The Effect of Thiazolidinediones in Polycystic Ovary Syndrome: A Systematic Review and Meta-Analysis of Randomised Controlled Trials
CONCLUSION: Both pioglitazone and rosiglitazone were associated with significant increases in body weight and BMI when compared with metformin or placebo. Pioglitazone significantly reduced triglycerides and fasting insulin when compared with placebo while rosiglitazone showed a modest reduction of LH when compared with metformin.PROSPERO REGISTRATION NO: CRD42020178783.PMID:38683294 | DOI:10.1007/s12325-024-02848-3 (Source: Adv Data)

Metformin inhibits nerve growth factor (NGF)-induced sympathetic neuron differentiation through p35/CDK5 inhibition
In this study, we investigated the impact of metformin on NGF-induced sympathetic neuronal differentiation using rat pheochromocytoma PC12 cells as a model. We examined the regulation of TrkA-p35/CDK5 signaling in NGF-induced PC12 differentiation. Our results demonstrate that metformin reduces NGF-induced PC12 differentiation by inactivating the TrkA receptor, subsequently inhibiting ERK and EGR1. Inhibition of this cascade ultimately leads to the down-regulation of p35/CDK5 in PC12 cells. Furthermore, metformin inhibits the activation of the pre-synaptic protein Synapsin-I, a substrate of CDK5, in PC12 differentiation. Ad...

A review on the nexus of autophagy genes from the perspective of polycystic ovary syndrome
Biol Cell. 2024 Apr 28:e202300069. doi: 10.1111/boc.202300069. Online ahead of print.ABSTRACTPolycystic ovary syndrome or PCOS is an endocrine disorder in women of reproductive age. It is a diversified multi factorial disorder and diagnosis is very complicated because of its overlapping symptoms some of which are irregular menstrual cycle, acne in face, excess level of androgen (AE), insulin resistance, obesity, cardiovascular disease, mood disorder and type 2 diabetes (T2DM). PCOS may be caused by hormonal imbalance, genetic and epigenetic vulnerability, hypothalamic and ovarian troubles. PCOS is essentially hyperandrogen...

Prescriptions for Antipsychotics: Youth with Intellectual/Developmental Disabilities Compared to Youth without Intellectual/Developmental Disabilities
CONCLUSION: Compared to youth without IDD, youth with IDD were more frequently dispensed antipsychotics and metformin. These findings have important implications for the health of youth with IDD.PMID:38678514 | DOI:10.1007/s10803-024-06344-z (Source: Journal of Autism and Developmental Disorders)

Low-dose metformin suppresses hepatocellular carcinoma metastasis via the AMPK/JNK/IL-8 pathway
CONCLUSION: The results indicate that low-dose metformin can inhibit HCC metastasis by suppressing IL-8 expression. Targeting the AMPK/JNK/IL-8 axis may be a promising treatment strategy for patients with HCC metastasis.PMID:38679570 | DOI:10.1177/03946320241249445 (Source: International Journal of Immunopathology and Pharmacology)

FAM83B regulates mitochondrial metabolism and anti-apoptotic activity in pulmonary adenocarcinoma
This study aims to understand how increased FAM83B expression impacts mitochondrial activity, cell apoptosis, and chemotherapy effectiveness in LUAD. Multiple assays, such as CCK8, wound healing, EdU, and transwell assays, were employed to confirm the augmented chemotherapy resistance, heightened cell proliferation, migration, and invasion caused by FAM83B overexpression in LUAD cells. Furthermore, MIMP, MTG, and ATP assays were utilized to quantify changes in mitochondrial metabolism. In vitro functional assays were performed to evaluate the influence of FAM83B overexpression on the malignant progression and resistance me...

Effects of Metformin Therapy on Thyroid Volume and Functions in Patients with Newly Diagnosed Type 2 Diabetes Mellitus: A Single-center Prospective Study
CONCLUSION: Our results suggest that metformin treatment significantly reduces body weight, BMI, thyroid volume, and serum TSH, ALT, and Anti-TPO levels in patients with newly diagnosed T2DM. Moreover, serum TSH levels showed a significant decrease in non-obese subjects, while thyroid volume showed a significant decrease in obese subjects.PMID:38676519 | DOI:10.2174/0118715303307313240315162000 (Source: Endocrine, Metabolic and Immune Disorders Drug Targets)

Metformin inhibits activation of NLRP3 inflammasome and inflammatory response in preeclamptic rats
CONCLUSIONS: MET demonstrates a protective effect against L-NAME-induced PE rats, potentially mediated through inhibition of the inflammatory response, downregulation of NLRP3 inflammasome expression in the placenta, and regulation of the balance between anti-angiogenic and pro-angiogenic factors.PMID:38677349 | DOI:10.1016/j.gene.2024.148509 (Source: Gene)

Impact of Metformin on Neocortical Development during Pregnancy: Involvements of ERK and p35/CDK5 Pathways
Chemosphere. 2024 Apr 25:142124. doi: 10.1016/j.chemosphere.2024.142124. Online ahead of print.ABSTRACTMetformin, the most commonly prescribed drug for the treatment of diabetes, is increasingly used during pregnancy to address various disorders such as diabetes, obesity, preeclampsia, and metabolic diseases. However, its impact on neocortex development remains unclear. Here, we investigated the direct effects of metformin on neocortex development, focusing on ERK and p35/CDK5 regulation. Using a pregnant rat model, we found that metformin treatment during pregnancy induces small for gestational age (SGA) and reduces relat...
