GSE160638 MYC induces immunotherapy and interferon- γ resistance through downregulation of JAK2

As the pathways mediating resistance to immunotherapy are largely unknown, we conducted transcriptome profiling of pre-immunotherapy tumor biopsies from melanoma patients that received PD-1 blockade (n=36) or adoptive cell therapy with tumor infiltrating lymphocytes (n=37). We identified two melanoma-intrinsic mutually exclusive gene programs, which are controlled by interferon- γ and MYC, and determi...

MYC induces immunotherapy and interferon- γ resistance through downregulation of JAK2
Cancer Immunol Res. 2023 Apr 19:CIR-22-0184. doi: 10.1158/2326-6066.CIR-22-0184. Online ahead of print.ABSTRACTImmunotherapy has revolutionized the treatment of advanced melanoma. Because the pathways mediating resistance to immunotherapy are largely unknown, we conducted transcriptome profiling of pre-immunotherapy tumor biopsies from melanoma patients that received PD-1 blockade or adoptive cell therapy with tumor-infiltrating lymphocytes. We identified two melanoma-intrinsic, mutually exclusive gene programs, which were controlled by interferon-γ (IFNγ) and MYC, and the association with immunotherapy outcome. MYC-over...

801 Novel nanoparticles mediate efficient siRNA/mRNA co-delivery to melanoma
Checkpoint immunotherapy has revolutionized treatment for patients with advanced or metastatic melanoma; however, many patients ’ tumors either fail to respond or develop secondary resistance. As a result, there is an urgent need for an off-the-shelf local treatment that can improve therapeutic outcomes by increasing immune activation and reducing immunosuppressive factors. We developed a library of novel lipophilic poly(b eta-amino ester) (PBAE)-based biodegradable nanoparticle (NP) formulations with different backbone, sidechain, and endcap monomers and various lipophilic/linear ratios to efficiently co-deliver mRNA an...

1249 Research of hiperfect-mediated RNA interference targeting hTERT on the biological behavior of the human malignant melanoma cell line A375
Objective: To Research of hiperfect-mediated RNA interference targeting human telomerase reverse transcriptase gene (hTERT) on the biological behavior of the human malignant melanoma cell line A375.Methods: Specific siRNAs with green fluorescent labeling on different concentrations were transfected into A375 cells by using transfection reagent (Hiperfect) at different volumes . The transfection efficiency of cells was determined by fluorescen microscop, and the concentration of siRNA and volume of Hipetfect were selected.

IDO2-siRNA carried by Salmonella combined with nifuroxazide attenuates melanoma growth
CONCLUSION: In conclusion, IDO2-siRNA combined with nifuroxazide therapy could serve a significant role in the treatment of melanoma-bearing mice, enhance the tumor immunity and provide an experimental basis for identifying a novel combination method for the treatment of melanoma clinically.PMID:36999419 | DOI:10.2174/1874467217666230329102030

Axl contributes to efficient migration and invasion of melanoma cells
by Hanshuang Shao, Diana Teramae, Alan Wells Axl, a member of the TAM receptor family has been broadly suggested to play a key role in tumor metastasis. However, the function of Axl in the invasion and metastasis of melanoma, the most lethal skin cancer, remains largely unknown. In the present study, we found that melanoma cell lines present variable protein levels of Axl and Tyro3; interestingly, MerTK is not noted at detectable levels in any of tested MGP (metastatic growth phase) cell lines. Treatment with recombinant human Gas6 significantly activates Akt in the Axl-expressing WM852 and IgR3 lines but just slightly in...

Attenuation of cancer proliferation by suppression of glypican-1 and its pleiotropic effects in neoplastic behavior
Oncotarget. 2023 Mar 21;14:219-235. doi: 10.18632/oncotarget.28388.ABSTRACTGlypicans (GPC1-6) are associated with tumorigenic processes and their involvement in neoplastic behavior has been discussed in different cancer types. Here, a cancer-wide GPC expression study, using clinical cancer patient data in The Cancer Genome Atlas, reveals net upregulation of GPC1 and GPC2 in primary solid tumors, whereas GPC3, GPC5 and GPC6 display lowered expression pattern compared to normal tissues. Focusing on GPC1, survival analyses of the clinical cancer patient data reveal statistically significant correlation between high expression...

Glycolysis regulator PFKP induces human melanoma cell proliferation and tumor growth
ConclusionCollectively, these results indicate that PFKP expression in melanoma cells increases proliferation and glycolytic activity in vitro and promotes tumorigenesis in vivo, suggesting that suppression of PKFP and inhibition of glycolysis may potently suppress melanoma progression.

Use of Iontophoresis Technology for Transdermal Delivery of a Minimal mRNA Vaccine as a Potential Melanoma Therapeutic
Biol Pharm Bull. 2023;46(2):301-308. doi: 10.1248/bpb.b22-00746.ABSTRACTmRNA vaccines have attracted considerable attention as a result of the 2019 coronavirus pandemic; however, challenges remain regarding use of mRNA vaccines, including insufficient delivery owing to the high molecular weights and high negative charges associated with mRNA. These characteristics of mRNA vaccines impair intracellular uptake and subsequent protein translation. In the current study, we prepared a minimal mRNA vaccine encoding a tumor associated antigen human gp10025-33 peptide (KVPRNQDWL), as a potential treatment for melanoma. Minimal mRNA...
