Synthesis and characterization of different sodium hyaluronate nanoparticles to transport large neurotherapheutic molecules through blood brain barrier after stroke
Publication date: Available online 16 January 2019Source: European Polymer JournalAuthor(s): Sebastián Peralta, Santos Blanco, Raquel Hernández, Herminia Castán, Eva Siles, Esther Martínez-Lara, María Encarnación Morales, María Ángeles Peinado, María Adolfina RuizAbstractSome biological drugs with proven neuroprotective capacity are unable to cross the blood brain barrier (BBB), preventing its use in neuroregenerative diseases such as stroke. The use of nanoparticles as a delivery system to transport large therapeutic molecules to the cerebral parenchyma may be a good option to overcome this limitation. To achieve ...

Stable isotope labeling derivatization and magnetic dispersive solid phase extraction coupled with UHPLC-MS/MS for the measurement of brain neurotransmitters in post-stroke depression rats administrated with gastrodin.
Abstract Synchronous measurement of brain neurotransmitters and blood drug metabolism by dual sites in vivo microdialysis can provide very important information for several neurological disorders such as post-stroke depression (PSD). In this work, d0/d3-6-N-methyl-rhodamine 6G-N-hydroxysuccinimidyl formate (d0/d3-MRSF) were firstly designed, synthesized and used as stable isotope labeling derivatization reagents for the determination of multiple neurotransmitters by ultra-high performance liquid chromatography-tandem mass spectrometry. The light d0-MRSF was used to label neurotransmitters in real samples and the ...

Omentin-1: protective impact on ischemic stroke via ameliorating atherosclerosis.
This study is the first to summarize the comprehensive mechanisms between omentin-1 and atherosclerosis and to review the shielding effect of omentin-1 on IS. We shed light on omentin-1 as a novel therapeutic target for combating IS. PMID: 33607071 [PubMed - as supplied by publisher]

Inflammatory Molecular Mediators and Pathways Involved in Vascular Aging and Stroke: A Comprehensive Review
Curr Med Chem. 2021 Sep 1. doi: 10.2174/0929867328666210901122359. Online ahead of print.ABSTRACTThere is an increase in the incidence of cardiovascular diseases with aging and it is one of the leading causes of death worldwide. The main cardiovascular pathologies include atherosclerosis, stroke, myocardial infarction, hypertension and stroke. Chronic inflammation is one of the significant contributors to the age-related vascular diseases. Therefore, it is important to understand the molecular mechanisms of the persistent inflammatory conditions occurring in the blood vessels as well as the signaling pathways involved. Her...

Molecules, Vol. 26, Pages 6365: Lack of Neuroprotective Effects of High-Density Lipoprotein Therapy in Stroke under Acute Hyperglycemic Conditions
Conclusion: We observed a nonsignificant decrease in cerebral infarct size in the HDL group. The deleterious consequences of reperfusion such as hemorrhagic transformation or inflammation were not improved by HDL infusion. In acute hyperglycemia, HDLs are not potent enough to counteract the adverse effects of hyperglycemia. The addition of antioxidants to therapeutic HDLs could improve their neuroprotective capacity.

Discovery of novel brain-penetrant GluN2B NMDAR antagonists via pharmacophore-merging strategy as anti-stroke therapeutic agents
Eur J Med Chem. 2021 Oct 8;227:113876. doi: 10.1016/j.ejmech.2021.113876. Online ahead of print.ABSTRACTIn this work, a novel structural series of brain-penetrant GluN2B NMDAR antagonists were designed, synthesized and biologically evaluated as anti-stroke therapeutic agents via merging the structures of NBP and known GluN2B ligands. Approximately half of them exhibited superior neuroprotective activity to NBP against NMDA-induced neurotoxicity in hippocampal neurons at 10 μM, and compound 45e and 45f exerted equipotent activity to ifenprodil, an approved GluN2B- selective NMDAR antagonist. In particular, 45e, with the mo...
