Assessment of No-Observed-Effect-Levels for DNA Adducts Formation by Genotoxic Carcinogens in Fetal Turkey Livers
Toxicology. 2023 Dec 21:153714. doi: 10.1016/j.tox.2023.153714. Online ahead of print.ABSTRACTFor genotoxic carcinogens, covalent binding to DNA is a critical initiating event in tumorigenesis. The present research investigated dose-effect relationships of three genotoxic carcinogens representing different structural classes, 2-acetylaminofluorene (2-AAF), benzo[a]pyrene (B[a]P) and quinoline (QUI), to assess the existence of no-observed-effect-levels (NOELs) for the formation of DNA adducts. Carcinogens were administered into the air sac of fertilized turkey eggs over wide dose ranges in three daily injections on days 22 ...

Assessment of the genotoxic and mutagenic effects induced by T-2 mycotoxin in HepG2 cells
Toxicology. 2023 Dec 19:153712. doi: 10.1016/j.tox.2023.153712. Online ahead of print.ABSTRACTThe T-2 toxin is a mycotoxin produced by molds belonging to Fusarium. Among the Fusarium mycotoxins, trichothecenes are frequently reported in food and feed, being the T-2 toxin (T-2) the mycotoxin which possesses the highest toxicity. According to EFSA, T-2 is found in various cereal grains used in food and feed products, mainly in oats, and it has a high environmental impact due to its mechanisms of toxicity. However, recent information on its genotoxic and mutagenic effects is lacking. This work aimed to evaluate the genotoxic ...

Multiomics was used to clarify the mechanism by which air pollutants affect chronic obstructive pulmonary disease: A human cohort study
Toxicology. 2023 Dec 18:153709. doi: 10.1016/j.tox.2023.153709. Online ahead of print.ABSTRACTExposure to air pollutants has been associated with various adverse health outcomes, including chronic obstructive pulmonary disease (COPD). However, the precise underlying mechanism by which air pollution impacts COPD through remains insufficiently understood. To elucidated the molecular mechanism by which air pollutant exposure contributes to alterations in the gut microbiome and metabolism in AECOPD patients, we employed metagenomics and untargeted metabolomics to analyse the gut microbial, faecal, and serum metabolites. The co...

A review of heavy metal risks around e-waste sites and comparable municipal dumpsites in major African cities: Recommendations and future perspectives
Toxicology. 2023 Dec 18:153711. doi: 10.1016/j.tox.2023.153711. Online ahead of print.ABSTRACTIn Africa, the effects of informal e-waste recycling on the environment are escalating. It is regularly transported from developed to developing nations, where it is disassembled informally in search of precious metals, thus increasing human exposure to harmful compounds. Africa has a serious problem with e-waste, as there are significant facilities in Ghana and Nigeria where imported e-waste is unsafely dismantled. however, because they are in high demand and less expensive than new ones, old electronic and electrical items are i...

DON induced DNA damage triggers absence of p53-mediated G2 arrest and apoptosis in IPEC-1 cells
In this study, we explored the detrimental impacts of DON on porcine intestinal epithelial cells (IPEC-1), serving as a representative model for porcine intestinal epithelial cells. After treating cells with DON for 24 h, DON can significantly inhibit the activity of cells, induce the production of reactive oxygen species (ROS), significantly reduce the content of glutathione and the activity of catalase, and increase the activity of superoxide dismutase and malondialdehyde, leading to an imbalance in intracellular redox status. In addition, DON can induce DNA double-strand breaks, and decrease mitochondrial membrane poten...

A Deep-Learning Approach for Identifying Prospective Chemical Hazards
Toxicology. 2023 Dec 15:153708. doi: 10.1016/j.tox.2023.153708. Online ahead of print.ABSTRACTWith the aim of helping to set safe exposure limits for the general population, various techniques have been implemented to conduct risk assessments for chemicals and other environmental stressors; however, none of these tools facilitate the identification of completely new chemicals that are likely hazardous and elicit an adverse biological effect. Here, we detail a novel in silico, deep-learning framework that is designed to systematically generate structures for new chemical compounds that are predicted to be chemical hazards. ...
