Dermorphin [D-Arg2, Lys4] (1-4) Amide Alleviates Frostbite-Induced Pain by Regulating TRP Channel-Mediated Microglial Activation and Neuroinflammation
In this study, we investigated the effect of dermorphin [D-Arg2, Lys4] (1 –4) amide (DALDA), a peripheral mu-opioid receptor agonist, on frostbite injury and hypersensitivity induced by deep freeze magnet exposure in rats. Animals with frostbite injury displayed significant hypersensitivity to mechanical, thermal, and cold stimuli which was significant ameliorated on tr eatment with different doses of DALDA (1, 3, and 10 mg/kg) and ibuprofen (100 mg/kg). Further, molecular biology investigations unveiled heightened oxido-nitrosative stress, coupled with a notable upregulation in the expression of TRP channels (TRPA1, TRP...

Synthesis and biological evaluation of new nicotinic acid derivatives as potential anti-inflammatory agents with enhanced gastric safety profile
Bioorg Chem. 2024 Mar;144:107136. doi: 10.1016/j.bioorg.2024.107136. Epub 2024 Jan 19.ABSTRACTTwo innovative series derived from nicotinic acid scaffold were synthesized and evaluated for their anti-inflammatory activity. Ibuprofen, celecoxib and indomethacin were used as standard drugs. All the newly synthesized compounds were in vitro screened for their anti-inflammatory activity adopting 3-(4,5-dimethylthiazol-2-yl)2,5-diphenyltetrazolium bromide dye (MTT), as well as Griess assays. The results showed that all compounds exhibited significant anti-inflammatory activity without affecting the viability of the macrophages c...

Organic Nucleation: Water Rearrangement Reveals the Pathway of Ibuprofen
Combining experimental and simulation approaches have been applied to investigate the nucleation of ibuprofen both in solution and at model hydrophobic interface. The macroscopic nucleation is followed, nanoscale species are identified, and the interfacial interaction between ibuprofen and water during nucleation is tackled. The multi-scale analysis implies that the structure of water changes substantially and reports the non classical pathway of ibuprofen nucleation. AbstractThe organic nucleation of the pharmaceutical ibuprofen is investigated, as triggered by the protonation of ibuprofen sodium salt at elevated pH. The ...

Genital Ulcers Are Not Always Because of Sexually Transmitted Infections: A Case Report of an Unusual Presentation of Fixed Drug Eruption in a Pediatric Patient
We report a case of fixed drug eruption to Ibuprofen that presented with balanitis and genital ulcers. Fixed drug eruption (FDE) is characterized by sharply demarcated pruritic, erythematous plaques and the diagnostic hallmark is its recurrence at previously affected sites (Maliyar et al., 2019). Management of symptomatic irreversible pulpitis using irrigation agitation techniques in anterior teeth
Bioinformation. 2023 Dec 31;19(12):1129-1133. doi: 10.6026/973206300191129. eCollection 2023.ABSTRACTEndodontic pain, a common complication after root canal treatment, affects 2.5% to 60% of patients. Therefore, it is of interest to compare apical negative pressure irrigation (EndoVac) with conventional needle irrigation to assess their impact on postoperative pain in permanent anterior teeth with symptomatic irreversible pulpitis. Fifty patients were randomly assigned to either the EndoVac or needle irrigation group. Pre and post-operative pain levels were assessed using a Visual Analog Scale and the amount of Ibuprofen t...

Preemptive use of ibuprofen and desensitizer decreases immediate tooth sensitivity after in-office bleaching: A triple-blind, randomized, placebo-controlled clinical trial
CONCLUSIONS: The combined use of 400 mg of IBU and KF2 appears to be beneficial in managing postbleaching tooth sensitivity. It provides a superior analgesic effect compared with placebo.PRACTICAL IMPLICATIONS: The preemptive administration of IBU plus KF2 reduces tooth sensitivity after tooth whitening when compared with placebo. This clinical trial was registered in the Brazilian Clinical Trials Registry Platform. The registration number is U1111-1249-8191.PMID:38244017 | DOI:10.1016/j.adaj.2023.11.011 (Source: Journal of the American Dental Association)

Facts and trends in dental antibiotic and analgesic prescriptions in Germany, 2012 –2021
ConclusionsSince 2013, the most frequently prescribed antibiotic by German dentists has been amoxicillin reaching nearly half of all dental antibiotic prescriptions in 2021. Simultaneously, the proportion of clindamycin has steadily decreased, but the level is still high compared to international data. During the past decade, ibuprofen as a first-line analgesic in German dentistry was continuously gaining in importance.Clinical relevanceAminopenicillins have the best risk –benefit balance in dentistry, but the use of antibiotics generally must be limited only to cases of severe infections or compromised patients. Pre-exi...

Molecules, Vol. 29, Pages 380: Dendrimer Coated Silica as a Sorbent for Dispersive Solid-Phase Extraction of Select Non-Steroidal Anti-Inflammatory Drugs from Water
ki Non-steroidal anti-inflammatory drugs (NSAIDs) have been recognized as a potentially serious threat to the natural environment. NSAIDs are popular painkillers, and the main pathway for them to reach natural water is via discharge from wastewater and sewage treatment plants. In order to monitor contamination caused by these drugs, as well as their impact on the environment, a new material based on Silica Gel 60, functionalized with a dendrimeric copolymer of methylamine and 1,4-butanediol diglycidyl ether (named MA-BDDE), was prepared. Initial physicochemical characterization of the MA-BDDE material was carried out u...

Molecules, Vol. 29, Pages 391: Green Synthesis of Er-Doped ZnO Nanoparticles: An Investigation on the Methylene Blue, Eosin, and Ibuprofen Removal by Photodegradation
We present a study on the green synthesis of undoped and Er-doped ZnO compounds using Mangifera indica gum (MI). A set of tests were conducted to assess the structure of the material. The tests included X-ray diffraction, Raman, and Fourier-transform infrared spectroscopy. Optical properties were studied using diffuse reflectance and photoluminescence. Morphological and textural investigations were done using SEM images and N2 adsorption/desorption. Furthermore, photocatalytic tests were performed with methylene blue (MB), yellow eosin (EY), and the pharmaceutical drug ibuprofen (IBU) under UV irradiation. The study demons...

IL-17A inhibitor-induced leucocytoclastic vasculitis is responsive to IL-23 blockade in a psoriatic arthritis patient
Introduction Secukinumab is a fully humanised monoclonal antibody directed at interleukin 17A (IL-17A) which has been widely used for the treatment of plaque psoriasis, psoriatic arthritis (PsA) and ankylosing spondylitis.1 The use of secukinumab increases the risk of mucocutaneous candidiasis, but little is known about other specific cutaneous adverse events (AEs). Of note, few cases of secukinumab-induced leucocytoclastic vasculitis (LCV) have been described in the literature.2 3 Case presentation A 59-year-old man with a history of plaque psoriasis and PsA refractory to first-line and second-line treatment presented wit...
