Hedgehog pathway in sarcoma: from preclinical mechanism to clinical application
AbstractSarcomas are a diverse group of malignant neoplasms of mesenchymal origin. They develop rarely, but due to poor prognosis, they are a challenging and significant clinical problem. Currently, available therapeutic options have very limited activity. A better understating of sarcomas ’ pathogenesis may help develop more effective therapies in the future. The Sonic hedgehog (Shh) signaling pathway is involved in both embryonic development and mature tissue repair and carcinogenesis. Shh pathway inhibitors are presently used in the treatment of basal cell carcinoma. Its increase d activity has been demonstrated in ma...

Invasive urothelial carcinoma with chordoid features or abundant myxoid stroma: A reappraisal of morphologic spectrum and risk stratification based on molecular classification
Pathol Res Pract. 2023 Oct 2;251:154839. doi: 10.1016/j.prp.2023.154839. Online ahead of print.ABSTRACTUrothelial carcinoma (UC) with myxoid stroma or chordoid features is a rare diagnosis. We retrospectively collected data from 17 cases of diagnosed UC with myxoid stroma, mucin production, or chordoid features. We aimed to investigate the molecular subtypes of this neoplasm and to assess subtype correlations with clinical outcomes. Immunohistochemical (IHC) staining with a panel composed of markers for basal subtypes (CK5/6, CK14, and CD44) and luminal subtypes (GATA3, FOXA1, and CK20) was performed. Morphologically, all ...

Differentiation between Chondrosarcoma and Synovial Chondromatosis of the Temporomandibular Joint Using CT and MR Imaging [HEAD & NECK]
CONCLUSIONS: CT and MR imaging features can distinguish chondrosarcoma from synovial chondromatosis of the temporomandibular joint with improved diagnostic performance when a subcombination of 9 imaging features is used. (Source: American Journal of Neuroradiology)

High dose, dual-release polymeric films for extended surgical bed paclitaxel delivery
We describe an implantable, biodegradable poly(1,2-glycerol carbonate) and poly(caprolactone) film with entrapped and covalently-bound paclitaxel enabling safe, controlled, and extended local delivery of paclitaxel achieving concentrations 10,000× tissue levels compared to systemic administration. Films containing entrapped and covalently-bound paclitaxel implanted in the tumor bed, immediately after resection of human cell line-derived chondrosarcoma and patient-derived xenograft liposarcoma and leiomyosarcoma in mice, improve median 90- or 200-day recurrence-free and overall survival compared to control mice. Thrombus or tumor?
A contrast defect was observed in inferior vena cava (IVC) of a 61-year-old male patient with de-differentiated chondrosarcoma, but it was not possible to distinguish whether it was a thrombus or a tumor. Using an angioscope, he was able to clearly diagnose a tumor. Key clinical messageContrast defects in veins are often diagnosed as benign thrombi, but depending on the patient ’s background it is necessary to differentiate between tumor thrombi. It is difficult to differentiate between these using contrast-enhanced CT alone, but with angioscopy it is easy to visually distinguish between a benign and tumor thrombi.Abstra...

Clinico-genomic profiling of conventional and dedifferentiated chondrosarcomas reveals TP53 mutation to be associated with worse outcomes
CONCLUSION: IDH mutations are prevalent in chondrosarcoma though were not associated with survival outcomes in this cohort. TP53 mutations were the next most common alteration and were associated with worse outcomes.PMID:37747813 | DOI:10.1158/1078-0432.CCR-23-1703 (Source: Clinical Cancer Research)
