A recalcitrant skin lesion and subsequent infection in a recreational intramural male athlete: a case report
J Athl Train. 2023 Nov 28. doi: 10.4085/1062-6050-0467.22. Online ahead of print.ABSTRACTA 35-year-old intramural male athlete presented to the athletic training staff with a 4.5cm x 2.2cm itchy, painful, swollen, and infected insidious skin lesion on his right lateral malleolus due to an underlying dermatological deficiency. Suspecting infection, the patient was referred to his nurse practitioner and was diagnosed with atopic dermatitis caused by a ceramide deficiency. He was placed on Cefalexin and Mupirocin 2% ointment but returned due to the lesion increasing to 8.5cm x 6cm although infection seemed controlled. He was ...

Chitosan and α-cellulose-based mupirocin topical film-forming spray: Optimization, in vitro characterization, antimicrobial studies and wound healing activity
Int J Biol Macromol. 2023 Oct 25:127622. doi: 10.1016/j.ijbiomac.2023.127622. Online ahead of print.ABSTRACTThe study aimed to develop a biopolymer-based mupirocin film-forming spray (MUP-FFS) for wound healing using chitosan and α-cellulose. MUP-FFS formulation was optimized by box-Behnken design, wherein the amount of chitosan, glycerol, and microfluidizer cycles showed a significant effect on the drying time and sprayability, but drug release remained unaffected. The optimized MUP-FFS formulation prepared by 13 microfluidizer cycles containing chitosan (0.125 %), glycerol (2.76 %) was quickly sprayable with 235 s dryin...

Bayesian modeling of the impact of antibiotic resistance on the efficiency of MRSA decolonization
by Fanni Ojala, Mohamad R. Abdul Sater, Loren G. Miller, James A. McKinnell, Mary K. Hayden, Susan S. Huang, Yonatan H. Grad, Pekka Marttinen Methicillin-resistantStaphylococcus aureus (MRSA) is a major cause of morbidity and mortality. Colonization by MRSA increases the risk of infection and transmission, underscoring the importance of decolonization efforts. However, success of these decolonization protocols varies, raising the possibility that some MRSA strains may be more persistent than others. Here, we studied how the persistence of MRSA colonization correlates with genomic presence of antibiotic resistance genes. O...

Purification and characterization of thioredoxin reductase enzyme from commercial Spirulina platensis tablets by affinity chromatography and investigation of the effects of some chemicals and drugs on enzyme activity
AbstractThioredoxin reductase (TrxR, enzyme code [E.C.] 1.6.4.5) is a widely distributed flavoenzyme that catalyzes nicotinamide adenine dinucleotide phosphate (NADPH)-dependent reduction of thioredoxin and many other physiologically important substrates.Spirulina platensis is a blue –green algae that is often used as a dietary supplement.S. platensis is rich in protein, lipid, polysaccharide, pigment, carotenoid, enzyme, vitamins and many other chemicals and exhibits a variety of pharmacological functions. In the present study, a simple and efficient method to purify TrxR fromS. platensis tablets is reported. The extrac...

Chronic Crusted Plaque After Skin Injury
An 11-year-old male presented to the emergency department with a lesion on his right proximal lower leg. Two months earlier, he had fallen onto a branch, leading to a laceration requiring repair with sutures. This wound was improving initially; however, six weeks later, surrounding redness and overlying crusting developed. The child presented to a walk-in clinic and was started on topical mupirocin ointment and oral cephalexin for presumed cellulitis. The surrounding erythema worsened despite treatment with antibiotics, and he presented to the emergency department. (Source: The Journal of Pediatrics)

Lupus Erythematosus Profundus with Multiple Overlying Cutaneous Ulcerations: A Rare Case
We present a case report of LEP with multiple cutaneous ulcers on the right cheek and scalp accompanied by cicatricial alopecia. No other systemic manifestations were noted. Histopathological examination revealed periadipocyte, perivascular, and perivascular infiltration of lymphocytes, eosinophils, and plasma cells, supporting the diagnosis of LEP. The topical treatments given to the patient were sunscreen, 2% mupirocin cream, and wound dressing with dialkyl carbamoyl chloride (DACC). The patient was also treated systemically with oral corticosteroids and hydroxychloroquine. Clinical improvements were observed in the 3rd ...

Antiseptics and mupirocin resistance in clinical, environmental, and colonizing coagulase negative Staphylococcus isolates
ConclusionsCoNS isolates colonizing healthcare workers showed a high prevalence of antiseptic resistance genes, while clinical infection samples were more resistant to antibiotics. CHDG demonstrated greater efficacy than BC and CTAB in our hospital. (Source: Antimicrobial Resistance and Infection Control)

Structure and Function of the  α-Hydroxylation Bimodule of the Mupirocin Polyketide Synthase
Angew Chem Int Ed Engl. 2023 Sep 28:e202312514. doi: 10.1002/anie.202312514. Online ahead of print.ABSTRACTMupirocin is a clinically important antibiotic produced by a trans-AT Type I polyketide synthase (PKS) in Pseudomonas fluorescens. The major bioactive metabolite, pseudomonic acid A (PA-A), is assembled on a tetrasubstituted tetrahydropyran (THP) core incorporating a 6-hydroxyl group proposed to be introduced via α-hydroxylation of the thioester of the acyl carrier protein (ACP) bound polyketide chain. Herein, we describe an in vitro approach combining purified enzyme components, chemical synthesis, isotopic labellin...

Genetic diversity of Staphylococcus aureus isolated from ear infections in Iran: Emergence of CC8/ST239-SCCmec III as major genotype
This study highlighted genetic variability and strong biofilm formation ability among our isolates revealing its crucial role in enhancing the resistance of this bacteria to drugs. Thus, it is necessary to continue the epidemiological analysis to improve the control of ear infections related to S. aureus.PMID:37721867 | DOI:10.1556/030.2023.02081 (Source: Acta Microbiologica et Immunologica Hungarica)
