Decreased hepatic enzymes reflect the decreased vitamin B6 levels in Parkinson's disease patients
Serum vitamin B6 level was strongly correlated with total levodopa dose in PD patients. AbstractThe study aims to investigate the vitamin B6 levels in Parkinson's disease (PD) patients and their association with liver enzymes and evaluate how much dysregulation is associated with levodopa dose. Furthermore, to evaluate the effect of Opicapone, a catechol-o-methyl-transferase inhibitor, on vitamin B6 levels by monitoring the AST and ALT levels in patients treated with Levodopa –Carbidopa Intestinal Gel Infusion (LCIG). For these aims, serum vitamin B6 levels were measured (PD,n = 72 and controls,n = 31). The vitam...

Enhanced Physical Stability of L-Ascorbic Acid in an Ionic Liquid
Chem Pharm Bull (Tokyo). 2024 Jan 26. doi: 10.1248/cpb.c23-00861. Online ahead of print.ABSTRACTIonic liquid (IL) technology was used to enhance the stability of L-ascorbic acid (AA). Pyridoxine was selected as the counter cation for anionic AA in IL. After AA was dissolved in water at 40°C, its ratio decreased to 3.2% after 7 days. In contrast, the IL formulation showed negligible degradation, with almost no loss of AA even after 28 days. These results suggest that the use of IL enhances the stability of AA.PMID:38281765 | DOI:10.1248/cpb.c23-00861 (Source: Chemical and Pharmaceutical Bulletin)

Identification of the pyridoxal 5 ′‐phosphate allosteric site in human pyridox(am)ine 5′‐phosphate oxidase
AbstractAdequate levels of pyridoxal 5 ′-phosphate (PLP), the catalytically active form of vitamin B6, and its proper distribution in the body are essential for human health. The PLP recycling pathway plays a crucial role in these processes and its defects cause severe neurological diseases. The enzyme pyridox(am)ine 5 ′-phosphate oxidase (PNPO), whose catalytic action yields PLP, is one of the key players in this pathway. Mutations in the gene encoding PNPO are responsible for a severe form of neonatal epilepsy. Recently, PNPO has also been described as a potential target for chemotherapeutic agents. Our labora tory h...

Genome analysis of “Candidatus Aschnera chinzeii,” the bacterial endosymbiont of the blood-sucking bat fly Penicillidia jenynsii (Insecta: Diptera: Nycteribiidae)
In this study, we report the first complete genome sequence of Aschnera from the bristled bat fly Penicillidia jenynsii. The Aschnera genome consisted of a 748,020 bp circular chromosome and a 18,747 bp circular plasmid. The chromosome encoded 603 protein coding genes (including 3 pseudogenes), 33 transfer RNAs, and 1 copy of 16S/23S/5S ribosomal RNA operon. The plasmid contained 10 protein coding genes, whose biological function was elusive. The genome size, 0.77 Mbp, was drastically reduced in comparison with 4–6 Mbp genomes of free-living γ-proteobacteria. Accordingly, the Aschnera genome was devoid of many important...

Insights into the pathogenesis of primary hyperoxaluria type I from the structural dynamics of alanine:glyoxylate aminotransferase variants
Mutations in alanine:glyoxylate aminotransferase (AGT) leading to protein misfolding cause primary hyperoxaluria type I (PH1). Here, we use hydrogen-deuterium exchange to characterize the structural dynamics of AGT variants. These dynamics are heterogeneously distributed across the structure and different functional sites. Disease-associated variants show local destabilization. Our work provides novel insight into the pathogenic mechanisms causing PH1. Primary hyperoxaluria type I (PH1) is caused by deficient alanine:glyoxylate aminotransferase (AGT) activity. PH1-causing mutations in AGT lead to protein mistargeting and a...

Enzymatic properties of ornithine decarboxylase from Clostridium aceticum DSM1496
In this study, we expressed ODC derived fromC. aceticum DSM1496 inEscherichia coli BL21 (DE3) and thoroughly examined its enzymatic properties. The enzyme has a molecular weight of 55.27  kDa and uses pyridoxal-5′-phosphate (PLP) as a coenzyme with aKm = 0.31 mM. ODC exhibits optimal activity at pH 7.5, and it maintains high stability even at pH 4.5. The peak reaction temperature for ODC is 30°C. Besides, it can be influenced by certain metal ions such as Mn2+. Althoughl-ornithine serves as the preferred substrate for ODC, the enzyme also decarboxylatesl-arginine andl-lysine simultaneously. The results indicate that...
