Oestrogen receptor-independent actions of oestrogen in cancer
Mol Biol Rep. 2023 Sep 20. doi: 10.1007/s11033-023-08793-8. Online ahead of print.ABSTRACTOestrogen, the primary female sex hormone, plays a significant role in tumourigenesis. The major pathway for oestrogen is via binding to its receptor [oestrogen receptor (ERα or β)], followed by nuclear translocation and transcriptional regulation of target genes. Almost 70% of breast tumours are ER + , and endocrine therapies with selective ER modulators (tamoxifen) have been successfully applied. As many as 25% of tamoxifen-treated patients experience disease relapse within 5 years upon completion of chemotherapy. In such cases, t...

Non-retroareolar male mucinous breast cancer without gynecomastia development in an elderly man: A case report
CONCLUSION: To the best of our knowledge, this is the first case report of MMBC in the non-retroareolar region of the nipple without gynecomastia development. Mucinous tumors are easily missed during diagnosis, and the incidence of axillary lymph node metastases of chest mucinous tumors has increased.PMID:37727495 | PMC:PMC10506025 | DOI:10.12998/wjcc.v11.i25.5954

Enantioselective De Novo Synthesis of α,α-Diaryl Ketones from Alkynes
Angew Chem Int Ed Engl. 2023 Sep 19:e202310078. doi: 10.1002/anie.202310078. Online ahead of print.ABSTRACTChiral α,α-diaryl ketones are structural motifs commonly present in bioactive molecules, and they are also valuable building blocks in synthetic organic chemistry. However, catalytic asymmetric synthesis of α,α-diaryl ketones bearing a tertiary stereogenic center remains largely unsolved. Herein, we report a catalytic de novo enantioselective synthesis of α,α-diaryl ketones from simple alkynes via chiral phosphoric acid (CPA) catalysis. A broad range of enolizable α,α-diaryl ketones are prepared in good yields...

Thyroid hormone enhances estrogen-mediated proliferation and cell cycle regulatory pathways in steroid receptor-positive breast Cancer
In this report, we show TH ± E2 ± tamoxifen inhibits cell cycle control signaling, reduces apoptosis, and enhances cell proliferation, tumor growth, tamoxifen resistance, and clonal expansion. Mechanistically these changes involve numerous genes and pathways, including critical cell cycle regulatory proteins and genes identified using various molecular methods. These studies facilitate a greater mechanistic understanding of the biological and molecular impact of TH on SR+ BC.PMID:37723865 | DOI:10.1080/15384101.2023.2249702

Molecular docking analysis of the tumor protein beta arrestin-1 with oxadiazole compounds
Bioinformation. 2023 Jan 31;19(1):111-116. doi: 10.6026/97320630019111. eCollection 2023.ABSTRACTBeta arrestins are a family of adaptor proteins that help in the regulation of signaling and trafficking of various G protein coupled receptors (GPCRs). Six oxadiazole derivatives taken from literature are analyzed for anti-cancer properties. The toxicity profiles of all the drugs were similar to Tamoxifen used as control. Data shows that compounds 2, 4, and 6 exhibited comparably significant molecular interactions with the cancerous protein for further consideration.PMID:37720289 | PMC:PMC10504516 | DOI:10.6026/97320630019111

Confounding Effects of Tamoxifen: Cautionary and Practical Considerations for the Use of Tamoxifen-Inducible Mouse Models in Atherosclerosis Research
CONCLUSIONS: In this study, we establish that tamoxifen administration results in decreased serum cholesterol, decreased plaque formation, and increased hepatic lipid accumulation. These alterations represent significant confounding variables in atherosclerosis research, and we urge future investigators to take these findings into consideration when planning and executing their own atherosclerosis experiments.PMID:37706321 | DOI:10.1161/ATVBAHA.123.319922
