Potential therapeutic role of melatonin in hepatobiliary diseases: current evidence and clinical observations
J Physiol Pharmacol. 2023 Apr;74(2). doi: 10.26402/jpp.2023.2.01. Epub 2023 Jul 10.ABSTRACTMelatonin (MEL) is produced and secreted by the pineal gland as well as the small intestine, liver, retina, lymphocytes, and melanocytes in the skin in both experimental animals as well as in humans. While pineal and retinas MEL is closely related to the light/dark cycle, the production of MEL by other so called extrapineal tissues is independent of such circadian rhythm. Among the primary mechanisms of action of MEL in humans, the most important are interaction of MEL with specific receptors (M1, M2, M3) and the MEL 'scavenging' act...

Using Human 'Personalized' Cybrids to Identify Drugs/Agents That Can Regulate Chronic Lymphoblastic Leukemia Mitochondrial Dysfunction
This study uses personalized chronic lymphoblastic leukemia (CLL) cybrid cells to test various drugs/agents designed to improve mitochondrial function and cell longevity. Age-matched control (NL) and CLL cybrids were created. The NL and CLL cybrids were treated with ibrutinib (Ibr-10 μM), mitochondrial-targeted nutraceuticals such as alpha lipoic acid (ALA-1 mM), amla (Aml-300 μg), melatonin (Mel-1 mM), resveratrol (Res-100 μM) alone, or a combination of ibrutinib with nutraceuticals (Ibr + ALA, Ibr + Aml, Ibr + Mel, or Ibr + Res) for 48 h. MTT (3-(4,5-dimethylthiazolyl-2)-2,5-diphenyltetrazoliumbromide), H2DCFDA(2',7' ...

Efficacy of melatonin and ramelteon for the acute and long-term management of insomnia disorder in adults: A systematic review and meta-analysis
J Sleep Res. 2023 Jul 11:e13939. doi: 10.1111/jsr.13939. Online ahead of print.ABSTRACTMelatonin has gained growing interest as a treatment of insomnia, despite contradictory findings, and a low level of evidence. A systematic review and meta-analysis was conducted following PRISMA criteria, to assess the efficacy of melatonin and ramelteon compared with placebo on sleep quantity and quality in insomnia disorder, while also considering factors that may impact their efficacy. This review included 22 studies, with 4875 participants, including 925 patients treated with melatonin, 1804 treated with ramelteon and 2297 receiving...

Effects of 24-Epibrassinolide, melatonin and their combined effect on cadmium tolerance in Primula forbesii Franch
This study aimed to investigate the interaction between 24-Epibrassinolide (EBR) and melatonin (MT) and their effects on cadmium (Cd)-stressed Primula forbesii Franch. P. forbesii seedlings were hydroponically acclimatized at 6-7 weeks, then treated with Cd (200 μmol L-1), 24-EBR (0.1 μmol L-1), and MT (100 μmol L-1) after two weeks. Cd stress significantly reduced crown width, shoot, root length, shoot fresh weight, and fresh and dry root weights. Herein, 24-EBR, MT, and 24-EBR+MT treatments attenuated the growth inhibition caused by Cd stress and improved the morphology, growth indexes, and ornamental characteristics ...

Exogenous melatonin ameliorates steroid-induced osteonecrosis of the femoral head by modulating ferroptosis through GDF15-mediated signaling
CONCLUSIONS: We proposed that MT attenuated SONFH by inhibiting ferroptosis through the regulation of GDF15, and supplementation with exogenous MT might be a promising method for the treatment of SONFH.PMID:37400902 | DOI:10.1186/s13287-023-03371-y

Melatonin improves the quality of rotenone-exposed mouse oocytes through association with histone modifications
In this study, we investigated the impact of rotenone on mouse oocyte quality and evaluated the protective effect of melatonin on oocytes exposed to rotenone. Our results showed that rotenone impaired mouse oocyte maturation and early embryo cleavage. However, melatonin prevented these negative effects by ameliorating rotenone-induced mitochondrial dysfunction and dynamic imbalance, intracellular Ca2+ homeostasis damage, ER stress, early apoptosis, meiotic spindle formation disruption, and aneuploidy in oocytes. Additionally, RNA sequencing analysis showed that rotenone exposure changed the expression of multiple genes inv...
