Clinico-genomic profiling of conventional and dedifferentiated chondrosarcomas reveals TP53 mutation to be associated with worse outcomes
CONCLUSION: IDH mutations are prevalent in chondrosarcoma though were not associated with survival outcomes in this cohort. TP53 mutations were the next most common alteration and were associated with worse outcomes.PMID:37747813 | DOI:10.1158/1078-0432.CCR-23-1703 (Source: Clinical Cancer Research)

Cancers, Vol. 15, Pages 4696: The Enigma of Atypical Cartilaginous Tumors: Surgery or Surveillance?
lle During the last 20 years, the treatment of atypical cartilaginous tumors (ACTs) of the long bones has undergone a dramatic change: while these formerly called chondrosarcomas G1 previously led to wide resections and big reconstructions with megaprostheses, today, the use curettage of the lesions and filling the defect e [...] (Source: Cancers)

Ollier Disease: A Case Report and Review of Treatment Options
In this report, we describe the case of an 11-year-old female who presented to her physician with pain and swelling of her right ring and small fingers after playing in a swimming pool with no obvious mechanism of trauma. A routine, plain radiographic evaluation of her hand revealed the presence of multiple enchondromatosis. We hope to use this case to highlight the surgical management options for young patients with Ollier disease and discuss circumstances in which surgical management may not be indicated.PMID:37731444 | PMC:PMC10509006 | DOI:10.7759/cureus.43815 (Source: Pain Physician)

Real ‐world experience of tyrosine kinase inhibitors in children, adolescents and adults with relapsed or refractory bone tumours: A Canadian Sarcoma Research and Clinical Collaboration (CanSaRCC) study
ConclusionOur real-world data show that TKIs have meaningful activity in recurrent BT with acceptable toxicities when started at modified dosing. Inclusion of TKIs in earlier lines of treatment and/or maintenance therapy could be questions for future research. (Source: Cancer Medicine)

MiR-4270 acts as a tumor suppressor by directly targeting Bcl-xL in human osteosarcoma cells
Front Oncol. 2023 Aug 31;13:1220459. doi: 10.3389/fonc.2023.1220459. eCollection 2023.ABSTRACTChondrosarcomas and osteosarcomas are malignant bone tumors with a poor prognosis when unresectable or metastasized. Moreover, radiotherapy and chemotherapy could be ineffective. MiRNAs represent an alternative therapeutic approach. Based on high-throughput functional screening, we identified four miRNAs with a potential antiproliferative effect on SW1353 chondrosarcoma cells. Individual functional validations were then performed in SW1353 cells, as well as in three osteosarcoma cell lines. The antiproliferative and cytotoxic effe...

NADPH oxidase 4 facilitates progression of chondrosarcoma via generation of reactive oxygen species
Acta Biochim Pol. 2023 Sep 18. doi: 10.18388/abp.2020_6580. Online ahead of print.ABSTRACTNicotinamide adenine dinucleotide phosphate oxidase 4 (NOX4) is an enzyme that regulates reactive oxygen species (ROS) generation, and its function in the development of chondrosarcoma remains unclear. In the present study, we studied NOX4 expression in chondrosarcoma by immunochemical examination, and analyzed the role of NOX4 in viability and apoptosis of human chondrosarcoma cell line SW1353 using NOX4 siRNA or NOX4 inhibitor GKT137831. NOX4 level significantly increased in tumor compared to that in para-carcinoma sample. The level...

Cancers, Vol. 15, Pages 4581: Mesenchymal Chondrosarcoma from Diagnosis to Clinical Trials
Anna M. Czarnecka Mesenchymal chondrosarcoma (MCS) is a rare subtype of chondrosarcoma with a poor prognosis. Although these tumors are sensitive to radiotherapy/chemotherapy, the standard treatment for localized MCS is only surgical resection, and there are no established treatment guidelines for patients with advanced and metastatic MCS. Due to the low incidence of MCS, the pathology of these tumors is still unknown, and other therapeutic options are lacking. Some studies show the potential role of the PDGF/PPI3K/AKT, PKC/RAF/MEK/ERK, and pRB pathways, and BCL2 overexpression in the pathogenesis of MCS. These finding...

Impact of Prior Cancer on the Prognosis of Patients with Chondrosarcoma: A Population-Based Study of the SEER Database
CONCLUSION: This study initially discovered that chondrosarcoma patients with past cancer have a bad prognosis. Different types of past cancer have varying effects on survival. We urgently propose that cancer trial exclusion criteria be set specifically by cancer classification, rather than accepting the unchangeable criterion for default.PMID:37678875 (Source: Alternative Therapies in Health and Medicine)
