Analysis of polymeric nanoparticle properties for siRNA/DNA delivery in a tumor xenograft tissue slice air-liquid interface model
CONCLUSION: The ex vivo 3D tissue slice ALI culture model is a powerful system that allows the effective evaluation of biological nanoparticle efficacy and biocompatibility in an intact tissue environment. It is comparably inexpensive, time-saving, and follows the 3R principle, while allowing the identification of critical nanoparticle properties and optimal candidates for in vivo applications.PMID:36541426 | DOI:10.1002/biot.202200415

Roles of MEF2A and HOXA5 in the transcriptional regulation of the bovine < em > Fox < /em > O1 gene
Anim Biotechnol. 2022 Nov 30:1-13. doi: 10.1080/10495398.2022.2150632. Online ahead of print.ABSTRACTThe Forkhead box factor 1 (FoxO1) gene plays a vital role in the growth and development of skeletal muscle. In the present study, expression analysis of the bovine FoxO1 gene exhibited the highest expression in longissimus dorsi muscle followed by its expression in adipose tissue. Moreover, high mRNA expression of FoxO1 gene was found in differentiated bovine myoblasts and adipocytes at day 6 of induced differentiation (p < 0.05). The regulatory pattern of the bovine FoxO1 gene was investigated through screening and dual...

Current advancements in self-assembling nanocarriers-based siRNA delivery for cancer therapy
Colloids Surf B Biointerfaces. 2022 Nov 2;221:113002. doi: 10.1016/j.colsurfb.2022.113002. Online ahead of print.ABSTRACTDifferent therapeutic practices for treating cancers have significantly evolved to compensate and/or overcome the failures in conventional methodologies. The demonstrated potentiality in completely inhibiting the tumors and in preventing cancer relapse has made nucleic acids therapy (NAT)/gene therapy as an attractive practice. This has been made possible because NAT-based cancer treatments are highly focused on the fundamental mechanisms - i.e., silencing the expression of oncogenic genes responsible fo...

An miRISC-initiated DNA nanomachine for monitoring MicroRNA activity in living cells
Biosens Bioelectron. 2022 Oct 29;220:114828. doi: 10.1016/j.bios.2022.114828. Online ahead of print.ABSTRACTMicroRNAs (miRNAs) play an important role in post-transcriptional regulation of gene expression. However, methods to accurately detect miRNA activity in living cells are still limited. Here we developed a DNA nanomachine initiated by a miRNA-induced silencing complex (miRISC) for imaging miRNA activity in living cells. miRISC-mediated RNA cleavage reaction activated the DNA nanomachine by the specific cleavage of an RNA strand on the machine, resulting in autonomous movement of the walking leg around the AuNP surface...
