Evaluation of diclofenac utilization patterns before and after digital risk minimization intervention in outpatient settings in Montenegro
ConclusionsAlthough a significant decrease in diclofenac prescription in both cohorts occurred, diclofenac is still prescribed, even in diagnoses contraindicated for its use. Further improvements of existing tools and the creation of new ones are necessary for minimization of cardiovascular and other risks related to diclofenac. (Source: Drugs and Therapy Perspectives)

Adsorption of anti-inflammatory and analgesic drugs traces in water on clay minerals
Chemosphere. 2024 Feb 20:141469. doi: 10.1016/j.chemosphere.2024.141469. Online ahead of print.ABSTRACTThe aim of this study was to assess the adsorption of four non-steroidal anti-inflammatory drugs (NSAIDs), namely Paracetamol (PRC), Diclofenac (DIC), Ibuprofen (IBU), and Ketoprofen (KET), using both batch and continuous experiments with clay. Various analytical techniques, including XRD, FTIR, SEM coupled to EDX, and Zeta potential, were employed to characterize both raw and calcined clay. XRD and FTIR analyses confirmed the kaolinite nature of the clay. SEM data revealed a lamellar structure formed in the clay after ca...

Comparing different versions of the Yoon-Nelson model in describing organic micropollutant adsorption within fixed bed adsorbers
This study aims to augment its modeling efficacy through two proposed refinements found in the literature: a logarithmic transformation and the incorporation of steric hindrance effects. We systematically evaluated the original Yoon-Nelson model alongside the modified versions, using breakthrough data associated with micropollutant adsorption on solid materials. Three distinct cases were scrutinized: (1) caffeine adsorption on activated carbon; (2) tetracycline adsorption on hierarchical porous carbon; and (3) diclofenac adsorption on organoclay. While all three models demonstrated comparable performance with highly symmet...

Structural modification based on the diclofenac scaffold: Achieving reduced colitis side effects through COX-2/NLRP3 selective inhibition
Eur J Med Chem. 2024 Feb 17;268:116257. doi: 10.1016/j.ejmech.2024.116257. Online ahead of print.ABSTRACTCOX-2/NLPR3-targeted therapy might be beneficial for the inflammation diseases. To discover novel anti-inflammatory compounds with favorable safety profiles, three new series of non-carboxylic diclofenac analogues bearing various ring systems, such as oxadiazoles 4a-4w, triazoles 6a-6m, and cyclic imides 7a and 7b, were synthesized. The synthesized analogues were evaluated for their inhibitory activity against COX-2 enzyme. Among them, compound 6k exhibited potent selective COX-2 inhibition (IC50 = 1.53 μM; selectivity...

Diclofenac-loaded PLGA nanoparticles downregulate LasI/R quorum sensing genes in pathogenic P. aeruginosa isolates
In this study, diclofenac sodium-loaded PLGA nanoparticles (DS-PLGA NPs) were produced by solvent evaporation technique and characterized using SEM, DLS, and zeta potential analyses. The antibacterial and antivirulence potential of DS-PLGA NPs against P. aeruginosa strains were examined using broth microdilution, crystal violet staining, hemolysis, and twitching quantification assays. Furthermore, the expression of the quorum sensing (QS) genes, lasI and lasR in P. aeruginosa strains after treatment with 1/2 MIC of DS-PLGA NPs was assessed using real-time PCR. SEM imaging of the synthesized NPs exhibited that the NPs have ...

Field Cancerization Therapies for the Management of Actinic Keratosis: An Updated Review
AbstractField cancerization theory highlights that the skin surrounding actinic keratoses (AK) is also at increased risk for possible malignant transformation; thus, field-directed treatments may both reduce the risk of AK recurrence and potentially reduce the risk of development of cutaneous squamous cell carcinoma (cSCC). Photodynamic therapy (PDT) with either aminolevulinic acid (ALA) or methylaminolevulinate (MAL), as well as topical treatments such as 5-fluorouracil (5-FU), diclofenac gel, piroxicam, imiquimod, and ingenol mebutate, have all shown higher efficacy than vehicle treatments. PDT is widely recognized for i...

Hydroponic materials improve organic micropollutant removal in vertical flow constructed wetlands treating wastewater
This study revealed notable removal of persistent and mobile micropollutants in cocopeat CWH, namely carbamazepine (80-86%) and diclofenac (97-100%). These results demonstrate the potential beneficial use of hydroponic materials as substratum in more advanced constructed wetlands designed to remove micropollutants.PMID:38346507 | DOI:10.1016/j.chemosphere.2024.141388 (Source: Chemosphere)

Evaluating an integrated nano zero-valent iron column system for emerging contaminants removal from different wastewater matrices - Identification of transformation products
In this study, the performance of a column system consisting of zero-valent iron nanoparticles (nZVI) incorporated into a cationic resin and synthesized from green tea extract with the addition of persulfate for the elimination of selected pharmaceuticals and endocrine disruptors from wastewater is evaluated. Ibuprofen, naproxen, diclofenac and ketoprofen were the target pharmaceuticals from non-steroidal anti-inflammatory drugs group, while bisphenol A was the target endocrine disruptor. In this context, different real wastewater effluent matrices were investigated: anaerobic membrane bioreactor (AnMBR), upflow anaerobic ...

Contaminants of emerging concern in water and sediment of the Venice Lagoon, Italy
This study investigates for the first time the contamination of water and sediment of the Venice Lagoon by twenty Contaminants of Emerging Concern (CECs): three hormones, six pharmaceutical compounds (diclofenac and five antibiotics, three of which are macrolides), nine pesticides (methiocarb, oxadiazon, metaflumizone, triallate, and five neonicotinoids), one antioxidant (BHT), and one UV filter (EHMC). Water and sediment samples were collected in seven sites in four seasons, with the aim of investigating the occurrence, distribution, and possible emission sources of the selected CECs in the studied transitional environmen...
