Cancers, Vol. 16, Pages 702: Pulmonary Metastasectomy for Adrenocortical Carcinoma & mdash; Not If, but When
Conclusions: PM appears to have a role in ACC patients. The number of nodules should not be an exclusion factor. Patients developing LM within a year of primary tumor resection may benefit from waiting before further surgeries, which may provide additional insight into who may benefit from PM. (Source: Cancers)

Diagnosis of adrenocortical carcinoma
Orv Hetil. 2024 Jan 28;165(4):123-131. doi: 10.1556/650.2024.32958. Print 2024 Jan 28.ABSTRACTA mellékvesekéreg-carcinoma ritka, rossz prognózisú megbetegedés. Az érintett betegek kórjóslatát döntően meghatározza, hogy a betegség korán, még lokoregionális stádiumban kerüljön felismerésre, amikor a sebészi tumoreltávolítással jelentősen növelhető a gyógyulás esélye. A mellékvese-daganatok dignitásának meghatározása gyakran jelent kihívást a preoperatív kivizsgálás során, a diagnózisalkotást hagyományos radiológiai és laboratóriumi vizsgálatok segítik. Újabban igen jó diagno...

The European Network for the Study of Adrenal Tumors staging system (2015): a United States validation
CONCLUSION: The accuracy of mENSAT is virtually the same as that of AJCC in predicting CSM (74.7 vs. 74.5%) and OM (73.7 vs. 73.5%). However, calibration is lower for mENSAT than for AJCC. In consequence, no obvious benefit appears to be associated with the use of mENSAT relative to AJCC in United States ACC patients.PMID:38266758 | DOI:10.1210/clinem/dgae047 (Source: The Journal of Clinical Endocrinology and Metabolism)

Carney complex predisposes to breast cancer: prospective study of 50 women
CONCLUSIONS: As CNC could predispose to breast carcinoma, an adequate screening strategy and follow-up should be discussed in affected women.CLINICAL TRIAL REGISTRATION: ClinicalTrial.gov NCT00668291.PMID:38252880 | DOI:10.1093/ejendo/lvae010 (Source: European Journal of Endocrinology)

Recovery of adrenal function after stopping mitotane in patients with adrenocortical carcinoma
CONCLUSIONS: Our study demonstrates that adrenal recovery occurs in most patients after stopping mitotane, particularly when followed up in specialized centers, but not in all. Elimination time of mitotane after treatment discontinuation is very long but individually quite variable.PMID:38244214 | DOI:10.1093/ejendo/lvae007 (Source: European Journal of Endocrinology)

Hypogonadism and sexual function in men affected by adrenocortical carcinoma under mitotane therapy
ConclusionAMT was associated with hypergonatropic hypogonadism and ED, while ART led to a significant improvement of cFT levels and sexual function in the hypogonadal ACC patients. Therefore, we suggest to evaluate LH, SHBG, TT and cFT and sexual function during AMT, and start ART in the hypogonadal ACC patients with sexual dysfunction. (Source: Frontiers in Endocrinology)

Wnt/ β-catenin signaling pathway in the tumor progression of adrenocortical carcinoma
Adrenocortical carcinoma (ACC) is an uncommon, aggressive endocrine malignancy with a high rate of recurrence, a poor prognosis, and a propensity for metastasis. Currently, only mitotane has received certification from both the US Food and Drug Administration (FDA) and the European Medicines Agency for the therapy of advanced ACC. However, treatment in the advanced periods of the disorders is ineffective and has serious adverse consequences. Completely surgical excision is the only cure but has failed to effectively improve the survival of advanced patients. The aberrantly activated Wnt/β-catenin pathway is one of the cat...

Pan-Cancer Analysis of P3H1 and Experimental Validation in Renal Clear Cell Carcinoma
In this study, for the first time, aspects associated with P3H1, such as the mRNA expression, any mutation, promoter methylation, and prognostic significance, the relationship between P3H1 and clinicopathological parameters, drug sensitivity, and immune cell infiltration were investigated by searching several databases including The Cancer Genome Atlas (TCGA), Genotype-Tissue Expression (GTEx), cBioPortal, and The Tumor Immune Evaluation Resource (TIMER2.0) using bioinformatics tools. The findings indicate significant differential expression of P3H1 in most tumors when compared to normal tissues, with a strong association ...

A pan-cancer analysis of the prognostic implication and oncogenic role of tubulin epsilon and delta complex 2 (TEDC2) in human tumors
In this study, we employed R software and various online bioinformatics analysis tools to investigate the functional attributes of TEDC2 in human tumours and its potential involvement in immune response. The status of TEDC2 expression was evaluated in samples from the TCGA and GEO datasets, as well as in tumour and corresponding normal samples from the TCGA database. Subsequently, Kaplan-Meier estimates, clinical correlations, and univariate Cox regressions were used to analyze the 33 types of tumors from TCGA and determine the prognostic significance of TEDC2. Moreover, nomogram models were formulated using three distinct...

Current Evidence on Local Therapies in Advanced Adrenocortical Carcinoma
Horm Metab Res 2024; 56: 91-98 DOI: 10.1055/a-2209-6022International guidelines emphasise the role of local therapies (LT) for the treatment of advanced adrenocortical carcinoma (ACC). However, large studies are lacking in this field. Therefore, we performed a review of the literature to synthesise current evidence and develop clinical guidance. PubMed database was searched for systematic literature. We identified 119 potentially relevant articles, of which 21 could be included in our final analysis. All were retrospective and reported on 374 patients treated with...

A reappraisal of transcriptional regulation by NR5A1 and beta-catenin in adrenocortical carcinoma
CONCLUSION: Overall, my analysis suggests a model where NR5A1 overexpression and beta-catenin activation principally act independently, rather than functionally interacting, to drive ACC malignancy.PMID:38155952 | PMC:PMC10753177 | DOI:10.3389/fendo.2023.1303332 (Source: Cell Research)
