Assessing potential drug-drug interactions between clofazimine and other frequently used agents to treat drug-resistant tuberculosis
Antimicrob Agents Chemother. 2024 Apr 10:e0158323. doi: 10.1128/aac.01583-23. Online ahead of print.ABSTRACTClofazimine is included in drug regimens to treat rifampicin/drug-resistant tuberculosis (DR-TB), but there is little information about its interaction with other drugs in DR-TB regimens. We evaluated the pharmacokinetic interaction between clofazimine and isoniazid, linezolid, levofloxacin, and cycloserine, dosed as terizidone. Newly diagnosed adults with DR-TB at Klerksdorp/Tshepong Hospital, South Africa, were started on the then-standard treatment with clofazimine temporarily excluded for the initial 2 weeks. Pha...

Efflux pump effects on levofloxacin resistance in < em > Mycobacterium abscessus < /em >
Antimicrob Agents Chemother. 2024 Apr 4:e0134823. doi: 10.1128/aac.01348-23. Online ahead of print.ABSTRACTMycobacterium abscessus (M. abscessus) inherently displays resistance to most antibiotics, with the underlying drug resistance mechanisms remaining largely unexplored. Efflux pump is believed to play an important role in mediating drug resistance. The current study examined the potential of efflux pump inhibitors to reverse levofloxacin (LFX) resistance in M. abscessus. The reference strain of M. abscessus (ATCC19977) and 60 clinical isolates, including 41 M. abscessus subsp. abscessus and 19 M. abscessus subsp. massi...

Influence of microplastics and environmentally persistent free radicals on the ability of biochar components to promote degradation of antibiotics by activated peroxymonosulfate
Environ Pollut. 2024 Apr 2:123827. doi: 10.1016/j.envpol.2024.123827. Online ahead of print.ABSTRACTMicroplastics (MPs) in sludge can affect the ability of biochar-activated peroxymonosulfate (PMS) to degrade antibiotics. In this work, biochar was prepared by mixing sludge and polystyrene (PS) through hydrothermal carbonization (HTC) and high-temperature pyrolysis processes. The resulting biochar was used to activate PMS to degrade ofloxacin (OFX), levofloxacin (LEV), and pefloxacin (PFX). The addition of PS significantly enhanced the ability of biochar/PMS to degrade antibiotics and the levels of environmentally persisten...

A novel molecularly imprinted electrochemical sensor from poly (3, 4-ethylenedioxythiophene)/chitosan for selective and sensitive detection of levofloxacin
Int J Biol Macromol. 2024 Apr 1:131321. doi: 10.1016/j.ijbiomac.2024.131321. Online ahead of print.ABSTRACTThe improper usage of levofloxacin (LEV) endangers both environmental safety and human public health. Therefore, trace analysis and detection of LEV have extraordinary significance. In this paper, a novel molecularly imprinted polymer (MIP) electrochemical sensor was developed for the specific determination of LEV by electrochemical polymerization of o-phenylenediamine (o-PD) using poly(3,4-ethylenedioxythiophene)/chitosan (PEDOT/CS) with a porous structure and rich functional groups as a carrier and LEV as a template...

Research on the sustainable effect of ZnS and MoS 2 decorated biochar nanocomposites for removing quinolones from antibiotic-polluted aqueous solutions
Ying-Jie Gan, Yue-hui Wang, Ya-ru Dang, Wen-jie Hao, Zheng-rong Hu, Zhi-wei Zhang, Yu-min Luo, Yan-bo Wu, Ai-jun Song, Zhong-bao Zhao - Volume 77(4) Schematic of the synthesis of ZMMBC and its use as an absorbant for quinolone antibiotics.MoS2 and ZnS modified biochar (ZMMBC) was synthesised by hydrothermal method. The removal potential of pefloxacin (PF), levofloxacin (LF) and norfloxacin (NF) was studied. It only takes 90 min for ZMMBC to reach its maximum adsorption capacity. The adsorption behaviour mainly includes complexation, pore filling, π–π interactions...

Estimation of country-specific tuberculosis resistance antibiograms using pathogen genomics and machine learning
CONCLUSIONS: This is the first attempt at global Mtb antibiogram estimation. DR prevalence in Mtb can be reliably estimated using public WGS and phenotypic resistance prediction for key antibiotics, but public WGS data demonstrates oversampling of isolates with higher resistance levels than MDR. Nevertheless, our results raise concerns about the empiric use of short-course fluoroquinolone regimens for drug-susceptible TB in South Asia and indicate underutilisation of ethionamide in MDR treatment.PMID:38548342 | DOI:10.1136/bmjgh-2023-013532 (Source: Cancer Control)
