Ionic detergent under pressure-vacuum as an innovative strategy to generate canine tracheal scaffold for organ engineering
Decellularized scaffolds applied in tissue engineering offer improvements, supplying the elevated necessity for organs and tissues for replacement. However, obtaining a functional trachea for autotransplantation or allotransplantation is tricky due to the organ anatomical and structural complexity. Most tracheal decellularization protocols are lengthy, expensive and could damage the tracheal extracellular matrix (ECM) architecture and functionality. Here we aim to evaluate the effectiveness of three different decellularization protocols combined with chemical and physical methods to obtain acellular canine tracheal scaffol...

Insulin-like growth factor-1 promotes human uterine leiomyoma cells proliferation via PI3K/AKT/mTOR pathway
The present research aims to evaluate the expression of insulin-like growth factor-1 (IGF-1) in uterine leiomyoma, and explore its relationship with the occurrence and development of uterine leiomyoma and potential signal pathways. QRT-PCR and enzyme linked immunosorbent assay (ELISA) were used for estimating the levels of IGF-1 in human uterine leiomyoma compared to myometrium. The expression of cell proliferation and PI3K/AKT/mTOR signaling pathway related proteins in uterine leiomyoma cells were evaluated by Western blot. Cell viability analysis was performed by CCK-8 assay. Lentivirus infection was used for IGF-1 overe...

Developmental characteristics of the glomerular and tubular portion of the nephron in the human foetal kidney cortex: Morphometrical and immunohistochemical analysis
This study aimed to morphometrically examine the development of glomeruli and tubules in the kidney cortex of human foetuses at different gestational ages (GAs). We also investigated the expression of the proliferation marker Ki-67 and apoptosis-related markers Bcl-2 and Bax during nephrogenesis using immunohistochemistry. Kidney samples from 38 human foetuses of both sexes with GA ranging from 13 to 40 weeks were analysed. The average area, perimeter, and Feret's diameter of the glomeruli gradually decreased up to a period of 29 –32 weeks of gestation and subsequently increased until a period of 37–40 weeks. There was...

Complex Material Properties of Gel-Amin: A Transparent and Ionically Conductive Hydrogel for Neural Tissue Engineering
The field of tissue engineering has benefited greatly from the broad development of natural and synthetic polymers. Extensive work in neural engineering has demonstrated the value of conductive materials to improve spontaneous neuron activity as well as lowering the necessary field parameters for exogenous electrical stimulation. Further, cell fate is directly coupled to the mechanical properties of the cell culture substrate. Increasing the conductivity of hydrogel materials often necessitates the addition of dopant materials, organic and inorganic that facilitate electron mobility. However, very little electron transfer ...

Development of a high-color flow cytometry panel for immunologic analysis of tissue injury and reconstruction in a rat model
The rat model is an important resource in biomedical research due to its similarities to the human immune system and its use for functional studies. However, because of the preponderance of mouse models in foundational and mechanistic immunologic studies, there is a relative lack of diverse, commercially available flow cytometry antibodies for immunological profiling in the rat model. Available antibodies are often conjugated to common fluorophores with similar peak emission wavelengths, making them hard to distinguish on conventional flow cytometers and restricting more comprehensive immune analysis. This can become a lim...

miR-654-5p suppresses migration and proliferation of vascular smooth muscle cells by targeting ADAMTS-7
Coronary artery disease (CAD) is the first leading cause of death worldwide. Therefore, novel therapeutic strategies need to be explored. Numerous publications reported that microRNA-654-5p (miR-654-5p) had anti-cancer activities in various cancers, and it was proved to modulate cell migration, invasion and proliferation, which played critical roles in CAD. However, its role in CAD is unknown. Thus, we aimed to evaluate the role of miR-654-5p in vascular smooth muscle cells (VSMCs) involved in CAD. A total of 25 CAD patients and 19 healthy individuals were enrolled to evaluate their circulating miR-654-5p levels. MiR-654-5...

Pentraxin-3 promotes LPS-induced pyroptosis in human periodontal ligament stem cells
This study indicated that PTX3 promotes LPS-induced pyroptosis and inflammation in hPDLSCs through activation of the caspase-1-dependent NLRP3 inflammasome. (Source: Cells Tissues Organs)

Effects of Pulsatility on Arterial Endothelial and Smooth Muscle cells
Continuous Flow Ventricular Assist Device (CFVAD) support in advanced heart failure patients causes diminished pulsatility, which has been associated with adverse events including gastrointestinal bleeding, end organ failure and arteriovenous malformation. Recently, pulsatility augmentation by pump speed modulation has been proposed as a means to minimize adverse events. Pulsatility primarily affects endothelial and smooth muscle cells in the vasculature. To study the effects of pulsatility and pulse modulation using CFVADs, we have developed a microfluidic co-culture model with human aortic endothelial (ECs) and smooth mu...

Static magnetic field promotes proliferation, migration, differentiation, and AKT activation of periodontal ligament stem cells
In this study, we revealed that a 320 mT SMF upregulates the protein expression levels of cytokines such as MCM7 and PCNA in proliferating PDLSCs. Cell counting kit-8 (CCK-8) results revealed that the SMF group had higher optical density (OD) values than the control group. The ratio of cells in the S phase to those in the G2/M phase was significantly increased after exposure to a 320 mT SMF. In scratch assays, the SMF-treated PDLSCs exhibited a higher migration rate than the sham-exposed group after 24 h of culture, indicating that the SMF promoted the migratory ability of PDLSCs. The activity level of the early differenti...

Successful in vivo Transplantation of Cultured and Enriched Testicular Germ Cells of Pre-Pubertal Bucks to Busulfan-Treated Homologous Recipients
The objective of the present study was to establish a workable approach for the production of germ cell (GC)-depleted recipient goat model using intra-testicular busulfan treatment and transplantation of cultured and enriched caprine-male GC (cmGCs) into the homologous recipients under ultrasonography (USG) guidance. The evaluation of post-transplantation colonization of donor cmGCs and restoration of the normal architecture of seminiferous tubules (ST) was performed. For this, the cmGCs of pre-pubertal male goats were isolated and enriched by differential platting for culture until the third passage. Thereafter, cells wer...
