Biomaterial-mediated intracellular control of macrophages for cell therapy in pro-inflammatory and pro-fibrotic conditions
We present a tunable biomaterial-based strategy to control macrophages intracellularly via small molecule-releasing microparticles. Poly(lactic-co-glycolic acid) microparticles encapsulating the anti-inflammatory and anti-fibrotic drug dexamethasone were administered to macrophages in vitro, with uptake rates controlled by different loading regimes. Microparticle dose and dexamethasone content directly affected macrophage phenotype and phagocytic capacity, independent of particle content per cell, leading to an overall pro-reparative, anti-inflammatory, anti-fibrotic phenotype with increased phagocytic and ECM degrading fu...

Combined treatment of dexamethasone mouthwash and low ‐level laser therapy in the management of aphthous‐like ulcers caused by nonsteroidal anti‐inflammatory drugs: A case report
Key Clinical MessageA case of major aphthous-like ulcer was described in a 50-year-old patient. The patient showed the main signs of aphthous stomatitis painful ulcer, 1 –2 cm in diameter, located on the ventral of the tongue, buccal mucosa, and the palate. These ulcers persisted for more than 3 weeks. The patient's self-administration of a nonsteroidal anti-inflammatory drug (NSAID) was suggested as the leading cause of aphthous-like ulcers in this case. ulce rs were treated with dexamethasone mouthwash and low-level laser therapy (LLLT). Bilateral Retinal Venous Occlusion in Atypical Hemolytic-Uremic Syndrome Due to Complement Factor H Mutation
CONCLUSION: Diagnosis of aHUS is challenging especially during pregnancy and the postpartum period. Although ocular involvement is quite rare, we described bilateral RVO in aHUS case with homozygous nonsense mutation (c.2134 G > T p.G712). Dexamethasone implant should be considered for the treatment of RVO in aHUS cases.PMID:38536978 | DOI:10.1080/09273948.2024.2333401 (Source: Ocular Immunology and Inflammation)

Bruton Tyrosine Kinase Inhibition: an Effective Strategy to Manage Waldenstr öm Macroglobulinemia
AbstractPurpose of ReviewThe treatment of Waldenstr öm macroglobulinemia (WM) has evolved over the past decade. With the seminal discoveries ofMYD88 andCXCR warts, hypogammaglobulinemia, infections, and myelokathexis (WHIM) mutations in WM cells, our understanding of the disease biology and treatment has improved. The development of a new class of agents, Bruton tyrosine kinase inhibitors (BTKi), has substantially impacted the treatment paradigm of WM. Herein, we review the current and emerging BTKi and the evidence for their use in WM.Recent FindingsClinical trials have established the role of covalent BTKi in the treatm...

Management of Trastuzumab Deruxtecan-related nausea and vomiting in real-world practice
CONCLUSIONS: The NK1 regimen improved vomiting control at cycle 1 and, when introduced at cycle 2, significantly improved both nausea and vomiting. The biased NK1 selection for higher-risk patients may have dampened the differences between groups at cycle 1. These findings support enhanced control of T-DXd-related nausea and vomiting with NK1RA.PMID:38529378 | PMC:PMC10961432 | DOI:10.3389/fonc.2024.1374547 (Source: Cancer Control)

Dehydrozaluzanin C- derivative protects septic mice by alleviating over-activated inflammatory response and promoting the phagocytosis of macrophages
In this study, we assessed the effect of dehydrozaluzanin C-derivative (DHZD), a modified compound from dehydrozaluzanin C (DHZC), as a potential HDT agent for severe infection. LPS-induced septic mouse model and Carbapenem resistant Klebsiella pneumoniae (CRKP) infection mouse model was used for testing in vivo. RAW264.7 cells, mouse primary macrophages, and DCs were used for in vitro experiments. Dexamethasone (DXM) was used as a positive control agent. DHZD ameliorated tissue damage (lung, kidney, and liver) and excessive inflammatory response induced by LPS or CRKP infection in mice. Also, DHZD improved the hypothermic...
