Evaluation of Extended-Release of Piroxicam Loaded Pectin-Zein Hydrogel Microspheres: In Vitro, Ex Vivo, and In Vivo Studies
CONCLUSION: P/Z HM had different drug release behaviors in in vitro and in vivo conditions. However, the ex vivo and in vivo characteristics were similar (R² = 0.99). This seemed reasonable to use the ex vivo method to predict the in vivo drug absorption behavior during the polymeric drug delivery system developmental studies. The P/Z HM formulation maintained the drug dose at the colon site for a long duration and could be applied for delivery of active pharmaceutical and food ingredients to the colon site.PMID:35249486 | DOI:10.2174/1567201819666220304092012 (Source: Current Drug Delivery)

Anti-inflammatory and Immunomodulatory Potency of Selenium-Enriched Probiotic Mutants in Mice with Induced Ulcerative Colitis
In this study, 32 BALB/c male mice were distributed to four groups: the control group, the Piroxicam group which was given 0.8 mg Piroxicam, SP and SB groups which were given 0.8 mg Piroxicam, and plus Lactobacillus plantarum and Bifidobacterium longum selenium-enriched mutants, respectively. Bodyweight; serum content of IgG, IgM, TNF-α, IL-2, IL-6, and IL-10; CBC; myeloperoxidase enzyme activity; histopathological examination of colon and spleen; and expression of TNF-α, IL-2, IL-6, and IL-10 genes in colon and spleen with qRT-PCR were determined. Bodyweight was found to reduce in the Piroxicam group and then recovery i...

1-Allyl-4-hydroxy-2,2-dioxo-N-(4-methoxyphenyl)-1H-2 λ 6,1-benzothiazine-3-carboxamide: polymorphic transition due to grinding with the loss of the biological activity
A study of two polymorphic forms of 1-allyl-4-hydroxy-2,2-dioxo-N-(4-methoxyphenyl)-1-2 λ 6,1-benzothiazine-3-carboxamide (a structural analogue of piroxicam) has revealed some regularities in the crystal structure formation due to different evaporation rates from the tested solvents. The monoclinic polymorph crystallized from ethyl acetate is formed due to a large number of very weak C — H...O and C — H... π interactions as well as one strong stacking interaction. The triclinic polymorph crystallized from N,N-dimethylformamide is formed due to a small number of weak specific interactions and a maximal number of stro...

Gastroprotective Effects of the Aqueous Extract from < em > Taraxacum officinale < /em > in Rats Using Ultrasound, Histology, and Biochemical Analysis
In conclusion, the gastroprotective effect of AETo involves reduction in oxidative stress and inflammatory injury and increase in mucin content. This study advances in the elucidation of mechanisms of gastric protection of T. officinale, contributes to the prospection of new molecules gastroprotective, and proposes the ultrasonographic analyses as a new gastroprotective assessment tool in preclinical studies.PMID:34970327 | PMC:PMC8714386 | DOI:10.1155/2021/8987232 (Source: Evidence-based Complementary and Alternative Medicine)

Design, Synthesis, Docking Study and Biological Evaluation of 4-Hydroxy-2 < em > H < /em > -benzo[ < em > e < /em > ][1,2]thiazine-3-carboxamide 1,1-dioxide Derivatives as Anti-HIV Agents
Iran J Pharm Res. 2021 Summer;20(3):1-12. doi: 10.22037/ijpr.2020.114153.14695.ABSTRACTA novel series of benzothiazine-3-carboxamide 1,1-dioxide derivatives by modifying the piroxicam scaffold was designed, synthesized, and evaluated as anti-HIV agents. The 1,2-benzothiazine-3-carboxamide 1,1-dioxide scaffold consists of hydroxy and carboxamide groups as a chelating motif to form an interaction with Mg2+ ions within the integrase active site as a target. Most of the compounds displayed encouraging anti-HIV activity in a cell-based assay. Among them, compounds 13d, 13l and 13m were the most potent with EC50 values ranging f...

Molecules, Vol. 26, Pages 7375: Modulating Properties of Piroxicam, Meloxicam and Oxicam Analogues against Macrophage-Associated Chemokines in Colorectal Cancer
ystek-Korpacka The mechanisms underlying the antineoplastic effects of oxicams have not been fully elucidated. We aimed to assess the effect of classic and novel oxicams on the expression/secretion of macrophage-associated chemokines (RTqPCR/Luminex xMAP) in colorectal adenocarcinoma cells, and on the expression of upstream the non-steroidal anti-inflammatory drug (NSAID)-activated genes NAG1, NFKBIA, MYD88, and RELA, as well as at the chemokine profiling in colorectal tumors. Meloxicam downregulated CCL4 9.9-fold, but otherwise the classic oxicams had a negligible/non-significant effect. Novel analogues with a thiazin...
