Histamine Potentiates SARS-CoV-2 Spike Protein Entry Into Endothelial Cells
Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) which causes coronavirus disease (COVID-19) is one of the most serious global health crises in recent history. COVID-19 patient symptoms range from life-threatening to mild and asymptomatic, which presents unique problems in identifying, quarantining, and treating the affected individuals. The emergence of unusual symptoms among survivors, now referred to as “Long COVID”, is concerning, especially since much about the condition and the treatment of it is still relatively unknown. Evidence so far also suggests that some of these symptoms can be attributed to v...

Famotidine activates the vagus nerve inflammatory reflex to attenuate cytokine storm
Conclusions. These observations reveal a previously unidentified vagus nerve-dependent anti-inflammatory effect of famotidine in the setting of cytokine storm which is not replicated by high dosages of other H2R antagonists in clinical use. Because famotidine is more potent when administered intrathecally, these findings are also consistent with a primarily central nervous system mechanism of action.PMID:35441176 | PMC:PMC9016653 | DOI:10.21203/rs.3.rs-1493296/v1 (Source: Cell Research)

Oral famotidine versus placebo in non-hospitalised patients with COVID-19: a randomised, double-blind, data-intense, phase 2 clinical trial
Conclusions Famotidine was safe and well tolerated in outpatients with mild to moderate COVID-19. Famotidine led to earlier resolution of symptoms and inflammation without reducing anti-SARS-CoV-2 immunity. Additional randomised trials are required. (Source: Gut)

Montmorillonite-Famotidine/Chitosan Bio-nanocomposite Hydrogels as a Mucoadhesive/Gastro-Retentive Drug Delivery System
The main purpose of the present study was to fabricate the mucoadhesive bio-nanocomposite hydrogels to prolong the drug retention time in the stomach. In these bio-nanocomposite hydrogels, chitosan (CH) was used as the bioadhesive matrix, montmorillonite (MMT) was applied to modulate the release rate, and tripolyphosphate (TPP) was the crosslinking agent. The test samples analyzed via different methods such as XRD, FT-IR, TGA, and SEM. Drug incorporation efficacy and mucoadhesive strength of these nanocomposite hydrogel beads were studied. Swelling and in vitro drug release behaviors of these bio-nanocomposite hydrogels we...

Isolated Hypoglossal Neuropathy due to Cervical Spondylosis Mimicking Lingual Angioedema
A woman in her 80s presented to the emergency department with a 4-month history of “tongue swelling,” mild drooling, difficulty in swallowing, and slurred speech that worsened during 1 day. Asymmetric lingual angioedema predominantly of the right side of the tongue secondary to lisinopril was suspected. The patient was treated with diphenhydramine, dexamethasone, famotidine, a nd tranexamic acid, then admitted to the hospital for observation. Lisinopril was discontinued after 4 years of treatment. A Novel Immune-Related Prognostic Signature Predicting Survival in Patients with Pancreatic Adenocarcinoma
J Oncol. 2022 Mar 18;2022:8909631. doi: 10.1155/2022/8909631. eCollection 2022.ABSTRACTPancreatic adenocarcinoma (PAAD) carries the lowest survival rate of all major organ cancers, which is of dismal prognosis and high mortality rate. Thus, the present study attempted to identify a few novel prognostic biomarkers and establish an immune-related prognostic signature which could predict the prognosis of PAAD. Four prognostic immune-related genes (IRGs) including S100A6, S100A10, S100A16, and SDC1 were screened by differentially expressed gene (DEG) identification and weighted gene coexpression network analysis (WGCNA). Subse...

DDSolver Software Application for Quantitative Analysis of In Vitro Drug Release Behavior of the Gastroretentive Floating Tablets Combined with Radiological Study in Rabbits
CONCLUSION: DDSolver software was helpful in deciding the optimized formulation of FMT floating tablets. The radiological examination in rabbits for gastric retention was consistent with the release data analysis in vitro.PMID:35249487 | DOI:10.2174/1567201819666220304203014 (Source: Current Drug Delivery)

Valproic acid –associated proximal tubulopathy
A 6-year-old Asian boy with ring chromosome 14, global developmental delay, seizure disorder, asthma, chronic respiratory failure requiring ventilatory support, and gastrostomy tube dependence presented with acute kidney injury and Fanconi syndrome. He had been treated with valproic acid (800 mg daily via gastrostomy tube) for 3.5 years for seizure disorder. Other medications included zonisamide, clobazam, fluticasone, albuterol, montelukast, and famotidine. (Source: Kidney International)

A Nanocrystal Platform Based on Metal-Phenolic Network Wrapping for Drug Solubilization
This study reports an exquisite platform for nanocrystal production based on a metal-phenolic network (MPN). MPN-wrapped nanocrystal particles (MPN-NPs) were fabricated through an anti-solvent precipitation method using tannic acid and FeIII or AlIII as coupling agents and characterized by dynamic light scattering, transmission electron microscope, ultraviolet and visible spectrophotometry, fourier-transform infrared spectroscopy, and X-ray powder diffraction. In vitro release, cytotoxicity, and stability were mainly studied with MPN-NPs loading paclitaxel. The suitability of MPN as a nanocrystal stabilizer was also invest...
