Genomic Sequencing Surveillance to Identify Respiratory Syncytial Virus Mutations, Arizona, USA
Emerg Infect Dis. 2023 Sep 13;29(11). doi: 10.3201/eid2911.230836. Online ahead of print.ABSTRACTWe conducted surveillance of respiratory syncytial virus (RSV) genomic sequences for 100 RSV-A and 27 RSV-B specimens collected during November 2022-April 2023 in Arizona, USA. We identified mutations within prefusion F-protein antigenic sites in both subtypes. Continued genomic surveillance will be critical to ensure RSV vaccine effectiveness.PMID:37705075 | DOI:10.3201/eid2911.230836

Klebsiella pneumoniae: adaptive immune landscapes and vaccine horizons
Trends Immunol. 2023 Sep 11:S1471-4906(23)00159-X. doi: 10.1016/j.it.2023.08.005. Online ahead of print.ABSTRACTKlebsiella pneumoniae is among the most common antibiotic-resistant pathogens causing nosocomial infections. Additionally, it is a leading cause of neonatal sepsis and childhood mortality across the globe. Despite its clinical importance, we are only beginning to understand how the mammalian adaptive immune system responds to this pathogen. Further, many studies investigating potential K. pneumoniae vaccine candidates or alternative therapies have been launched in recent years. Here, we review the current state o...

Infections due to Salmonella sp. in children with chronic granulomatous disease: Our experience from North India
Clin Immunol. 2023 Sep 11:109769. doi: 10.1016/j.clim.2023.109769. Online ahead of print.ABSTRACTInfections with non-typhoidal Salmonella sp. have been documented in children with chronic granulomatous disease (CGD), but the prevalence of salmonella infection in children with CGD in underdeveloped countries is unknown. We assessed the clinical profiles of CGD patients diagnosed at our tertiary care centre in north India and had Salmonella sp.infections. We found three patients with Salmonella sp. bloodstream infections (2-proven, 1-probable) among the 99 CGD patients. After receiving cotrimoxazole prophylaxis following a C...

Pharma - manufacturing: the unappreciated and overlooked indispensable skill
Curr Opin Immunol. 2023 Sep 11;84:102385. doi: 10.1016/j.coi.2023.102385. Online ahead of print.ABSTRACTThe process of vaccine production, manufacturing, is time-intensive, complex, expensive, and highly technical, requiring close coordination and collaboration among multiple companies with different inputs, from active pharmaceutical ingredients to glass, and specializations, and with the supply chains spread across many countries. Covid-19 pandemic highlighted that neglecting and ignoring the need for a global effort in vaccine manufacturing and delivery can have alarming, and devastating, repercussions, especially when ...

Obtaining a high titer of polyclonal antibodies from rats to the SARS-CoV-2 nucleocapsid protein and its N- and C-terminal domains for diagnostic test development
J Immunol Methods. 2023 Sep 11:113558. doi: 10.1016/j.jim.2023.113558. Online ahead of print.ABSTRACTSevere Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) is an enveloped, plus-stranded RNA virus responsible for the Coronavirus Disease 2019 (COVID-19). Patients infected with COVID-19 may be asymptomatic or have symptoms ranging from mild manifestations to severe cases of the disease that could lead to death. The SARS-CoV-2 genome encodes 4 structural proteins, including the Spike protein (S), the Nucleocapsid protein (N), membrane protein (M) and, the envelope (E). The N protein forms a major component of the ribonu...

Suramin inhibits SARS-CoV-2 nucleocapsid phosphoprotein genome packaging function
Virus Res. 2023 Sep 11:199221. doi: 10.1016/j.virusres.2023.199221. Online ahead of print.ABSTRACTThe coronavirus disease 2019 (COVID-19) pandemic is fading, however its etiologic agent severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) continues posing - despite the availability of licensed vaccines - a global health threat, due to the potential emergence of vaccine-resistant SARS-CoV-2 variants. This makes the development of new drugs against COVID-19 a persistent urgency and sets as research priority the validation of novel therapeutic targets within the SARS-CoV-2 proteome. Among these, a promising one is the...

Macrophage memory: Types, mechanisms, and its role in health and disease
Immunology. 2023 Sep 13. doi: 10.1111/imm.13697. Online ahead of print.ABSTRACTOn the basis of the mechanisms of action and characteristics of immune effects, immunity is commonly categorized into innate and adaptive immunity. Adaptive immunity is associated with the response to non-self-entities and is characterized by high specificity and memory properties. In contrast, innate immunity has traditionally been considered devoid of memory characteristics. However, an increasing number of studies have sought to challenge this conventional immunological dogma and shown that innate immune cells exhibit a more robust and rapid ...

COVID-19 vaccine booster doses provide increased protection against COVID-19 hospitalization compared with previously vaccinated individuals: Interim findings from the REFORCO-Brazil real-world effectiveness study during Delta and Omicron
CONCLUSIONS: Our results support the use of AZD1222 and other adenoviral/mRNA vaccine boosters to maintain protection against COVID-19 hospitalization from Omicron subvariants, including in elderly and immunocompromised individuals at increased risk of accelerated waning or severe outcomes.PMID:37704499 | DOI:10.1016/j.vaccine.2023.08.085

Prophylactic HPV vaccines in patients with HPV-associated diseases and cancer
Vaccine. 2023 Sep 11:S0264-410X(23)00990-8. doi: 10.1016/j.vaccine.2023.08.047. Online ahead of print.ABSTRACTIndividuals with human papillomavirus (HPV)-related disease remain at risk for subsequent HPV infection and related disease after treatment of specific lesions. Prophylactic HPV vaccines have shown benefits in preventing subsequent HPV-related disease when administered before or soon after treatment. Based on our understanding of the HPV life cycle and vaccine mechanism of action, prophylactic HPV vaccination is not expected to clear active persistent HPV infection or unresected HPV-associated dysplastic tissue rem...

Integrative transcriptomic and metabolomic analysis in mice reveals the mechanism by which ginseng stem-leaf saponins enhance mucosal immunity induced by a porcine epidemic diarrhea virus vaccination
Vaccine. 2023 Sep 11:S0264-410X(23)01085-X. doi: 10.1016/j.vaccine.2023.09.017. Online ahead of print.ABSTRACTPorcine epidemic diarrhea virus (PEDV) is a main cause of severe enteric disease in piglets, leading to millions of dollars lost annually in the global pig industry. Parenteral vaccination is limited in generating sufficient mucosal immunity, which is crucial for early defense against PEDV. Here, we orally administered ginseng stem-leaf saponins (GSLS) to mice before parenteral vaccination and found that GSLS significantly enhanced the phagocytosis of dendritic cells, promoted the activities of CD4+ T cells and inc...
