Perinatal exposure to the fungicide ketoconazole alters hypothalamic control of puberty in female rats
ConclusionnRH secretion and the hypothalamic transcriptome are highly sensitive to perinatal exposure to both DES and KTZ. The identified pathways should be exploredfurther to identify biomarkers for future testing strategies for EDC identification and when enhancing the current standard information requirements in regulation. DNA damage and up-regulation of PARP-1 induced by columbin in vitro and in vivo
Toxicol Lett. 2023 Mar 9:S0378-4274(23)00106-6. doi: 10.1016/j.toxlet.2023.03.003. Online ahead of print.ABSTRACTColumbin (CLB) is the most abundant (>1.0%) furan-containing diterpenoid lactone in herbal medicine Tinospora sagittate (Oliv.) Gagnep. The furano-terpenoid was found to be hepatotoxic, but the exact mechanisms remain unknown. The present study demonstrated that administration of CLB at 50mg/kg induced hepatotoxicity, DNA damage and up-regulation of PARP-1 in vivo. Exposure to CLB (10μM) induced GSH depletion, over-production of ROS, DNA damage, up-regulation of PARP-1 and cell death in cultured mouse primar...

GSE208545 Transcriptional profiling of developing rat ovary following intrauterine exposure to known endocrine disruptors
Contributors : Indusha Kugathas ; Frederic Chalmel ; Severine Mazaud-Guittot ; Terje SvingenSeries Type : Expression profiling by high throughput sequencingOrganism : Rattus norvegicusThe main objective of the FREIA project is to identify markers of endocrine disrupting effects in rodents to overcome the lack of sensitive markers of female reproductive toxicity. For that 2 archetypes of endocrine disruption are studied, ketoconazole (KTZ) and diethylstilbestrol (DES) at three different stages (post-natal days 6, 14, and 22) and at three different doses 3, 6 and 12 milligram per kilogram by bodyweight per day for KTZ an...

Rapid equilibrium dialysis, ultrafiltration or ultracentrifugation? Evaluation of methods to quantify the unbound fraction of substances in plasma
Biochem Biophys Res Commun. 2023 Feb 10;651:114-120. doi: 10.1016/j.bbrc.2023.02.021. Online ahead of print.ABSTRACTIn pharmacokinetics plasma protein binding (PPB) is a well-established parameter impacting drug disposition. The unbound fraction (fu) is arguably regarded the effective concentration at the target site. Pharmacology and toxicology, increasingly use in vitro models. The translation of in vitro concentrations to in vivo doses can be supported by toxicokinetic modelling, e.g. physiologically based toxicokinetic models (PBTK). PPB of a test substance is an input parameter for PBTK. We compared three methods to q...

Differential adoption of castration ‐resistant prostate cancer treatment across facilities in a national healthcare system
ConclusionMany facilities persisted in their use of ketoconazole as first-line CRPC therapy, even when other facilities had adopted the new standard of care abiraterone and enzalutamide. Further work is needed to identify the effect of this late adoption on outcomes important to patients. (Source: Cancer Medicine)
