p38 MAPK activation and STIM1-Orai3 association mediate TRPC6 externalization
Am J Physiol Cell Physiol. 2023 Apr 24. doi: 10.1152/ajpcell.00425.2022. Online ahead of print.ABSTRACTEndothelial cell (EC) migration is critical for the repair of monolayer disruption following angioplasties, but migration is inhibited by lipid oxidation products, including lysophosphatidylcholine (lysoPC), which open canonical transient receptor potential 6 (TRPC6) channels. TRPC6 activation requires an increase in intracellular Ca2+ concentration ([Ca2+]i), the source of which is unknown. LysoPC can activate phospholipase A2 to release arachidonic acid (ArA). ArA can activate arachidonic acid-regulated calcium (ARC) ch...

Tumor suppressor miR-218 directly targets epidermal growth factor receptor (EGFR) expression in triple-negative breast cancer, sensitizing cells to irradiation
CONCLUSION: This study characterizes the antagonistic relationship between miR-218 and EGFR. It also demonstrates downstream functional effects of miR-218 overexpression, leading to anti-tumorigenic cellular changes.PMID:37088795 | DOI:10.1007/s00432-023-04750-x

Visfatin-induced upregulation of lipogenesis via EGFR/AKT/GSK3 β pathway promotes breast cancer cell growth
Cell Signal. 2023 Apr 19:110686. doi: 10.1016/j.cellsig.2023.110686. Online ahead of print.ABSTRACTBreast cancer (BC) incidence and associated mortality have increased in tandem with the growth in obesity among the females worldwide. An adipokine, visfatin, has been shown to potentially impact glucose, lipid, and protein metabolism, and promote cancer growth however, the mechanism underlying the effect of visfatin on lipid metabolism dysregulation contributing to BC cell survival, proliferation, and metastasis has not been elucidated. Herein, we have investigated the role of visfatin on the induction of Sterol regulatory e...

USP7 reduction leads to developmental failure of mouse early embryos
Exp Cell Res. 2023 Apr 18;427(2):113605. doi: 10.1016/j.yexcr.2023.113605. Online ahead of print.ABSTRACTAs a member of Ubiquitin-specific protease subfamily, ubiquitin specific protease 7 (USP7) has been reported to participate in a variety of cellular processes, including cell cycle, apoptosis, DNA damage response, and epigenetic modification. However, its function in preimplantation embryos is still obscure. To investigate the functions of USP7 during preimplantation embryo development, we used siRNA to degrade endogenous USP7 messenger RNA. We found that USP7 knockdown significantly decreased the development rate of mo...

IL-1 β stimulates a novel, IKKα -dependent, NIK -independent activation of non-canonical NFκB signalling
In this study, we examined the activation of non-canonical nuclear factor Kappa B (NFκB) signalling in U2OS cells, a cellular metastatic bone cancer model. Whilst Lymphotoxin α1β2 (LTα1β2) stimulated the expected slow, delayed, sustained activation of serine 866/870 p100 phosphorylation and increased cellular expression of p52 NFκB, we found that canonical agonists, Interleukin-1β (IL-1β) and also Tumour necrosis factor-α (TNFα) generated a rapid transient increase in pp100, which was maximal by 15-30 min. This rapid phosphorylation was also observed in other cells types, such as DU145 and HCAECs suggesting the p...

MYC induces immunotherapy and interferon- γ resistance through downregulation of JAK2
Cancer Immunol Res. 2023 Apr 19:CIR-22-0184. doi: 10.1158/2326-6066.CIR-22-0184. Online ahead of print.ABSTRACTImmunotherapy has revolutionized the treatment of advanced melanoma. Because the pathways mediating resistance to immunotherapy are largely unknown, we conducted transcriptome profiling of pre-immunotherapy tumor biopsies from melanoma patients that received PD-1 blockade or adoptive cell therapy with tumor-infiltrating lymphocytes. We identified two melanoma-intrinsic, mutually exclusive gene programs, which were controlled by interferon-γ (IFNγ) and MYC, and the association with immunotherapy outcome. MYC-over...

The HDAC inhibitor domatinostat induces type I interferon α in Merkel cell carcinoma by HES1 repression
CONCLUSION: Our results demonstrate that the direct anti-tumor effect of HDACi domatinostat on MCC cells is at least in part mediated via decreased HES1 expression allowing the induction of IFNα, which in turn causes apoptosis.PMID:37071208 | DOI:10.1007/s00432-023-04733-y
