A Collagen ‐coated Sponge Silk Scaffold for Functional Meniscus Regeneration
In this study, we fabricate a silk sponge from 9% (w/v) silk fibroin solution through freeze drying, and then coat its internal space and external surface with collagen sponge. Subsequently, various characteristics of the silk ‐collagen scaffold are evaluated, and cytocompatibility of the construct is assessed in vitro and subcutaneously. The efficacy of this composite scaffold for meniscal regeneration is evaluated through meniscus reconstruction in a rabbit meniscectomy model. It is found that the internally coated co llagen sponge enhances the cytocompatibility of the silk sponge, and the external layer of collagen sp...

Reduced SIRT1/AMPK in Amyotrophic Lateral Sclerosis Patient ‐Derived Mesenchymal Stem Cells can be restored by resveratrol
In this study, we examined SIRT1/AMPK activities and identified significant decreases in the ALS‐MSCs compared to normal healthy control origi nated BM‐MSCs. This decline was restored by pre‐treatment with resveratrol (RSV), measured using quantitative PCR, NAD/NADH assay, and immunoblot analysis. Neuro‐progenitor markers were increased in RSV treated ALS‐MSCs (RSV/ALS‐MSCs). The differentiated ALS‐MSCs exhibited a cell body an d dendritic shape similar to neurons. RSV/ALS‐MSCs showed significantly increased differentiation rate as compared to the untreated differentiated ALS‐MSCs (ALS‐dMSCs). The neuri...

Subnormothermic short ‐term cultivation improves the vascularization capacity of adipose tissue‐derived microvascular fragments
In this study we analyzed the effects of normo‐ (37 °C) and subnormothermic (20 °C) short‐term cultivation on their viability and network forming capacity. Ad‐MVF from green fluorescent protein (GFP)+ and GFP‐ C57BL/6 mice were cultivated for 24 h at 37 °C or 20 °C. Freshly isolated, non‐cultivated ad‐MVF served as controls. Number, length, viability, proliferation and angiogenic activity of the ad‐MVF were assessed by microscopic analysis and proteome profiling. GFP+ ad ‐MVF were seeded onto collagen‐glycosaminoglycan matrices, which were implanted into dorsal skinfold chambers of GFP‐ ...

Tissue ‐engineered nanoclay based 3D in vitro breast cancer model for studying breast cancer metastasis to bone
AbstractBreast cancer (BrCa) preferentially spreads to bone and colonizes within the bone marrow to cause bone metastases. To improve the outcome of patients with breast cancer bone metastasis, we need to understand better the mechanisms underlying bone metastasis. Researchers have relied heavily uponin vivo xenografts due to limited availibility of human bone metastasis samples. A significant limitation of these is that they do not have a human bone microenvironment. To address this issue, we have developed a nanoclay based 3Din vitro model of breast cancer bone metastasis using human breast cancer cells mimicking late st...

Corin is a key regulator of endochondral ossification and bone development via modulation of vascular endothelial growth factor A expression
This study tested the hypothesis that, through modulation of angiogenic signalling pathways, corin is a critical regulator of osteogenic differentiation and endochondral ossification. In vitro, corin expression in hASC was suppressed via siRNA knockdown and vascular en dothelial growth factor A (VEGF‐A) expression was quantified via reverse transcription polymerase chain reaction. In vivo, a murine corin knockout model (female, 10 weeks) was used to determine the effect of corin deficiency on long bone development. Wild‐type and corin knockout long bones were compared via haematoxylin and eosin staining to assess ...

Electrosprayed extracellular matrix nanoparticles induce a pro ‐regenerative cell response
AbstractDecellularized extracellular matrix (ECM) is an effective tissue repair scaffold. Additionally, ECM has recently been shown to be protective to the lungs. However, current processing is inadequate for effective delivery of ECM to the lungs. Processing methods, such as milling, produce large variability in particle sizes. The size variation produced is ineffective at treating the lung because only a small range of sizes reach the distal regions of the alveoli. The aim of this work is to formulate decellularized ECM to reach the distal lung while retaining the pro ‐regenerative effects of ECM. We first digested the...

The effect of alginate –gelatin encapsulation on the maturation of human myelomonocytic cell line U937
AbstractToday, many attempts have been collected in the field of tissue engineering for reconstitution of injured bone marrow capacity by transplantation of functional cell source. By having a three ‐dimensional condition, microcapsules are appropriate candidates for cells transplantation to target sites. Here, we examined the effect of alginate–gelatin microcapsules on functional maturation of human myelomonocytic cell line U937 after 7 days in vitro. U937 cells were encapsulated by the m ixture of alginate–gelatin and cultured for 7 days. Trypan blue staining was used to show cell survival rate. M...

Macrophage functions in wound healing
AbstractMacrophages play a crucial role in regeneration and consecutive phases of wound healing. In this review we summarize current knowledge on the ontogeny, origin, phenotypical heterogeneity and functional exchangeability of macrophages participating in these processes. We also describe the genetic, pharmacologic and bioengineering methods for manipulation of macrophage phenotype and functions and their potential for development of the novel, clinically applicable, therapies. Tuning Forkhead Box D3 overexpression to promote specific osteogenic differentiation of human embryonic stem cells while reducing pluripotency in a three ‐dimensional culture system
AbstractClinical use of human embryonic stem cells (hESCs) in bone regeneration applications requires that their osteogenic differentiation be highly controllable as well as time ‐ and cost‐effective. The main goals of the current work were thus (a) to assess whether overexpression of pluripotency regulatorForkhead Box D3 (FOXD3) can enhance the osteogenic commitment of hESCs seeded in three ‐dimensional (3D) scaffolds and (b) to evaluate if the degree ofFOXD3 overexpression regulates the strength and specificity of hESC osteogenic commitment. In conducting these studies, an interpenetrating hydrogel network consisti...

Hydrogels in adipose tissue engineering —Potential application in post‐mastectomy breast regeneration
AbstractCurrent methods of breast reconstruction are associated with significant shortcomings, including capsular contracture, infection, rupture, the need for reoperation in implant ‐based reconstruction, and donor site morbidity in autologous reconstruction. These limitations result in severe physical and psychological issues for breast cancer patients. Recently, research has moved into the field of adipose tissue engineering to overcome these limitations. A wide range of re generative strategies has been devised utilising various scaffold designs and biomaterials. A scaffold capable of providing appropriate biochemica...

Temporal Effects of Cytokine Treatment on Lubricant Synthesis and MMP Activity of Fibroblast ‐like Synoviocytes
This study assessed the HA, lubricin, and matrix metalloproteinase‐2 (MMP‐2) secretion profile of FLS over 12 days of culture. FLS were subjected to continuous, intermittent, and s equential cytokine treatments of interleukin‐1 beta (IL‐1β), tumor necrosis factor‐alpha (TNF‐α), and transforming growth factor‐beta 1 (TGF‐β1). HA was assessed via an ELISA‐like assay for content, and agarose gel electrophoresis for molecular weight distribution. Relative lubricin content was determined by western blot. Pro MMP‐2 and active MMP‐2 were quantified by gelatin zymography. All intermittent and ...
