Three-dimensional structure of liver vessels and spatial distribution of hepatic immune cells
Journal of Innovative Optical Health Sciences, Ahead of Print. As the largest internal organ of the human body, the liver has an extremely complex vascular network and multiple types of immune cells. It plays an important role in blood circulation, material metabolism, and immune response. Optical imaging is an effective tool for studying fine vascular structure and immunocyte distribution of the liver. Here, we provide an overview of the structure and composition of liver vessels, the three-dimensional (3D) imaging of the liver, and the spatial distribution and immune function of various cell components of the liver. Espe...

Detection of cells by flow cytometry: Counting, imaging, and cell classification
Journal of Innovative Optical Health Sciences, Ahead of Print. The study of circulating cells in the blood stream is critical, as it covers many fields of biomedicine, including immunology, cell biology, oncology, and reproductive medicine. In-vivo flow cytometry (IVFC) is a new tool to monitor and count cells in real time for long durations in their native biological environment. This review describes two main categories of IVFC, i.e., labeled and label-free IVFC. It focuses on label-free IVFC and introduces its technological development and related biological applications. Because cell recognition is the basis of flow cy...

Transmissive multifocal laser speckle contrast imaging through thick tissue
In this study, we propose a transmissive multifocal LSCI for depth-resolved blood flow in thick tissue, further extending the transmissive LS CI for tissue thickness up to [math]. The limited-DOF imaging system is applied to the multifocal acquisition, and the depth of the vessel is estimated using a robust visibility parameter [math] in the coherent domain. The accuracy and linearity of depth estimation are tested by Monte Carlo simulati ons. Based on the proposed method, the model of contrast analysis resolving the depth information is established and verified in a phantom experiment. We demonstrated its effectiveness in...

Comparison of uniform resampling and nonuniform sampling direct-reconstruction methods in k-space for FD-OCT
Journal of Innovative Optical Health Sciences, Ahead of Print. The nonuniform distribution of interference spectrum in wavenumber [math]-space is a key issue to limit the imaging quality of Fourier-domain optical coherence tomography (FD-OCT). At present, the reconstruction quality at different depths among a variety of processing methods in [math]-space is still uncertain. Using simulated and experimental interference spectra at different depths, the effects of common six processing methods including uniform resampling (linear interpolation (LI), cubic spline interpolation (CSI), time-domain interpolation (TDI), and K-B w...

DecodeSTORM: A user-friendly ImageJ plug-in for quantitative data analysis in single-molecule localization microscopy
Journal of Innovative Optical Health Sciences, Ahead of Print. Quantitative data analysis in single-molecule localization microscopy (SMLM) is crucial for studying cellular functions at the biomolecular level. In the past decade, several quantitative methods were developed for analyzing SMLM data; however, imaging artifacts in SMLM experiments reduce the accuracy of these methods, and these methods were seldom designed as user-friendly tools. Researchers are now trying to overcome these difficulties by developing easy-to-use SMLM data analysis software for certain image analysis tasks. But, this kind of software did not pa...
