GSE243667 Increased AID Results in Mutations at the CRLF2 Locus Implicated in Latin American ALL Health Disparities
Contributors : Richard L Frock ; Valeria Rangel ; Jason N Sterrenberg ; Nicholas R PannunzioSeries Type : OtherOrganism : Homo sapiensActivation-induced cytidine deaminase (AID) is a B cell-specific base editor required during class switch recombination and somatic hypermutation for B cell maturation and antibody diversification. However, it has also been implicated as a factor in the etiology of several B cell malignancies. Evaluating the AID-induced mutation load in patients at-risk for certain types of blood cancers is critical in assessing disease severity and treatment options. Here, we have developed a digital PCR (...

Understanding hemoglobin contribution to high-dose methotrexate disposition —population pharmacokinetics in pediatric patients with hematological malignancies
ConclusionThe developed population pharmacokinetic model can contribute to the therapy optimization during HDMTX in pediatric patients with ALL and NHL. In addition to renal function and body weight, it describes the influence of hemoglobin on CL, allowing better understanding of its contribution to the disposition of HDMTX. RuZn NPs with electroactivity and oxidase-like property for dual-mode anti-cancer drug monitoring
Talanta. 2024 Apr 8;274:126075. doi: 10.1016/j.talanta.2024.126075. Online ahead of print.ABSTRACT6-mercaptopurine (6-MP) as the effective anti-cancer drug was used for the treatment of Crohn's disease and acute lymphoblastic leukaemia, but the response to maintenance therapy was variable with individual differences. In order to control the dosage and decrease the side effects of 6-MP, a sensitive and stable assay was urgently needed for 6-MP monitoring. Herein, RuZn NPs with electrochemical oxidation property and oxidase-like activity was proposed for dual-mode 6-MP monitoring. Burr-like RuZn NPs were prepared and explore...

The DMT1 isoform lacking the iron ‐response element regulates normal and malignant hematopoiesis via NOTCH pathway activation
In this report, we investigated the function of DMT1 nonIRE in normal and malignant hematopoiesis. Knockdown of Dmt1 nonIRE in mice showed that it has non-canonical functions in hematopoietic stem cell differentiation: its knockdown suppressed development along the myeloid and lymphoid lineages, while promoting the production of platelets. These phenotypic effects on the hematopoietic system induced by Dmt1 nonIRE knockdown were linked to suppression of Notch/Myc pathway activity. Conversely, our data indicate a non-canonical function for DMT1 nonIRE overexpression in boosting NOTCH pathway activity in T-cell leukemia home...
