High TRB3 expression induces chondrocyte autophagy and senescence in osteoarthritis cartilage
CONCLUSIONS: Interfering with TRB3 expression in cartilage may serve as a target in the prevention and treatment of age-related osteoarthritis.PMID:35776529 | DOI:10.18632/aging.204066

BNIP3 mediates the different adaptive responses of fibroblast-like synovial cells to hypoxia in patients with osteoarthritis and rheumatoid arthritis
CONCLUSION: In summary, RA-FLSs maintained intracellular redox balance through mitophagy to promote cell survival under hypoxia. The mitophagy of OA-FLSs was too little to maintain the redox balance of mitochondria, resulting in apoptosis. The difference of mitophagy between OA-FLSs and RA-FLSs under hypoxia is mediated by the level of BNIP3 expression.PMID:35690741 | DOI:10.1186/s10020-022-00490-9

RPL38 knockdown inhibits the inflammation and apoptosis in chondrocytes through regulating METTL3-mediated SOCS2 m6A modification in osteoarthritis
CONCLUSION: RPL38 knockdown inhibited osteoarthritic chondrocyte dysfunction and alleviated OA progression through promoting METTL3-m6A-mediated SOCS2 expression.PMID:35596790 | DOI:10.1007/s00011-022-01579-x

Valdecoxib attenuates lipid-induced hepatic steatosis through autophagy-mediated suppression of endoplasmic reticulum stress
This study investigated the effects of VAL on lipid accumulation and lipogenesis in human primary hepatocytes. Treatment with VAL suppressed lipid accumulation and expressions of lipogenic genes, such as processed sterol regulatory element binding proteins (SREBP1) and stearoyl-CoA desaturase-1 (SCD1) in palmitate-treated hepatocytes. Furthermore, VAL ameliorated dose-dependently palmitate-induced ER stress markers. Treatment of hepatocytes with VAL increased AMPK phosphorylation and SIRT6 expression. siRNA-mediated suppression of AMPK or SIRT6 abolished the effects of VAL on lipid accumulation, lipogenesis, and endoplasmi...

Advanced Glycation End Products Induced Mitochondrial Dysfunction of Chondrocytes through Repression of AMPK α-SIRT1-PGC-1α Pathway
Conclusion: Taken together, AGEs reduced the AMPK α/SIRT1/PGC-1α signaling in chondrocytes, leading to mitochondrial dysfunction as a result of increased oxidative stress, inflammation, and apoptosis. These results indicated that target AMPK may be as a novel therapeutic strategy for AGEs-related OA prevention.Pharmacology

Pin1-mediated regulation of articular cartilage stem/progenitor cell aging
In this study, we investigated the role of Pin1 in the aging of rat knee joint CSPCs. We isolated CSPCs from rat knee joints and demonstrated that, in long-term in vitro culture, Pin1 protein levels are significantly reduced. At the same time, expression of the senescence-related β-galactosidase and the senescence marker p16INK4A were markedly elevated. In addition, Pin1 overexpression reversed the progression of cellular senescence, as evidenced by the down-regulation of senescence-related β-galactosidase, increased EdU positive cells and diminished levels of p16INK4A. In contrast, Pin1 siRNA incorporation promoted CSPC...

Epigenetic Regulation of Nutrient Transporters in Rheumatoid Arthritis Fibroblast ‐Like Synoviocytes
ConclusionThis study demonstrates that changes in the epigenetic landscape of genes related to nutrient transporters and metabolic pathways identify RA-specific targets, including critical SLC transporters, enzymes, and transcription factors, to develop novel therapeutic agents. This article is protected by copyright.
