Hypoxic and osmotic expression of Kir2.1 potassium channels in retinal pigment epithelial cells: Contribution to vascular endothelial growth factor expression
Exp Eye Res. 2021 Aug 20:108741. doi: 10.1016/j.exer.2021.108741. Online ahead of print.ABSTRACTRetinal pigment epithelial (RPE) cells express different subtypes of inwardly rectifying potassium (Kir) channels. We investigated whether human and rat RPE cells express genes of strongly rectifying Kir2 channels. We also determined the hypoxic and hyperosmotic regulation of Kir2.1 gene expression in cultured human RPE cells and the effects of siRNA-mediated knockdown of Kir2.1 on VEGFA expression, VEGF secretion, proliferation, and viability of the cells. Extracellular hyperosmolarity was induced by addition of NaCl or sucrose...

Black carbon induces complement activation via NLRP3 inflammasome in human corneal epithelial cells
CONCLUSIONS: BC particles, including FBC and OBC, triggered complement activation, increasing the protein level of C5b-9 in cultured HCECs. siRNA targeting NLRP3 to inhibit NLRP3 generation reduced C5b-9 protein level in HCECs treated with FBC or OBC particles, indicating that BC induces complement activation potentially through the NLRP3 inflammasome in HCECs.PMID:34375537 | DOI:10.1080/02713683.2021.1967404

Corneal lymphangiogenesis in dry eye disease is regulated by substance P/neurokinin-1 receptor system through controlling expression of vascular endothelial growth factor receptor 3
CONCLUSIONS: SP/NK1R system promotes lymphangiogenesis in vitro and NK1R antagonism suppresses pathologic corneal lymphangiogenesis in DED in vivo.PMID:34311077 | DOI:10.1016/j.jtos.2021.07.003

Inhibition of enhancer of zeste homolog 2 prevents corneal myofibroblast transformation in vitro
CONCLUSION: Inhibition of EZH2 suppresses corneal myofibroblast activation and ECM protein synthesis, and could serve as a novel therapeutic target for preventing corneal scarring.PMID:33992624 | DOI:10.1016/j.exer.2021.108611

Astaxanthin mediated regulation of VEGF through HIF1 α and XBP1 signaling pathway: An insight from ARPE-19 cell and streptozotocin mediated diabetic rat model
Exp Eye Res. 2021 Mar 28:108555. doi: 10.1016/j.exer.2021.108555. Online ahead of print.ABSTRACTBreakdown of outer blood-retina barrier (BRB) has been associated with the pathogenesis of diabetic retinopathy (DR) and diabetic macular edema (DME). Vascular endothelial growth factor (VEGF) might play a detrimental role in the pathogenesis of DME, a major clinical manifestation of DR. In the present study, we investigated the inhibitory mechanism of astaxanthin on VEGF and its upstream signaling pathways under in vitro and in vivo conditions. Astaxanthin has been observed to downregulate VEGF expression under hyperglycemic (H...

MicroRNA Let-7c-5p-mediated regulation of ERCC6 disrupts autophagic flux in age-related cataract via the binding to VCP
CONCLUSION: In ARC, Let-7c-5p-mediated downregulation of ERCC6 might prevent the degradation of autophagic vacuoles. CSB binds to VCP, inducing autophagosomes to combine with lysosomes and be degraded.PMID:33703976 | DOI:10.1080/02713683.2021.1900273

RNA therapeutics in ophthalmology - translation to clinical trials.
Abstract The use of RNA interference technology has proven to inhibit the expression of many target genes involved in the underlying pathogenesis of several diseases affecting various systems. First established in in vitro and later in animal studies, small interfering RNA (siRNA) and antisense oligonucleotide (ASO) therapeutics are now entering clinical trials with the potential of clinical translation to patients. Gene-silencing therapies have demonstrated promising responses in ocular disorders, predominantly due to the structure of the eye being a closed and compartmentalised organ. However, although the effic...

LncRNA-MALAT1/miRNA-204-5p/Smad4 Axis Regulates Epithelial-Mesenchymal Transition, Proliferation and Migration of Lens Epithelial Cells.
Conclusions: Our findings reveal that MALAT1 may regulate EMT, proliferation, and migration of LECs as a ceRNA by "sponging" miR-204-5p and targeting Smad4, and serve as a promising therapeutic target in preventing PCO. PMID: 33327804 [PubMed - as supplied by publisher]

Minocycline prevents the inflammatory response after retinal detachment, where microglia phenotypes being regulated through A20.
Abstract Retinal detachment (RD) is a severe sight-threatening complication that can be caused by a multitude of retinal diseases. It has been evidenced that minocycline exerts neuroprotective effects by targeting microglia in the pathogenesis of massive ocular lesions including RD, but mechanisms remain elusive. We carried out this research to elucidate the potential mediators that link RD-induced vision loss with microglia reactivity by discussing effects of minocycline on cytokine levels and A20, a negative regulator of inflammation. Minocycline or vehicle was intraperitoneally administrated immediately after R...
