Three species multiplexing of fluorescent dyes and gold nanoclusters recovered with fluorescence lifetime correlation spectroscopy
Biochim Biophys Acta Gen Subj. 2024 Mar 27:130611. doi: 10.1016/j.bbagen.2024.130611. Online ahead of print.ABSTRACTBiosensor applications often require the simultaneous detection of multiple analytes, with a clear need to go beyond the traditional multiplexing relying on distinct fluorescent dyes across the visible spectrum. Fluorescence lifetime correlation spectroscopy (FLCS) is a powerful approach taking advantage of the fluorescence lifetime information to separate the contributions of different fluorescent species with overlapping emission spectra. However, so far FLCS detection has been demonstrated only on binary m...

Global control of RNA polymerase II
Biochim Biophys Acta Gene Regul Mech. 2024 Mar 27:195024. doi: 10.1016/j.bbagrm.2024.195024. Online ahead of print.ABSTRACTRNA polymerase II (Pol II) is the multi-protein complex responsible for transcribing all protein-coding messenger RNA (mRNA). Most research on gene regulation is focused on the mechanisms controlling which genes are transcribed when, or on the mechanics of transcription. How global Pol II activity is determined receives comparatively less attention. Here, we follow a Pol II molecule's life from complex assembly to nuclear import, enzymatic activity, and degradation. We focus on how Pol II spends its ti...

METTL3 orchestrates glycolysis by stabilizing the c-Myc/WDR5 complex in triple-negative breast cancer
CONCLUSION: METTL3 binds to the c-Myc/WDR5 complex and promotes glycolysis, which plays a powerful role in promoting TNBC progression. Our findings further broaden our understanding of the role and mechanism of action of METTL3, and may open up new therapeutic avenues for effective treatment of TNBC with high c-Myc expression.PMID:38547933 | DOI:10.1016/j.bbamcr.2024.119716 (Source: Biochimica et Biophysica Acta)

Compound SJ-12 attenuates streptozocin-induced diabetic cardiomyopathy by stabilizing SERCA2a
Biochim Biophys Acta Mol Basis Dis. 2024 Mar 26;1870(5):167140. doi: 10.1016/j.bbadis.2024.167140. Online ahead of print.ABSTRACTHeart failure (HF) is one of the major causes of death among diabetic patients. Although studies have shown that curcumin analog C66 can remarkably relieve diabetes-associated cardiovascular and kidney complications, the role of SJ-12, SJ-12, a novel curcumin analog, in diabetic cardiomyopathy and its molecular targets are unknown. 7-week-old male C57BL/6 mice were intraperitoneally injected with single streptozotocin (STZ) (160 mg/kg) to develop diabetic cardiomyopathy (DCM). The diabetic mice w...

Knockout of Brca1-interacting factor Ola1 in female mice induces tumors with estrogen suppressible centrosome amplification
Biochim Biophys Acta Mol Basis Dis. 2024 Mar 25:167138. doi: 10.1016/j.bbadis.2024.167138. Online ahead of print.ABSTRACTObg-like ATPase 1 (OLA1) is a binding protein of Breast cancer gene 1 (BRCA1), germline pathogenic variants of which cause hereditary breast cancer. Cancer-associated variants of BRCA1 and OLA1 are deficient in the regulation of centrosome number. Although OLA1 might function as a tumor suppressor, the relevance of OLA1 deficiency to carcinogenesis is unclear. Here, we generated Ola1 knockout mice. Aged female Ola1+/- mice developed lymphoproliferative diseases, including malignant lymphoma. The lymphoma...

CX3CR1 deficiency promotes resolution of hepatic ischemia-reperfusion injury by regulating homeostatic function of liver infiltrating macrophages
Biochim Biophys Acta Mol Basis Dis. 2024 Mar 25:167130. doi: 10.1016/j.bbadis.2024.167130. Online ahead of print.ABSTRACTHepatic ischemia-reperfusion injury(HIRI) remains to be an unsolved risk factor that contributes to organ failure after liver surgery. Our clinical retrospective study showed that lower donor liver CX3-C chemokine receptor-1(CX3CR1) mRNA expression level were correlated with upregulated pro-resolved macrophages receptor MERTK, as well as promoted restoration efficiency of allograft injury in liver transplant. To further characterize roles of CX3CR1 in regulating resolution of HIRI, we employed murine liv...

The Y49H cytochrome c variant enhances megakaryocytic maturation of K-562 cells
Biochim Biophys Acta Mol Basis Dis. 2024 Mar 24:167134. doi: 10.1016/j.bbadis.2024.167134. Online ahead of print.ABSTRACTFive pathogenic variants in the gene encoding cytochrome c (CYCS) associated with mild autosomal dominant thrombocytopenia have been reported. Previous studies of peripheral blood CD34+ or CD45+ cells from subjects with the G42S CYCS variant showed an acceleration in megakaryopoiesis compared to wild-type (WT) cells. To determine whether this result reflects a common feature of the CYCS variants, the c.145T>C mutation (Y49H variant) was introduced into the endogenous CYCS locus in K-562 cells, which u...

Dipeptidyl-peptidase 9 regulates the dynamics of tumorigenesis and metastasis in breast cancer
Biochim Biophys Acta Mol Basis Dis. 2024 Mar 24:167133. doi: 10.1016/j.bbadis.2024.167133. Online ahead of print.ABSTRACTThe cytosolic dipeptidyl-aminopeptidase 9 (DPP9) cleaves protein N-termini post-proline or -alanine. Our analysis of DPP9 mRNA expression from the TCGA 'breast cancer' data set revealed that low/intermediate DPP9 levels are associated with poor overall survival of breast cancer patients. To unravel the impact of DPP9 on breast cancer development and progression, the transgenic MMTV-PyMT mouse model of metastasizing breast cancer was used. In addition, tissue- and time-controlled genetic deletion of DPP9 ...

FXR/Menin-mediated epigenetic regulation of E2F3 expression controls β-cell proliferation and is increased in islets from diabetic GK rats after RYGB
Biochim Biophys Acta Mol Basis Dis. 2024 Mar 24:167136. doi: 10.1016/j.bbadis.2024.167136. Online ahead of print.ABSTRACTFarnesoid X receptor (FXR) improves the function of islets, especially in the setting of Roux-en-Y gastric bypass (RYGB). Here we investigated how FXR activation regulates β-cell proliferation and explored the potential link between FXR signaling and the menin pathway in controlling E2F3 expression, a key transcription factor for controlling adult β-cell proliferation. Stimulation with the FXR agonist GW4064 or chenodeoxycholic acid (CDCA) increased E2F3 expression and β-cell proliferation. Consistent...
