Interferon inhibits the release of herpes simplex virus-1 from the axons of sensory neurons
In this study, we determined the effects of type I, II, and III IFNs on HSV-1 release from sensory axons. Using compartmentalized microfluidic devices separating axons from the neuronal cell bodies, we showed that treating axons with type I (IFNα and IFNβ), type II (IFNγ), and type III (IFNλ) IFNs inhibited the release of HSV-1 from axons, without altering the anterograde axonal transport of viral components. Furthermore, we showed that only type II IFN induced a cell-wide response following axonal treatment, with phosphorylated STAT1 and STAT3 present along axons and in the neuronal cell body, whereas type I and III I...

Recurrent Erythema Multiforme Major Following COVID-19 Infection
Cureus. 2023 Jul 29;15(7):e42646. doi: 10.7759/cureus.42646. eCollection 2023 Jul.ABSTRACTErythema multiforme (EM) is a rare and potentially serious skin condition that can present as a myriad of mucocutaneous lesions. EM can be commonly confused with other cutaneous etiologies, leading to misdiagnosis and delay in proper treatment. This paper describes a case of recurrent erythema multiforme following COVID-19 infection in a 34-year-old male with no prior medical history. The patient had an extensive rash of the oral and genital mucosal areas, diffuse cutaneous involvement, and an extended length of recovery. This unexpec...

BMS-265246, a Cyclin-Dependent Kinase Inhibitor, Inhibits the Infection of Herpes Simplex Virus Type 1
In this study, we examined a panel of CDK inhibitors that target CDKs in the present study. BMS-265246 (BMS), a CDK 1/2 inhibitor, was found to effectively limit HSV-1 multiplication in Vero, HepG2, and Hela cells. A mechanism of action study suggested that BMS inhibits the early stages of viral replication when added early in the viral infection. The suppression of multiple steps in viral replication by BMS was revealed when HSV-1 infected cells were treated at different time periods in the viral life cycle. Our results suggest that BMS is a potent anti-HSV-1 agent and unique in that it may interfere with multiple steps i...

Molecular Characterization of UL50 (dUTPase) Gene of Bovine Herpes Virus 1
In this study, we analyzed the amino acid sequence of UL50 (dUTPase) using bioinformatics tools and found that it was highly conserved among herpesvirus family. Then, it was cloned and expressed in Escherichia coli Rosetta (DE3), induced by isopropy1-b-D-thiogalactopyranoside (IPTG) and the recombinant UL50 protein was purified to immunize rabbits for the preparation of polyclonal antiserum. The results indicated that the UL50 gene of BoHV-1 was composed of 978 nucleotides, which encoded 323 amino acids. Western blot analysis revealed that polyclonal sera against UL50 reacted with a band of 34 kDa. Furthermore, immunofluor...

Therapeutic Prime/Pull Vaccination of HSV-2 Infected Guinea Pigs with the Ribonucleotide Reductase 2 (RR2) Protein and CXCL11 Chemokine Boosts Antiviral Local Tissue-Resident and Effector Memory CD4 < sup > + < /sup > and CD8 < sup > + < /sup > T Cells and Protects Against Recurrent Genital Herpes
bioRxiv. 2023 Aug 9:2023.08.08.552454. doi: 10.1101/2023.08.08.552454. Preprint.ABSTRACTFollowing acute herpes simplex virus type 2 (HSV-2) infection, the virus undergoes latency in sensory neurons of the dorsal root ganglia (DRG). Intermittent virus reactivation from latency and shedding in the vaginal mucosa (VM) causes recurrent genital herpes. While T-cells appear to play a role in controlling virus reactivation and reducing the severity of recurrent genital herpes, the mechanisms for recruiting these T-cells into DRG and VM tissues remain to be fully elucidated. The present study investigates the effect of CXCL9, CXCL...

Public Awareness About Sexually Transmitted Diseases in Taif, Saudi Arabia
CONCLUSION: This study shed light on the limited knowledge and misconceptions surrounding STDs in Taif city. The findings highlighted knowledge gaps, including confusion between different STDs and misconceptions about transmission modes. The results revealed a positive correlation between higher maternal education and increased knowledge scores. These findings underscore the urgency for health authorities to develop awareness campaigns and educational initiatives to promote accurate information and foster healthier attitudes toward sexual activity in these regions.PMID:37609085 | PMC:PMC10441818 | DOI:10.7759/cureus.42302 (Source: Herpes)
