Targeting long non-coding RNA PVT1/TGF- β/Smad by p53 prevents glioma progression
This study discussed the impact of p53/lncRNA plasmacytoma variant translocation 1 (lncRNA PVT1)/transforming growth factor beta (TGF-β)/Smad axis on the biological characteristics of glioma. Glioma and normal tissues were collected, in which relative lncRNA PVT1 and p53 expression was assessed. Pearson's analysis was adopted for the correlation analysis between lncRNA PVT1 and p53. Short interfering RNA (siRNA) against lncRNA PVT1 (siRNA-PVT1), siRNA-p53 or both was transfected into the glioma cells to evaluate effects of lncRNA PVT1 and p53 on cell proliferation, migration, invasion, and apoptosis. Mouse xenograft model...

Propofol inhibits proliferation and migration of glioma cells by up-regulating lncRNA GAS5
Toxicol In Vitro. 2022 Jan 24:105321. doi: 10.1016/j.tiv.2022.105321. Online ahead of print.ABSTRACTPropofol has been documented to exhibit anti-tumor effects in cancers. However, the underlying mechanisms of its inhibition remain unclear. We conducted this study to investigate the effect of propofol on lncRNA GAS5 in glioma cells, and to probe the role of GAS5 in the anti-tumor effects of propofol. Human glioma cell line U251 were treated with propofol. Cell proliferation, migration, invasion, cell cycle and apoptosis were evaluated, and the expression of GAS5 was measured. After that, we knockdown the expression of GAS5 ...

A Nanoparticle-Conjugated Anti-TBK1 siRNA Induces Autophagy-Related Apoptosis and Enhances cGAS-STING Pathway in GBM Cells
CONCLUSION: The rGO-PEG could be an efficient system facilitating the delivery of specific siRNA. TBK1si/rGO-PEG could be a novel strategy for the treatment of GBM.PMID:34931127 | PMC:PMC8684524 | DOI:10.1155/2021/6521953

EZH2 as a new therapeutic target in brain tumors: Molecular landscape, therapeutic targeting and future prospects
Biomed Pharmacother. 2021 Dec 11;146:112532. doi: 10.1016/j.biopha.2021.112532. Online ahead of print.ABSTRACTBrain tumors are responsible for high mortality and morbidity worldwide. The brain tumor treatment depends on identification of molecular pathways involved in progression and malignancy. Enhancer of zeste homolog 2 (EZH2) has obtained much attention in recent years in field of cancer therapy due to its aberrant expression and capacity in modulating expression of genes by binding to their promoter and affecting methylation status. The present review focuses on EZH2 signaling in brain tumors including glioma, gliobla...

Enhanced expression of Pentraxin-3 in glioblastoma cells correlates with increased invasion and IL8-VEGF signaling axis
In this study, we examined the role of PTX3 in GB growth, angiogenesis, and invasion using in vitro and in vivo GB models, proteomic profiling, molecular and biochemical approaches. Under in vitro conditions, PTX3 over-expression in U87 cells correlated with cell cycle progression, increased migratory potential, and proliferation under hypoxic conditions. Conditioned media containing PTX3 enhanced the angiogenic potential of endothelial cells. While silencing of PTX3 by siRNA decreased the proliferation, migration, and angiogenic potential of U87 cells in vitro. Importantly, PTX3 over-expression increased tumor growth, ang...
