Persistent Opioid Use After Hospital Admission From Surgery in New Zealand: A Population-Based Study
CONCLUSIONS: Approximately 1 in 11 opioid-naïve patients who were dispensed opioids on surgical discharge, developed POU. Potentially modifiable risk factors for POU, related to how opioids were prescribed included changing opioids after discharge, receiving multiple opioids, and higher total dose of opioids given on discharge. Clinicians should discuss the possibility of developing POU with patients before and after surgery and consider potentially modifiable risk factors for POU when prescribing analgesia on discharge after surgery.PMID:38493440 | DOI:10.1213/ANE.0000000000006911 (Source: Anesthesia and Analgesia)

Does morphine attenuate the Inhibition of Platelet Aggregation for both oral and parenteral P2Y12 inhibitors?
Cangrelor is an intravenous P2Y12 inhibitor characterized by rapid and reversible Inhibition of Platelet Aggregation (IPA) which was shown to reduce the risk of periprocedural ischemic complications in patients undergoing percutaneous coronary intervention (PCI) [1]. Pivotal studies investigating pharmacodynamic (PD) and clinical effects of cangrelor included a relatively low proportion of patients with ST-segment elevation myocardial infarction (STEMI), albeit in this setting cangrelor showed treatment effects which were consistent with the overall trial results [1]. Paynantheine more effectively reverses multidrug resistance in malignant EPG85.257RDB and MCF7MX cells than morphine and speciociliatine
Cell Biol Int. 2024 Mar 13. doi: 10.1002/cbin.12152. Online ahead of print.ABSTRACTThe possible interactions of morphine, paynantheine and speciociliatine alkaloids with ATP-binding cassette (ABC) transporters was investigated. The compounds were docked against ABCG2 and ABCB1 to predict the binding mode of alkaloids in active binding sites. The cytotoxicity of morphine, paynantheine and speciociliatine for EPG85.257RDB and MCF7MX cells was determined and ABCB1 and ABCG2 gene and protein expression were determined. The binding score of paynantheine to ABCB1 was higher in the docking studies. Paynantheine and speciociliatin...
