Human-Derived collagen hydrogel as an antibiotic vehicle for topical treatment of bacterial biofilms
This study aims to assess the capacity of human -derived collagen hydrogel to provide sustained antibiotic release to disrupt bacterial biofilms and decrease bacterial load while maintaining host cell viability and scaffold integrity. Human collagen harvested from flexor tendons underwent processing to yield a gellable liquid, and subsequently wa s combined with varying concentrations of gentamicin (50–500 mg/L) or clindamycin (10–100 mg/L). The elution kinetics of antibiotics from the hydrogel were analyzed using liquid chromatography-mass spectrometry. The gel was used to topically treat Methicillin-resistantStaphylo...

Collaborative Cross mice have diverse phenotypic responses to infection with Methicillin-resistant < i > Staphylococcus aureus < /i > USA300
by Aravindh Nagarajan, Kristin Scoggin, Jyotsana Gupta, Manuchehr Aminian, L. Garry Adams, Michael Kirby, David Threadgill, Helene Andrews-PolymenisStaphylococcus aureus (S.aureus) is an opportunistic pathogen causing diseases ranging from mild skin infections to life threatening conditions, including endocarditis, pneumonia, and sepsis. To identify host genes modulating this host-pathogen interaction, we infected 25 Collaborative Cross (CC) mouse strains with methicillin-resistantS.aureus (MRSA) and monitored disease progression for seven days using a surgically implanted telemetry system. CC strains varied widely in thei...

Raw meat-based diet for pets: a neglected source of human exposure to Salmonella and pathogenic Escherichia coli clones carrying mcr, Portugal, September 2019 to January 2020
ConclusionSome RMBDs from European brands available in Portugal can be a vehicle for clinically relevant MDRSalmonella and pathogenicE. coli clones carrying genes encoding resistance to the last-line antibiotic colistin. Proactive actions within the One Health context, spanning regulatory, pet-food industry and consumer levels, are needed to mitigate these public health risks. (Source: Eurosurveillance)

The Spx stress regulator confers high-level β-lactam resistance and decreases susceptibility to last-line antibiotics in methicillin-resistant < em > Staphylococcus aureus < /em >
Antimicrob Agents Chemother. 2024 May 1:e0033524. doi: 10.1128/aac.00335-24. Online ahead of print.ABSTRACTInfections caused by methicillin-resistant Staphylococcus aureus (MRSA) are a leading cause of mortality worldwide. MRSA has acquired resistance to next-generation β-lactam antibiotics through the horizontal acquisition of the mecA resistance gene. Development of high resistance is, however, often associated with additional mutations in a set of chromosomal core genes, known as potentiators, which, through poorly described mechanisms, enhance resistance. The yjbH gene was recently identified as a hot spot for adaptiv...

Photosensitive AIEgens sensitize bacteria to oxidative damage and modulate the inflammatory responses of macrophages to salvage the photodynamic therapy against MRSA
Biomaterials. 2024 Apr 26;309:122583. doi: 10.1016/j.biomaterials.2024.122583. Online ahead of print.ABSTRACTThe urgent need for antimicrobial agents to combat infections caused by multidrug-resistant bacteria facilitates the exploration of alternative strategies such as photosensitizer (PS)-mediated photoinactivation. However, increasing studies have discovered uncorrelated bactericidal activities among PSs possessing similar photodynamic and pathogen-targeted properties. To optimize the photodynamic therapy (PDT) against infections, we investigated three type-I PSs of D-π-A AIEgens TI, TBI, and TTI. The capacities of re...

Mariculture waters as yet another hotbed for the creation and transfer of new antibiotic-resistant pathogenome
Environ Int. 2024 Apr 26;187:108704. doi: 10.1016/j.envint.2024.108704. Online ahead of print.ABSTRACTWith the rapid growth of aquaculture globally, large amounts of antibiotics have been used to treat aquatic disease, which may accelerate induction and spread of antibiotic-resistant bacteria (ARB) and antibiotic resistance genes (ARGs) in aquaculture environments. Herein, metagenomic and 16S rRNA analyses were used to analyze the potentials and co-occurrence patterns of pathogenome (culturable and unculturable pathogens), antibiotic resistome (ARGs), and mobilome (mobile genetic elements (MGEs)) from mariculture waters ne...

Novel natural osthole-inspired amphiphiles as membrane targeting antibacterials against methicillin-resistant Staphylococcus aureus (MRSA)
Eur J Med Chem. 2024 Apr 26;271:116449. doi: 10.1016/j.ejmech.2024.116449. Online ahead of print.ABSTRACTMethicillin-resistant Staphylococcus aureus (MRSA) is a widespread pathogen causing clinical infections and is multi-resistant to many antibiotics, making it urgent need to develop novel antibacterials to combat MRSA. Herein, we designed and prepared a series of novel osthole amphiphiles 6a-6ad by mimicking the structures and function of antimicrobial peptides (AMPs). Antibacterial assays showed that osthole amphiphile 6aa strongly inhibited S. aureus and 10 clinical MRSA isolates with MIC values of 1-2 μg/mL, comparab...

Molecules, Vol. 29, Pages 2084: UVC Stokes and Anti-Stokes Emission of Ca9Y(PO4)7 Polycrystals Doped with Pr3+ Ions
k Dereń The recent COVID-19 pandemic has made everyone aware of the threat of viruses and the growing number of antibiotic-resistant bacteria. It has become necessary to find new methods to combat these hazards. One tool that could be used is UVC radiation, i.e., 100–280 nm. Currently, the available sources of this light are mercury vapor lamps. However, the modern world requires more compact, mercury-free, and less energy-consuming light sources. This work presents the results of our research on a new material in which efficient UVC radiation was obtained. Here, we present the results of research on Ca9...

On-site detection of methicillin-resistant Staphylococcus aureus (MRSA) utilizing G-quadruplex based isothermal exponential amplification reaction (GQ-EXPAR)
Talanta. 2024 Apr 13;275:126073. doi: 10.1016/j.talanta.2024.126073. Online ahead of print.ABSTRACTMethicillin-resistant Staphylococcus aureus (MRSA) has a high incidence in infectious hospitals and communities, highlighting the need for early on-site detection due to its resistance to methicillin antibiotics. The present study introduces a highly sensitive detection system for mecA, a crucial methicillin marker, utilizing an RCA-based isothermal exponential amplification reaction. The G-quadruplex-based isothermal exponential amplification reaction (GQ-EXPAR) method designs probes to establish G-quadruplex secondary struc...
