JAMA Neurology
Mission Statement: The mission of JAMA Neurology is to publish and disseminate scientific information primarily important for physicians caring for people with neurologic disorders and for those interested in the structure and function of the normal and diseased human nervous system. The specific aims are to (1) publish timely original research, including clinical trials that will directly improve clinical neurologic care and that will inform efforts to improve neurological health and promote health care equity; (2) report translational research that is pertinent to the understanding of neurologic disease; (3) address topi...

Comparative Efficacy of Disease-Modifying Treatments for Relapsing Multiple Sclerosis —Variations in Real-World Experience—Reply
In Reply We thank Dr Wolf and colleagues for highlighting the importance of clinical context when evaluating effectiveness of multiple sclerosis (MS) therapies. As demonstrated in the comparative effectiveness of natalizumab and fingolimod, where seemingly discordant results emerged from different registries, characteristics of the study population play a central role in study findings. It is, therefore, important to differentiate between studies of treatment efficacy and effectiveness. (Source: JAMA Neurology)

Comparative Efficacy of Disease-Modifying Treatments for Relapsing Multiple Sclerosis —Variations in Real-World Experience
With great interest we read the study by Roos et al comparing the efficacy of 2 anti-CD20 disease-modifying therapies (DMTs) commonly used for relapsing multiple sclerosis (MS). As no head-to-head randomized clinical trials (RCTs) have been completed, the comparative effectiveness of ocrelizumab and rituximab on relapse reduction was evaluated in patients from the MSBase and Danish MS registries. Over a mean follow-up of 1.4 years, the annualized relapse rate (ARR) for patients treated with rituximab was 0.20, whereas for ocrelizumab it was 0.09. Noninferiority was not demonstrated for treatment with rituximab when compare...
