Corin is a key regulator of endochondral ossification and bone development via modulation of vascular endothelial growth factor A expression
This study tested the hypothesis that, through modulation of angiogenic signalling pathways, corin is a critical regulator of osteogenic differentiation and endochondral ossification. In vitro, corin expression in hASC was suppressed via siRNA knockdown and vascular en dothelial growth factor A (VEGF‐A) expression was quantified via reverse transcription polymerase chain reaction. In vivo, a murine corin knockout model (female, 10 weeks) was used to determine the effect of corin deficiency on long bone development. Wild‐type and corin knockout long bones were compared via haematoxylin and eosin staining to assess tiss...

Activation of MET promotes resistance to sorafenib in hepatocellular carcinoma cells via the AKT/ERK1/2-EGR1 pathway.
In this study, we found that addition of HGF to sorafenib-treated HCC cells activated MET and re-stimulated the downstream AKT and ERK1/2 pathways. Thereby, restored sorafenib-treated HCC cells proliferation, migration and invasion ability, and rescued cells from apoptosis. In addition, we found that HGF treatment of HCC cells induced early growth response protein (EGR1) expression, which is involved in sorafenib resistance. Importantly, the HGF rescued effect in sorafenib-treated HCC cells could be abrogated by inhibiting MET activation with PHA-665752 or by downregulating EGR1 expression with small interfering RNA (siRNA...

The application of the CRISPR-Cas9 genome editing machinery in food and agricultural science: Current status, future perspectives, and associated challenges.
Abstract The recent progress in genetic engineering has brought multiple benefits to the food and agricultural industry by enhancing essential characteristics of agronomic traits. Powerful tools in the field of genome editing, such as siRNA-mediated RNA interference for targeted suppression of gene expression and transcription activator-like effector nucleases (TALENs) and zinc-finger nucleases (ZFNs) for DNA repair have been widely used for commercial purposes. However, in the last few years, the discovery of the CRISPR-Cas9 system has revolutionized genome editing and has attracted attention as a powerful tool f...

Expression analysis of proteasome maturation protein (POMP) gene in Liaoning Cashmere goat.
Abstract Liaoning Cashmere goat is a precious genetic resource of China. To explore the relationship between POMP and cashmere growth, we analyzed the expression of POMP. POMP encodes a hudrophilic protein which is most closely related to bos. RT-PCR showed that POMP was expressed in skin, heart, liver, spleen, lung, and kidney tissues. Real-time PCR showed that the expression of POMP was more active in the secondary hair follicles than the primary hair follicles in anagen. In situ hybridization showed that POMP was obviously expressed in the Inner Root Sheath (IRS) but no expression in Outer Root. The treatment o...

Molecular characterization, expression analysis of Chemerin gene and its potential role in intramuscular adipocyte differentiation of goat.
Abstract Chemerin is a cytokine secreted by adipose cells and plays an important regulatory role in adipocyte differentiation and lipid metabolism. However, the function of Chemerin on intramuscular fat (IMF) deposition is still unknown in goat. Here, we cloned the cDNA of goat Chemerin gene using quantitative real-time PCR (qRT-PCR) technique and further clarified the expression pattern of Chemerin in various tissues of goat. The siRNA against Chemerin were synthesized to explore the potential role in intramuscular preadipocyte differentiation of goat. Our results showed that the full-length open reading frame (O...
