Use of apixaban in adults with congenital heart disease and atrial arrhythmias: The PROTECT-AR study
Adults with congenital heart disease (ACHD) and atrial arrhythmias (AA) face an increased risk of thromboembolic events. Limited data exist on the use of non-vitamin K oral anticoagulants for thromboprophylaxis in ACHD. We aimed to assess the effectiveness and safety of apixaban in ACHD patients with AA. Randomized controlled trials of switching to a new medication vs continuing a chronic medication may underestimate the risk of adverse events in those continuing because of asymmetric depletion of susceptibles
For older adults with atrial fibrillation (AF), direct oral anticoagulants (DOACs) reduce a person's risk of stroke and bleeding, compared with vitamin k antagonists (VKAs) [1 –3]. However, a recently published randomized controlled trial (RCT) of frail older adults who were on a VKA and randomized to continue a VKA or switch to a DOAC was stopped early for futility due to greater risk of bleeding in adults switched to a DOAC [4]. Some have interpreted this to mean that VKAs are safer than DOACs in frail older adults [5]. (Source: Journal of Clinical Epidemiology)

Combining SSRIs With Oral Anticoagulants Found to Increase Risk of Major Bleeding
People who take serotonin reuptake inhibitors (SSRIs) and oral anticoagulants have an increased risk of multiple types of major bleeding compared with people who take only oral anticoagulants, astudy inJAMA Network Open has found. The study findings also showed that bleeding risk differs depending on the type of anticoagulant.Christel Renoux, M.D., Ph.D., of Jewish General Hospital in Montreal and colleagues examined data from 331,305 patients in the United Kingdom aged 60 years or older who had atrial fibrillation and began taking oral anticoagulants between January 2, 1998, and March 29, 2021. The researchers included an...

Plasma Cell-Free Adenomatous Polyposis Coli Gene Promoter Methylation as a Prognostic Biomarker for Hepatocellular Carcinoma
CONCLUSIONS: Hypermethylation of APC promoter appears to be a potential biomarker that could predict patient survival and disease progression outcome in patients with intermediate stage HCC after local ablation treatment.PMID:38527449 | DOI:10.1159/000538455 (Source: Oncology)

Matrix Gla protein and the long-term incidence and progression of coronary artery and aortic calcification in the Multi-Ethnic Study of Atherosclerosis
CONCLUSIONS: We found a positive association of the inactive form of matrix Gla protein, dp-ucMGP, and long-term incidence/progression of CAC, ATAC, and DTAC. Future studies should investigate dp-ucMGP as a calcification regulator and MGP as a possible therapeutic target to slow progression of calcification in the vasculature.PMID:38527383 | DOI:10.1016/j.atherosclerosis.2024.117505 (Source: Atherosclerosis)

Challenges and lessons learned from a long-term postauthorisation safety study programme of rivaroxaban in Europe
Objectives To describe opportunities and challenges experienced from the four pharmacoepidemiological database studies included in the rivaroxaban post authorisation safety study (PASS) programme and propose ways to maximise the value of population-based observational research when addressing regulatory requirements. Design PASS programme of rivaroxaban carried out as part of the regulatory postapproval commitment to the European Medicines Agency. Setting Clinical practice in Germany, the Netherlands, Sweden and the UK (electronic health records)—undertaken by pharmacoepidemiology research teams using country-speci...

Int J Syst Evol Microbiol. 2024 Mar;74(3). doi: 10.1099/ijsem.0.006298.ABSTRACTA novel bacterial strain, APC 4016T, was previously isolated from the skin of a snub-nosed spiny eel, Notacanthus chemnitzii, from a depth of 1000 m in the northern Atlantic Ocean. Cells were aerobic, cocci, motile, Gram-positive to Gram-variable staining, and gave rise to orange-pigmented colonies. Growth occurred at 4-40 °C (optimum, 25-28 °C), pH 5.5-12 (optimum, pH 7-7.5), and 0-12 % (w/v) NaCl (optimum, 1 %). 16S rRNA gene phylogenetic analysis confirmed that strain APC 4016T belonged to the genus Planococcus and was most closely related ...

Direct oral anticoagulants compared with warfarin in patients with left ventricular thrombus: a cohort study from China
CONCLUSIONS: This study showed no significant difference in thrombus resolution between DOAC and warfarin. DOAC might be an alternative to warfarin for the treatment of LV thrombus. However, further large prospective studies are required to explore the efficacy and safety of DOAC in patients with LV thrombus.PMID:38505052 | PMC:PMC10944738 | DOI:10.21037/jtd-23-1582 (Source: Journal of Thoracic Disease)
