A cost-effective and labor-saving method for detecting HLA-B27 status via sequence-encoded fluorescence amplification assay
Identification of Human leukocyte antigen B27 (HLA-B27) by flow cytometry (FCM) has been widely applied in clinical practice for auxiliary diagnosis of ankylosing spondylitis (AS). However, FCM requires fresh-prepared samples and relies on expensive equipment, reagents, and experienced operator. To provide a cheaper and more convenient method for HLA-B27 detection, we proposed a new method termed sequence-encoded fluorescence amplification assay (SEFA), which specially recognized sequences of HLA-B27 gene (HLA-B*27) covering current common subtypes in a single closed tube. Immune-mediated inflammatory disorders (IMIDs) including inflammatory bowel disease (IBD), rheumatoid arthritis (RA), psoriasis/psoriatic arthritis (PSO), and ankylosing spondylitis (AS), are associated with increased risk of major adverse cardiovascular events (MACE). There is uncertainty regarding the cardiovascular safety of newer medications for IMIDs such as biologics and small molecules (anti-IL-23, anti-IL-12/23, anti-TNFs, and JAKi). This network meta-analysis pooled 40 studies (36 RCTs) that reported MACE (myocardial infarction, cerebrovascular accident, unstable angina, cardiovascular death, or heart failure) in ...

The cost-effectiveness of a bimekizumab versus IL-17A inhibitors treatment-pathway in patients with active axial spondyloarthritis in Scotland
Conclusions remained unchanged in most scenarios. Results' drivers included BASDAI50 response rate and disease management cost.CONCLUSION: The bimekizumab treatment-pathway represents a cost-effective option across the axSpA disease spectrum in Scotland. Bimekizumab is cost-effective compared to a secukinumab-pathway that includes dose up-titration, and has similar costs and QALYs to an ixekizumab-pathway.PMID:38650583 | DOI:10.1080/13696998.2024.2342209 (Source: Journal of Medical Economics)

Programmed cell death in autoimmune diseases: ferroptosis
Ann Biol Clin (Paris). 2024 Apr 19;82(1):33-42. doi: 10.1684/abc.2024.1866.ABSTRACTFerroptosis is an iron dependent cell death driven by lipid peroxidation. Over the past decade, increasing evidence has confirmed that ferroptosis plays an irreplaceable role in the occurrence and development of many diseases, including various cancers, neurodegenerative diseases, cardiovascular diseases and autoimmune diseases. Autoimmune disease is an inflammatory disease characterized by the breakdown of immune tolerance. Nowadays, accumulating evidence indicates that ferroptosis is closely related to the pathogenesis of autoimmune diseas...

Extensive colitis associated with ixekizumab
We present the clinical case of a 56-year-old former smoker female, with a family history of maternal ulcerative colitis and personal history of ankylosing spondylitis treated with Ixekizumab for 3 months, who was admitted for fever, left iliac fossa pain and diarrhea without pathological products of 2 weeks of evolution. Abdominopelvic computed tomography scan identified pancolitis. Complete colonoscopy revealed continuous involvement of the proximal colon (right and transverse colon presented deep ulcerations and mucosal friability) with preservation of the terminal ileum. After many complementary tests and according to ...

Lower total cholesterol and triglyceride levels in ankylosing spondylitis than non-inflammatory rheumatic disease controls in a 1978-98 study: a potential effect of increased physical energetics in manual occupations in the pre-2000 chronologic era
CONCLUSIONS: In the 1978-98 survey, AS had significantly lower mean serum cholesterol and triglyceride levels than NIRD control patients. Associated manual labour occupations may have significantly contributed to results, possibly related to increased energy expenditures from physical activity in the pre-2000 era.PMID:38634375 | DOI:10.55563/clinexprheumatol/u41mn1 (Source: Clinical and Experimental Rheumatology)

Exploring the Effects of Ixekizumab on Pain in Patients with Ankylosing Spondylitis Based on Objective Measures of Inflammation: Post Hoc Analysis from a Large Randomized Clinical Trial
ConclusionThis post hoc analysis is supportive of the hypothesis that ixekizumab reduces pain in AS by additional mechanisms other than the reduction of measurable inflammation.Trial Registration NumberNCT02696785. (Source: Rheumatology and Therapy)

Angiogenesis-related immune response may be the prelude to the syndesmophyte formation in Ankylosing spondylitis
CONCLUSIONS: We explored the intimate relationship between local inflammation and bone formation in AS from the perspective of nascent vascularisation. Hence, our study lays the foundation for elucidating a unified AS pathogenesis through the immune-angiogenesis-osteogenesis axis.PMID:38631220 | DOI:10.1016/j.intimp.2024.112040 (Source: International Immunopharmacology)
