Synthesis and evaluation of pentacyclic triterpenoids conjugates as novel HBV entry inhibitors targeting NTCP receptor
In this study, pentacyclic triterpenoids (PTs) including glycyrrhetinic acid (GA), oleanolic acid (OA), ursolic acid (UA) and betulinic acid (BA) were modified via molecular hybridization with podophyllotoxin respectively, and resulted in thirty-two novel conjugates. The anti-HBV activities of conjugates were evaluated in HepG2.2.15 cells. The results showed that 66% of the conjugates exhibited lower toxicity to the host cells and had significant inhibitory effects on the two HBV antigens, especially HBsAg. Notably, the compounds BA-PPT1, BA-PPT3, BA-PPT4, and UA-PPT3 not only inhibited the secretion of HBsAg but also supp...

Cellular strategies to induce immune tolerance after liver transplantation: Clinical perspectives
World J Gastroenterol. 2024 Apr 7;30(13):1791-1800. doi: 10.3748/wjg.v30.i13.1791.ABSTRACTLiver transplantation (LT) has become the most efficient treatment for pediatric and adult end-stage liver disease and the survival time after transplantation is becoming longer due to the development of surgical techniques and perioperative management. However, long-term side-effects of immunosuppressants, like infection, metabolic disorders and malignant tumor are gaining more attention. Immune tolerance is the status in which LT recipients no longer need to take any immunosuppressants, but the liver function and intrahepatic histol...

Risk factors for hepatocellular carcinoma associated with hepatitis C genotype 3 infection: A systematic review
CONCLUSION: Although, studies have shown that HCV genotype-3 infection is an independent risk factor for end-stage liver disease, HCC, and liver-related death, there is a lack of evidence for specific risk factors for HCC secondary to HCV genotype-3. Only cirrhosis and age have demonstrated an association; however, the number of studies is very small, and more research is required to investigate risk factors for HCC secondary to HCV genotype-3.PMID:38660636 | PMC:PMC11037048 | DOI:10.4251/wjgo.v16.i4.1596 (Source: Cancer Control)

Is salvage Plug-Assisted Retrograde Transvenous Obliteration (PARTO) safe and effective for bleeding gastric varices ?- A preliminary single-center experience
ConclusionsPARTO is a safe and effective procedure for bleeding gastric varices without any deterioration in liver function even after six months. Patient selection is critical to prevent complications. Further prospective studies with larger sample size are required to validate our findings. (Source: Emergency Radiology)

Every sheriff needs a deputy: targeting non-parenchymal cells to treat hepatic fibrosis
The landscape of anti-fibrotic drug development for liver disease is littered with failed clinical trials. In 2020, the CCR2 and CCR5 receptor antagonist, Cenicriviroc, failed to achieve its primary endpoint of fibrosis improvement of ≥1 stage without worsening of steatohepatitis at 12 months despite promising preclinical and phase 2 data[1]. This failure was particularly disappointing as it was one of the most promising immune-targeted therapeutics to reach phase 3 testing. In 2023, the FXR agonist, obeticholic acid, met the p rimary endpoint that Cenicriviroc could not, but the FDA voted against its approval for treatm...

The contribution of genetics and epigenetics to MAFLD susceptibility
AbstractMetabolic dysfunction-associated fatty liver disease (MAFLD) is the most common liver disease worldwide. The risk of developing MAFLD varies among individuals, due to a combination of environmental inherited and acquired genetic factors. Genome-wide association and next-generation sequencing studies are leading to the discovery of the common and rare genetic determinants of MAFLD. Thanks to the great advances in genomic technologies and bioinformatics analysis, genetic and epigenetic factors involved in the disease can be used to develop genetic risk scores specific for liver-related complications, which can improv...

Exercise, digital health and chronic disease: feasibility, effectiveness and utilisation (PhD Academy Award)
What did I do? My thesis investigated the feasibility, effectiveness and utilisation of digital health physical activity/exercise interventions in chronic disease cohorts. Contemporary literature was reviewed for applicable digital health interventions,1 a feasibility randomised controlled trial was undertaken to evaluate the feasibility of a patient-centred digital health exercise and diet intervention for people living with kidney and liver disease in tertiary care,2 3 and the utilisation of telehealth exercise physiology (EP) services in Australia was examined.4 Why did I do it? Only 25% of all Australian adults aged 18...

Association between sleep duration and a new onset of nonalcoholic fatty liver disease
Conclusions In women, a short sleep duration was independently associated with the onset of NAFLD, thus suggesting that an adequate sleep duration can be a promising preventive factor for the onset of NAFLD in women.PMID:38658342 | DOI:10.2169/internalmedicine.3119-23 (Source: Internal Medicine)
