Local Silencing of Connective Tissue Growth Factor by siRNA/Peptide Improves Dermal Collagen Arrangements
Conclusion:CTGF expression was down-regulated to 40% of control by CTGF siRNA/KALA (1:24) complexes (p <  0.01) and collagen lattice contraction was inhibited. However, down-regulated of CTGF by CTGF siRNA/MPG∆NLS complexes was not statistically significant. CTGF KALA-treated wound appeared with well formed-basket weave pattern of collagen fibrils with mean diameter of 128  ± 22 nm (n = 821). Mismatch siRNA/KALA-treated wound showed a high frequency of parallel small diameter fibrils (mean 90 ± 20 nm, n = 563).Conclusion:Controlling over-expression of CTGF by peptide-mediated siRNA delivery cou...

Cyclic peptide-based nanostructures as efficient siRNA carriers.
Authors: Panigrahi B, Singh RK, Mishra S, Mandal D Abstract RNA interference shows a great strategy for biological studies; however, delivering of small interfering RNA (siRNA) remains challenging. Although several delivery vehicles, including cell-penetrating peptides, have been developed, their implementation is often restricted because of their endosomal entrapment. Herein, we report the formation of self-assembled nanostructures from rationally designed cyclic peptides and explore them for efficient delivery of functional biomacromolecules such as siRNA into mammalian cells. The newly obtained soft materials ma...

Dendrobium moniliforme stem extract inhibits lipoteichoic acid-induced inflammatory responses by upregulation of heme oxygenase-1.
In this study, we investigated anti-inflammatory effects of the aqueous extract of D. moniliforme stems (DM) in response to lipoteichoic acid (LTA), a major constituent of the cell wall of Gram-positive bacteria. DM inhibited LTA-induced expression of a pro-inflammatory mediator inducible nitric oxide synthase (iNOS) in the murine macrophages. And DM induced expression of heme oxygenase-1 (HO-1) at the transcriptional level. Conversely, the knockdown of HO-1 expression by siRNA markedly reversed the inhibitory effects of DM on LTA-induced iNOS expression. We also demonstrated that nuclear translocation of Nrf2 was increase...

Development and characterization of a novel cationic PEGylated niosome-encapsulated forms of doxorubicin, quercetin and siRNA for the treatment of cancer by using combination therapy.
Authors: Hemati M, Haghiralsadat F, Yazdian F, Jafari F, Moradi A, Malekpour-Dehkordi Z Abstract The aim of this study was to optimize the cationic PEGylated niosome-containing anti-cancer drugs and siRNA to enhance the therapeutic response. Therefore, various surfactant-based (tween-60) vesicles of doxorubicin (DOX; a chemotherapeutic drug) and quercetin (QC; a chemosensitizer) were prepared. To load siRNA on niosomes, 1, 2-dioleoyl-3-trimethylammonium-propane (DOTAP) was used as a cationic lipid. The optimum formulation containing tween-60:cholesterol:DPPC:DOTAP:DSPE-PEG2000 at 49.5:5.5:15:25:5 demonstrated that ...

The combinational effect of E6/E7 siRNA and anti-miR-182 on apoptosis induction in HPV16-positive cervical cells.
Authors: Javadi H, Lotfi AS, Hosseinkhani S, Mehrani H, Amani J, Soheili ZS, Hojati Z, Kamali M Abstract In the present research, we assumed that reducing the amounts of E6 and E7 oncoproteins by a specific siRNA sequence and recovering p53 and RB proteins, along with the recovery of the FOXO1 protein by applying anti-miR-182, would increase apoptosis and reduce proliferation rate in cancer cells. The HPV16-positive CaSki cervical cancer cell line was used. 48 hours after transfection of siRNA for targeting E6 and E7 oncoproteins and anti-miR-182, expression of its cellular targets p53, p21 and FOXO1 was assessed...

Intracellular miRNA or siRNA delivery and function.
Abstract Nanocarrier-mediated delivery and release of short non-coding RNA (e.g., miRNA or siRNA) into the cells with subsequent suppression of the populations of some of the mRNAs and proteins is of interest in the context of the development of a new generation of drugs. Bearing in mind such applications, the author shows the specifics of the corresponding transient kinetics by using three generic models without and with feedback resulting in bistability in the gene expression. In the absence of feedback, the suppression of the mRNA and protein population is transient. In the case of bistable kinetics, non-coding...

BST2 promotes cell proliferation, migration and induces NF- κB activation in gastric cancer.
CONCLUSION: BST2 possesses the oncogenic potential in GC by regulating the proliferation, apoptosis, and migratory ability of GC cells, thereby BST2 could be a potential therapeutic target for the treatment of GC. PMID: 29774441 [PubMed - as supplied by publisher]

Near-infrared light-activatable siRNA delivery by microcapsules for combined tumour therapy.
Authors: Rui Y, Pang B, Zhang J, Liu Y, Hu H, Liu Z, Ama Baidoo S, Liu C, Zhao Y, Li S Abstract A polyelectrolyte microcapsule-based layer-by-layer (LbL) technique has been widely used as a multifunctional vehicle for combined tumor therapy. Meanwhile, with the rapid development of combined tumour therapy, the challenge for designing multifunctional drug delivery system has attracted much more attention. Herein, we developed a new type of microcapsule (MC) system called MPA@siRNA@DOX@MC, which conjugated with siRNA and DOX as well as ICG-Der-02 (MPA) by electrostatic absorption. MPA as indocyanine green (ICG) fluor...
