Targeted Nanoparticle for Co ‐delivery of HER2 siRNA and a Taxane to Mirror the Standard Treatment of HER2+ Breast Cancer: Efficacy in Breast Tumor and Brain Metastasis
This research paper describes a new therapeutic candidate based on a nanoparticle that co-delivers siRNA against human epidermal growth factor receptor type 2 (HER2), trastuzumab, and docetaxel to treat advanced HER2+ breast cancer. The nanotherapeutic is more effective and safer than the free drug counterparts. It inhibits drug-resistant orthotopic and brain-metastatic HER2+ breast tumors in mice. AbstractThe first-line treatment of advanced and metastatic human epidermal growth factor receptor type 2 (HER2+) breast cancer requires two HER2-targeting antibodies (trastuzumab and pertuzumab) and a taxane (docetaxel or pacli...

SHMT1 siRNA-Loaded hyperosmotic nanochains for blood-brain/tumor barrier post-transmigration therapy
Biomaterials. 2021 Dec 30;281:121359. doi: 10.1016/j.biomaterials.2021.121359. Online ahead of print.ABSTRACTThe near-perivascular accumulation in solid tumors and short-lived span in circulation, derails even the most competent nanoparticles (NPs) from achieving their maximum therapeutic potential. Moreover, delivering them across the blood brain/tumor barrier (BBB/BTB) is further challenging to sought anticancer effect. To address these key challenges, we designed a linearly aligned nucleic acid-complexed polydixylitol-based polymeric nanochains (X-NCs), with inherent hyperosmotic properties enabling transmigration of th...

EZH2 as a new therapeutic target in brain tumors: Molecular landscape, therapeutic targeting and future prospects
Biomed Pharmacother. 2021 Dec 11;146:112532. doi: 10.1016/j.biopha.2021.112532. Online ahead of print.ABSTRACTBrain tumors are responsible for high mortality and morbidity worldwide. The brain tumor treatment depends on identification of molecular pathways involved in progression and malignancy. Enhancer of zeste homolog 2 (EZH2) has obtained much attention in recent years in field of cancer therapy due to its aberrant expression and capacity in modulating expression of genes by binding to their promoter and affecting methylation status. The present review focuses on EZH2 signaling in brain tumors including glioma, gliobla...

Enhanced expression of Pentraxin-3 in glioblastoma cells correlates with increased invasion and IL8-VEGF signaling axis
In this study, we examined the role of PTX3 in GB growth, angiogenesis, and invasion using in vitro and in vivo GB models, proteomic profiling, molecular and biochemical approaches. Under in vitro conditions, PTX3 over-expression in U87 cells correlated with cell cycle progression, increased migratory potential, and proliferation under hypoxic conditions. Conditioned media containing PTX3 enhanced the angiogenic potential of endothelial cells. While silencing of PTX3 by siRNA decreased the proliferation, migration, and angiogenic potential of U87 cells in vitro. Importantly, PTX3 over-expression increased tumor growth, ang...

Cancers, Vol. 13, Pages 6160: Microglial Cytokines Induce Invasiveness and Proliferation of Human Glioblastoma through Pyk2 and FAK Activation
In this study, 20 human glioblastoma specimens were evaluated with the use of RT-PCR and western blotting. A Pierson correlation test demonstrated a correlation (0.6–1.0) between the gene expression levels for platelet-derived growth factor β(PDGFβ), stromal-derived factor 1α (SDF-1α), IL-6, IL-8, and epidermal growth factor (EGF) in tumor-purified microglia and levels of p-Pyk2 (Y579/Y580) and p-FAK(Y925) in glioma cells. siRNA knockdown against Pyk2 or FAK in three primary glioblastoma cell lines, developed from the investigated specimens, in combination...

MALAT1 Improves Functional Recovery after Traumatic Brain Injury through Promoting Angiogenesis in Experimental Mice
Brain Res. 2021 Nov 19:147731. doi: 10.1016/j.brainres.2021.147731. Online ahead of print.ABSTRACTAs a highly evolutionary conserved lncRNA, MALAT1 was first demonstrated to associate with metastasis of lung tumor by promoting angiogenesis. Activated vasculature was recently indicated to assist neurogenesis by secreting neurotrophic factor Ang1 (Angiopoietin-1). The purpose of this study is to investigate the potential role of MALAT1 in angiogenesis following traumatic brain injury (TBI). Adult male mice were subjected to controlled cortical impact (CCI) and brain microvacular endothelial cells were exposed to oxygen-gluco...
