RXR α cooperates with KLF8 to promote the differentiation of intramuscular preadipocytes in goat.
RXRα cooperates with KLF8 to promote the differentiation of intramuscular preadipocytes in goat. Anim Biotechnol. 2020 Feb 27;:1-11 Authors: Xu Q, Wang Y, Zhang Y, Zhu J, Lin Y Abstract Retinoid X receptor α (RXRα) is thought to be a key regulator in lipid metabolism, glucose metabolism, adipogenic differentiation, gene expression, and inflammatory response. However, it is not clear whether RXRα has any role in intramuscular preadipocyte of goat. In the current investigation, we report that adenovirus overexpression of RXRα promotes lipid accumulation in intramuscular adipocyte and up-regulates t...

Inhibitory effects of msFGFR2c on the epithelial-to-mesenchymal transition of AE2 cells in pulmonary fibrosis.
Abstract In interstitial fibrosis, alveolar epithelial type II (AE2) cells fail to repair damaged epithelium. However, whether this dysfunction is related to fibroblast growth factor (FGF) signal pathway and how it affects the fibrotic process remains unclear. In our study, the medium of the human foetal lung fibroblast cell line MRC-5 (Med) can induce epithelial-to-mesenchymal transition (EMT) in AE2 cells, we also found that TGF-β in Med can induce FGF-2 and CTGF expression in AE2 cells. TGF-β or CTGF exposure trigger a FGFR2 subtype b to c transition which can be supressed by siRNA-CTGF. All together, since F...

Newly synthesized peptide, Ara ‐27, exhibits significant improvement in cell‐penetrating ability compared to conventional peptides
In conclusion, this study describes a novel CPP, Ara‐27, which exhibit significant improvement in intracellular uptake compared to conventional CPPs, without affecting cell viability. Thus, development of Ara‐27 based peptides may lead to imp roved delivery of functional cargo such as small molecules, siRNA, and drugs forin vivo studies. This article is protected by copyright. Mechanical loading induces HIF-1 α expression in chondrocytes via YAP.
CONCLUSION: Our results indicated that cyclic mechanical stress promoted HIF-1α stabilization and YAP is involved in mechanical stress induced HIF-1α up-regulation. PMID: 32451801 [PubMed - as supplied by publisher]

Noncoding RNAs in Medicinal Plants and Their Regulatory Roles in Bioactive Compound Production.
CONCLUSION: So far, scientists have identified thousands of miRNA candidates from over 50 medicinal plant species and 11794 lncRNAs from Salvia miltiorrhiza, Panax ginseng, and Digitalis purpurea. Among them, more than 30 miRNAs and five lncRNAs have been predicted to regulate bioactive compound production. The regulation may achieve through various regulatory modules and pathways, such as the miR397-LAC module, the miR12112-PPO module, the miR156-SPL module, the miR828-MYB module, the miR858-MYB module, and other siRNA and lncRNA regulatory pathways. Further functional analysis of herbal ncRNAs will provide useful informa...

Hepatitis B virus DNA polymerase displays an anti-apoptotic effect by interacting with elongation factor 1-alpha2 in hepatoma cells.
In this study, we aimed at investigating the impact of HBV Pol on host cellular processes, mainly apoptosis, and the underlying mechanisms. We showed a marked reduction in apoptotic rates in the HBV Pol-expressed HepG2 cells compared to controls. Moreover, a series of assays-i.e. yeast two-hybrid, GST pull-down, co-immunoprecipitation, and confocal laser scanning microscopy-identified the host factor eEF1A2 to be associated with HBV Pol. Furthermore, knockdown of the eEF1A2 gene by siRNA abrogated the HBV Pol-mediated anti-apoptotic effect with apoptosis induced by endoplasmatic reticulum (ER) stress inducer thapsigargin (...

Leucine regulates porcine muscle fiber type transformation via adiponectin signaling pathway
Anim Biotechnol. 2021 Mar 11:1-9. doi: 10.1080/10495398.2021.1892709. Online ahead of print.ABSTRACTLeucine can promote slow-twitch muscle fibers formation, and this effect may be mediated by AMPK signaling pathway. In addition, adiponectin (AdipoQ) plays an important role in regulation of muscle fiber type transformation. AdipoQ is located in the upstream of AMPK and its secretion can be regulated by leucine. Therefore, the aim of this study was to explore whether leucine affects muscle fiber type transformation through AdipoQ signaling pathway. Our data showed that 4 mM leucine significantly increased protein expression ...

Procyanidin B2 induces porcine skeletal slow-twitch myofiber gene expression by AMP-activated protein kinase signaling pathway
In this study, our aim is to investigate the effect of dimer procyanidin B2 [epicatechin-(4β-8)-epicatechin] (PB2) on porcine skeletal myofiber gene expression in vitro. Our data showed PB2 promoted the protein expression of slow myosin heavy chain (MyHC) in porcine myotubes, concomitant with the increases in mRNA levels of MyHC I, MyHC IIa and Tnni1. We also found PB2 activated AMPK signaling in porcine myotubes. NRF1 and CaMKKβ that are two important upstream factors of AMPK, and Sirt1 and PGC-1α that are two major downstream factors of AMPK, were also up-regulated by PB2. The mechanism study showed the effect of PB2 ...

An adventitious agent-free clonal cell line that is highly susceptible to foot -and-mouth disease virus
In this study, we developed a three-prong methodology to completely remove BVDV from LFBKαVβ6 cells. Combined treatment with siRNA against BVDV NS5A, porcine interferon alpha and ribavirin resulted in the elimination of BVDV, as determined by immunohistochemistry analysis, quantitative RT-PCR and RNA sequencing. Importantly, elimination of BVDV from LFBKαVβ6 did not affect FMDV growth and plaque phenotype from different serotypes isolated and propagated in the clean cell line, newly named MGPK αVβ6-C5. Additionally, isolation of FMDV from field oro-pharyngeal samples, was successful at the same sensitivity as in BVDV...
