GABA transporter mGat4 is involved in multiple neural functions in mice
Biochim Biophys Acta Mol Cell Res. 2024 Apr 30:119740. doi: 10.1016/j.bbamcr.2024.119740. Online ahead of print.ABSTRACTγ-Aminobutyric acid (GABA) is the major inhibitory neurotransmitter in the central nervous system. The termination of GABA transmission is through the action of GABA transporters (GATs). mGAT4 (encoded by Slc6a11) is another GAT besides GAT1 (encoded by Slc6a1) that functions in GABA reuptake in CNS. Research on the function of mGAT4 is still in its infancy. We developed an mGat4 knockout mouse model (mGat4-/- mice) and performed a series of behavioral analyses for the first time to study the effect of m...

Inhibition of δ-catenin palmitoylation slows the progression of prostate cancer
In this study, we utilized prostate cancer cells overexpressing mutant δ-catenin (J6A cells) to induce a depalmitoylation phenotype and investigate its effect on prostate cancer. Our results indicated that depalmitoylation of δ-catenin not only reduced its membrane expression but also promoted its degradation in the cytoplasm, resulting in a decrease in the effect of EGFR and E-cadherin signaling. Consequently, depalmitoylation of δ-catenin reduced the proliferation and metastasis of prostate cancer cells. Our findings provide novel insights into potential therapeutic strategies for controlling the progression of prosta...

Procollagen-lysine 2-oxoglutarate 5-dioxygenase 2 promotes collagen cross-linking and ECM stiffening to induce liver fibrosis
Biochim Biophys Acta Mol Basis Dis. 2024 May 1;1870(5):167205. doi: 10.1016/j.bbadis.2024.167205. Online ahead of print.ABSTRACTProcollagen-lysine 2-oxoglutarate 5-dioxygenase 2 (Plod2) is a key collagen lysyl hydroxylase mediating the formation of collagen fiber and stabilized collagen cross-links, and has been identified in several forms of fibrosis. However, the potential role and regulatory mechanism of Plod2 in liver fibrosis remain unclear yet. Mouse liver fibrosis models were induced by injecting carbon tetrachloride (CCl4) intraperitoneally. The morphology and alignment of collagen was observed under transmission a...

Two pathways to understanding electron transfer in reaction centers from photosynthetic Bacteria: A comparison of Rhodobacter sphaeroides and Rhodobacter capsulatus mutants
Biochim Biophys Acta Bioenerg. 2024 Apr 29:149047. doi: 10.1016/j.bbabio.2024.149047. Online ahead of print.ABSTRACTThe rates, yields, mechanisms and directionality of electron transfer (ET) are explored in twelve pairs of Rhodobacter (R.) sphaeroides and R. capsulatus mutant RCs designed to defeat ET from the excited primary donor (P*) to the A-side cofactors and re-direct ET to the normally inactive mirror-image B-side cofactors. In general, the R. sphaeroides variants have larger P+HB- yields (up to ~90 %) than their R. capsulatus analogs (up to ~60 %), where HB is the B-side bacteriopheophytin. Substitution of Tyr for ...

Key interactions of RNA polymerase with 6S RNA and secondary channel factors during pRNA synthesis
Biochim Biophys Acta Gene Regul Mech. 2024 Apr 29:195032. doi: 10.1016/j.bbagrm.2024.195032. Online ahead of print.ABSTRACTSmall non-coding 6S RNA mimics DNA promoters and binds to the σ70 holoenzyme of bacterial RNA polymerase (RNAP) to suppress transcription of various genes mainly during the stationary phase of cell growth or starvation. This inhibition can be relieved upon synthesis of short product RNA (pRNA) performed by RNAP from the 6S RNA template. Here, we have shown that pRNA synthesis depends on specific contacts of 6S RNA with RNAP and interactions of the σ finger with the RNA template in the active site of ...

Regulation of VEGF gene expression by bisacridine derivative through promoter i-motif for cancer treatment
CONCLUSION: B09 could specifically regulate VEGF gene expression, possibly through interacting with promoter i-motif structure. As a lead compound, B09 could be further developed for innovative anti-cancer agent targeting VEGF.PMID:38685534 | DOI:10.1016/j.bbagen.2024.130631 (Source: Biochimica et Biophysica Acta)

Interaction of L-phenylalanine with carbonyl groups in mixed lipid membranes
Biochim Biophys Acta Biomembr. 2024 Apr 28:184328. doi: 10.1016/j.bbamem.2024.184328. Online ahead of print.ABSTRACTThe interaction of L-Phe with the membrane components, i.e., lipids and proteins, has been discussed in the current literature due to the interest to understand the effect of single amino acids in relation to the formation of amyloid aggregates. In the present work, it is shown that L-Phe interacts with 9:1 DMPC (1,2-dimyristoyl-sn-glycero-3 phosphocholine)/DPPC (1,2-dipalmitoyl-sn-glycero-3 phosphocholine) mixtures but not in the 1:9 one. An important observation is that the interaction disappears when DPPC ...

The molecular arrangement of ceramides in the unit cell of the long periodicity phase of stratum corneum models shows a high adaptability to different ceramide head group structures
In this study, the impact of the CER head group structure on the lipid arrangement and barrier functionality was investigated using lipid models forming the LPP. The results demonstrate that the positions of CER N-(tetracosanoyl)-sphingosine (CER NS) and CER N-(tetracosanoyl)-phytosphingosine (CER NP), two essentials CER subclasses, are not influenced by the addition of another CER subclass (N-(tetracosanoyl)-dihydrosphingosine (CER NdS), N-(2R-hydroxy-tetracosanoyl)-sphingosine (CER AS) or D-(2R-hydroxy-tetracosanoyl)-phytosphingosine (CER AP)). However, differences are observed in the lipid organization and the hydrogen ...
