A hemorrhagic transformation model of mechanical stroke therapy with acute hyperglycemia in mice
Aim: Clinical benefit for mechanical thrombectomy (MT) in stroke was recently demonstrated in multiple large prospective studies. The aim of this therapy is to achieve a complete reperfusion in a short delay, given that reperfusion damage is dependent on the time of ischemia. Acute hyperglycemia is an important risk factor of poor outcome in stroke patients, including those that underwent MT. Here, we investigated the effect of acute hyperglycemia in a mouse model of ischemic stroke subjected to different times (30, 90 and 180 min) of middle cerebral artery occlusion (MCAO).

Aim: Anti-platelet therapy with aspirin (ASA) reduces nonfatal myocardial infarction, stroke, and cardiovascular death by irreversibly inhibiting platelet COX-1 and TxA2 production. Activation of protease-activated receptors in patients with new onset of atrial fibrillation contributes to systemic atherosclerotic risk
Aim: Atrial fibrillation (AF) remains one of the major causes of stroke, heart failure, sudden death, and cardiovascular morbidity in the world. A complex array of pathophysiological changes including fibrosis and inflammation increasing the propensity of the atria to develop or maintain AF. At the same time, these alterations are involved in the occurrence of the systemic hypercoagulable state in which protease-activated receptors (PARs) 1 and 2 are discussed to be mediators. We wanted to evaluate the pathomechanistic link between inflammation, hypercoagulability and the atherosclerotic risk in the context of PAR1 and PAR2 activation.

Intraindividual variability of lipoprotein(a) and implications for the decision-making process for lipoprotein(a) lowering therapy
Aim: Lipoprotein (a) is an independent risk factor for myocardial infarction, stroke and calcific aortic valve stenosis. Circulating Lp(a) levels are largely genetically determined by SNP and a wide heterogeneity in kringle IV type 2 repeats. Furthermore, significant racial differences exist. Lifestyle modifications like nutrition and sports have no or less effect on Lp(a) serum levels.

Vulnerable atherosclerotic plaque model in atherosclerotic swine and a potential target site for intervention
Aim: Unstable/vulnerable plaques are characterized by a thin cap fibroatheroma and necrotic core, and may result in serious clinical conditions, including transient ischemic attack, stroke, aphasia, and other motor defects. The precise mechanism of plaque rupture remains to be defined to identify target site(s) for intervention. Here, we report a novel swine model of vulnerable plaque and identified a potential target site for intervention.

PET-CT imaging of atherosclerosis in Ldlr-/- mice treated with an anti-LDL(-) nanoformulation
Aim: Atherosclerosis is a chronic inflammatory disease underlying the myocardial infarction and ischemic stroke. The electronegative low-density lipoprotein (LDL-), an in vivo modified subfraction of LDL, is pro-inflammatory and plays a key role in atherosclerosis contributing to foam cells formation. In the nanotechnology field, some strategies have emerged such as the use of nano-carriers with synthetic polymer nanoparticles and nanoparticle-protein conjugates as promising approaches for the prevention and treatment of diseases.

Secretory phospholipase A2 modified high-density lipoprotein potently suppresses platelet activation
Aim: Levels of secretory phospholipases A2 (sPLA2) highly increase under acute and chronic inflammatory conditions. sPLA2 is mainly associated with high-density lipoproteins (HDL) and generates bioactive lysophospholipids implicated in acute and chronic inflammatory processes.
