Methionine gamma lyase: Structure-activity relationships and therapeutic applications
Biochim Biophys Acta Proteins Proteom. 2023 Dec 24:140991. doi: 10.1016/j.bbapap.2023.140991. Online ahead of print.ABSTRACTMethionine gamma lyase (MGL) is a bacterial and plant enzyme that catalyzes the conversion of methionine in methanthiol, 2-oxobutanoate and ammonia. The enzyme belongs to fold type I of the pyridoxal 5'-dependent family. The catalytic mechanism and the structure of wild type MGL and variants were determined in the presence of the natural substrate as well as of many sulfur-containing derivatives. Structure-function relationship studies were pivotal for MGL exploitation in the treatment of cancer, bact...

Mental Health Conditions, Including Depression and Stress, Are Associated with Increased Odds of Gastric Cancer-Insights into the Role of Diet: A Case-Control Study
Nutrients. 2023 Nov 30;15(23):4981. doi: 10.3390/nu15234981.ABSTRACTSeveral risk factors, including nutritional/lifestyle ones, play a role in gastric cancer etiology. Further interactions with mental health have also been emphasized. We hypothesized that individuals with mental disorders would exhibit compromised nutrient intake, increasing their risk of gastric cancer. The state of mental health was evaluated in 82 patients with gastric cancer and 95 healthy controls using the 21-item Depression-Anxiety-Stress Scale. The participants' dietary intakes were evaluated by a 168-item food frequency questionnaire. Based on ful...

Trimethylamine N-oxide, β-alanine, tryptophan index, and vitamin B6-related dietary patterns in association with stroke risk
The aim of this study was to examine the associations of dietary patterns derived by reduced-rank regression (RRR) model reflecting variation in novel biomarkers (trimethylamine N-oxide, β-alanine, tryptophan index, and vitamin B6) with stroke risk. Maternal B-vitamin and vitamin D status before, during, and after pregnancy and the influence of supplementation preconception and during pregnancy: Prespecified secondary analysis of the NiPPeR double-blind randomized controlled trial
This study presents one prespecified outcome from the randomized controlled NiPPeR trial, aiming to identify longitudinal patterns of maternal vitamin status from preconception, through early and late pregnancy, to 6 months postdelivery, and determine the influence of vitamin supplementation. Methods and findingsIn the NiPPeR trial, 1,729 women (from the United Kingdom, Singapore, and New Zealand) aged 18 to 38 years and planning conception were randomized to receive a standard vitamin supplement (control;n = 859) or an enhanced vitamin supplement (intervention;n = 870) starting in preconception and continued throughout pr...

GSE191212 Engineering class-B vitamin biosynthesis in Saccharomyces cerevisiae
Contributors : Anna K Wronska ; Thomas Perli ; Marcel van den Broek ; Jean-Marc DaranSeries Type : Expression profiling by high throughput sequencingOrganism : Saccharomyces cerevisiaeSaccharomyces cerevisiae is bradytroph for class B vitamins, it means that yeast cells exhibit slower growth in the absence of an external source of these metabolites. Alleviating these nutritional requirements for optimal growth performance would represent a valuable phenotypic characteristic for industrial strains since this would result in cheaper processes that would also be less susceptible to contaminations. In the present study, sub...

Erythromycin, retapamulin, pyridoxine, folic acid, and ivermectin inhibit cytopathic effect, papain-like protease, and MPRO enzymes of SARS-CoV-2
ConclusionThe IC50 for all the drugs, except ivermectin, was at the clinically achievable plasma concentration in humans, which supports a possible role for the drugs in the management of COVID-19. The lack of inhibition of CPE by ivermectin at clinical concentrations could be part of the explanation for its lack of effectiveness in clinical trials. Sociodemographic, lifestyle, and environmental determinants of vitamin D levels in pregnant women in Spain
CONCLUSIONS: This comprehensive study shows that younger age, higher body mass index, higher deprived areas, higher air pollution and lower UV rays and humidity are associated with lower 25(OH)D3 levels.PMID:37984291 | DOI:10.1016/j.envint.2023.108293 (Source: Environment International)

Crystal structure of an aspartate aminotransferase Lpg0070 from Legionella pneumophila
Biochem Biophys Res Commun. 2023 Nov 10;689:149230. doi: 10.1016/j.bbrc.2023.149230. Online ahead of print.ABSTRACTLegionella pneumophila aspartate aminotransferase (Lpg0070) is a member of the transaminase and belongs to the pyridoxal 5'-phosphate (PLP)-dependent superfamily. It is responsible for the transfer of α-amino between aspartate and α-ketoglutarate to form glutamate and oxaloacetate. Here, we report the crystal structure of Lpg0070 at the resolution of 2.14 Å and 1.7 Å, in apo-form and PLP-bound, respectively. Our structural analysis revealed the specific residues involved in the PLP binding and free form ag...

Research on sweat metabolomics of athlete ’s fatigue induced by high intensity interval training
Conclusion: The change of hypoxanthine concentration in sweat can be used as a good biomarker for the diagnosis of exercise-induced fatigue, while the change of pyruvate content in sweat can be used as a discriminant index for the energy metabolism mode of the body before and after exercise. The main metabolic pathways involved in differential metabolites produced before and after HIIT exercise-induced fatigue are purine metabolism and amino acid metabolism.
