Prenylflavonoids from fruit of Macaranga tanarius promote glucose uptake via AMPK activation in L6 myotubes
AbstractSkeletal muscle is a major tissue of glucose consumption and plays an important role in glucose homeostasis. Prenylflavonoids, a component ofMacaranga tanarius fruits, have been reported to have antioxidant, antibacterial, and anticancer effects. However, the effects of these compounds on skeletal muscle glucose metabolism are unclear. Here, we isolated five prenylflavonoids fromM. tanarius fruits, and investigated the mechanism of action of these compounds on skeletal muscle cells using L6 myotubes. We found that isonymphaeol B and 3 ′-geranyl naringenin increased glucose uptake in a dose-dependent manner. Furth...

The hepatoprotective efficacy and biological mechanisms of three phenylethanoid glycosides from cistanches herba and their metabolites based on intestinal bacteria and network pharmacology
AbstractEchinacoside (ECH), acteoside (ACT), and isoacteoside (ISAT), the typical phenylethanoid glycosides (PhGs) in cistanches herba, have various pharmacological activities. However, the ECH, ACT and ISAT have extremely low oral bioavailability, which is related to their metabolism under the intestinal flora. Previous studies showed that intestinal metabolites were the hepatoprotective substances in vivo, but the research on whether PhGs has effects without intestinal bacteria has not been studied. In this paper, ECH, ACT and ISAT were incubated with human or rat intestinal bacteria for 36 h. After incubating with human...

Polymorphism analysis of TCP gene region to intraspecific analysis of Paeonia lactiflora , and application for authentication of Paeoniae Radix
AbstractWe attempted to conduct an intraspecific analysis of 30 peony cultivars in Japan and to authenticate five herbal medicine samples derived from Paeoniae Radix by polymorphism analysis of theTEOSINTE BRANCHED1, CYCLOIDEA, and PCF (TCP) gene region. We focused on cultivar-dependent differences in leaf margin undulation and analyzed the sequence of the relatedTCP gene region. As a result, we found that the nucleotide sequences of 29 cultivars ofPaeonia lactiflora except ‘America’ exhibit interspecific variations compared with the nucleotide sequences ofPaeonia suffruticosa andPaeonia tenuifolia. Therefore, in the d...

Four new xanthones and their cytotoxicity from the stems of Garcinia schomburgkiana
AbstractFour new xanthones, named schomburgones C-F (1 ‒4), along with six known xanthones (5 ‒10) were isolated from the stems ofGarcinia schomburgkiana. Their structures were determined by spectroscopic analysis especially 1D and 2D NMR spectroscopies. The isolated compounds were evaluated for their cytotoxicity against five human cancer cell lines. Furanoxanthones4 ‒6 showed potent cytotoxicity against four cell lines (KB, HeLa S3, MCF-7 and Hep G2) with IC50 values in the range of 0.18 ‒9.95 µM.Graphic abstract (Source: Journal of Natural Medicines)

Podogigants A and B, two new potentiators of amphotericin B activity, from Sordariomycete Podostroma giganteum
AbstractTwo new compounds, podogigants A (1) and B (2), were isolated from the culture broth ofPodostroma giganteum. This is the first report on the identification of secondary metabolites inP. giganteum. The structures of1 and2 were elucidated through spectroscopic analysis, including 2D NMR spectroscopy assisted by chemical derivatization, which revealed the presence of farnesyl- and geranyl-hydroquinone structures, respectively. Compounds1 and2 exhibited no antifungal activity even at a concentration of 64  μg/mL, whereas they potentiated amphotericin B (AmB) activity against several species of fungi. In particular,1 ...
