Molecules, Vol. 28, Pages 7099: Silica-Based Stationary Phase with Surface Bound N-Acetyl-glucosamine for Hydrophilic Interaction Liquid Chromatography
ssi A hydrophilic silica-based stationary phase with surface bound N-acetylglucosamine (GlcNAc-silica) was prepared in house and characterized physically via Fourier transform infrared (FTIR) analysis and thermogravimetric analysis (TGA) and chromatographically over a wide range of mobile phase compositions. While both FTIR and TGA confirmed the attachment of the GlcNAc ligands to the silica surface, the chromatographic evaluation of GlcNAc-silica with polar and slightly polar standard solutes (e.g., sugars, nucleic acid fragments, phenolic, and benzoic acid derivatives) yielded the typical hydrophilic interaction liqu...

Dechlorination of wastewater from shell-based glucosamine processing by mangrove wetland-derived fungi
In this study, mangrove wetland-derived fungi were first tested for their salt tolerance, and the highly tolerant isolates were cultured in shrimp processing wastewater and the chloride concentration was monitored. Notably, the filamentous fungal species Aspergillus piperis could remove over 70% of the chloride in the wastewater within 3 days, with the fastest biomass increase (2.01 times heavier) and chloride removal occurring between day one and two. The chloride ions were sequestered into the fungal cells. The genome of this fungal species contained Cl− conversion enzymes, which may have contributed to the ion remov...

Composition, structural properties and immunomodulatory activity of several aqueous Pleurotus β-glucan-rich extracts
Int J Biol Macromol. 2023 Oct 10:127255. doi: 10.1016/j.ijbiomac.2023.127255. Online ahead of print.ABSTRACTIn this work, aqueous extracts from six different Pleurotus species were obtained and their yield, gross composition, β-glucan content, monosaccharide profile, thermal stability, molecular weight distribution, and FT-IR were analyzed before and after purification through ethanol precipitation of the carbohydrate-rich fractions. The bioactivity (anti-inflammatory and immunomodulatory activity) of the various fractions obtained was also analyzed in three different cell cultures and compared with a lentinan control. Th...

Multicomponent analysis of dietary supplements containing glucosamine and chondroitin: comparative low- and high-field NMR spectroscopic study
Anal Sci. 2023 Oct 11. doi: 10.1007/s44211-023-00433-2. Online ahead of print.ABSTRACTWith the prevalence of glucosamine- and chondroitin-containing dietary supplements for people with osteoarthritis in the marketplace, it is important to have an accurate and reproducible analytical method for the quantitation of these compounds in finished products. NMR spectroscopic method based both on low- (80 MHz) and high- (500-600 MHz) field NMR instrumentation was established, compared and validated for the determination of chondroitin sulfate and glucosamine in dietary supplements. The proposed method was applied for analysis of 2...

Computational and Biophysical Approaches to Identify Cell Wall-Associated Modulators in Salmonella enterica serovar Typhi
Methods Mol Biol. 2024;2727:35-55. doi: 10.1007/978-1-0716-3491-2_4.ABSTRACTAn increase in the number of antibiotic-resistant bacterial pathogens, in recent times, has posed a great challenge for treating the affected patients. This has paved the way for the development and design of antibiotics against the previously less explored newer targets. Among these, peptidoglycan (PG) biosynthesis serves as a promising target for the design and development of novel drugs. The peptidoglycan cell wall synthesis in bacteria is essential for its viability. The enzyme class, Mur ligases, plays a key role in PG biosynthesis. Therefore,...

Visualization of a Cell Wall Hydrolase Inhibitor in Fusobacterium nucleatum by Immunofluorescence Microscopy
Methods Mol Biol. 2024;2727:27-33. doi: 10.1007/978-1-0716-3491-2_3.ABSTRACTInnately present in tears, saliva and mucosal secretions, lysozyme provides a critical defensive strategy to the host by cleaving the β-1,4-glycosidic bonds between N-acetylmuramic acid and N-acetyl-D-glucosamine residues of peptidoglycan of invading bacteria, leading to bacterial lysis. To counter this class of cell wall hydrolase enzymes, bacteria produce several lysozyme inhibitors, a representative of which, MliC, was identified in Escherichia coli, Pseudomonas aeruginosa, and various bacterial species. The Gram-negative oral anaerobe Fusobact...

Antioxidant activity of glucosamine and its effects on ROS production, Nrf2, and O-GlcNAc expression in HMEC-1 cells
Curr Res Toxicol. 2023 Sep 22;5:100128. doi: 10.1016/j.crtox.2023.100128. eCollection 2023.ABSTRACTGlucosamine (GlcN) is the most used supplement for osteoarthritis treatment. In vitro studies have related GlcN to beneficial and detrimental effects on health. The aim of this study was to evaluate the effects of O-linked-N-acetylglucosaminylation (O-GlcNAc) on GlcN-induced ROS production and Nrf2 expression in human dermal microvascular endothelial cells-1 (HMEC-1) and to evaluate the antioxidant capacity of GlcN compared to well-known antioxidants. For this, we evaluate the antioxidant capacity by in vitro assays. Besides,...

Synthesis of pyrrole-2-carboxylic acid from cellulose- and chitin-based feedstocks discovered by the automated route search
In this study, a protocol was developed to identify potential transformation pathways that allow for the use of feedstocks from different origins. By analyzing existing knowledge on chemical reactions in large databases, several promising synthetic routes were shortlisted, with the reaction of D-glucosamine and pyruvic acid being the most interesting to make pyrrole-2-carboxylic acid (PCA). The optimized synthetic conditions resulted in 50% yield of PCA, with insights gained from temperature variant NMR studies. The use of substrates obtained from two different bio-feedstock bases, namely cellulose and chitin, allowed for ...
