes in hepatic enzymes
Cell Mol Biol (Noisy-le-grand). 2024 Apr 28;70(4):23-28. doi: 10.14715/cmb/2024.70.4.4.ABSTRACTThe main risk factors for non-alcoholic fatty liver disease (NAFLD) are strongly associated with obesity, diabetes, hyperlipidemia, and metabolic syndrome. The best clinical evaluation of the liver is done through studying changes in liver enzymes' activity, especially alanine aminotransferase, aspartate aminotransferase, and alkaline phosphatase. Therefore, this study aimed to investigate the relationship between changes in factors such as blood glucose, cholesterol, glycosylated hemoglobin, and triglyceride and changes in hepat...

Process Evaluation and Investigation of Cultural Adaptations for an Online Parent-Based Intervention Using a Mixed-Method Approach
AbstractMost alcohol intervention research focuses on program efficacy, yet few studies have investigated the acceptability of a program ’s design and implementation to the target population or adapting existing alcohol interventions to different populations. To address these gaps in the literature, we (1) examined participant responsiveness to and implementation quality of FITSTART+, a web-app delivered parent-based alcohol interv ention designed for incoming first-year college students in the United States, and (2) gathered feedback on how this intervention could be adapted to other populations of parents. A sample of ...
